Abstract ObjectivesThis study explored differing levels of macronutrients in breast milk in relation to maternal anemia and hemoglobin. MethodsArchived milk specimens and data from a cross‐sectional sample of 208 breastfeeding mothers in northern Kenya, originally collected in 2006, were analyzed; data included milk fat, maternal hemoglobin concentration, and anemia status (anemia defined as hemoglobin <12 g/dL). Total protein and lactose were measured and energy was calculated. To explore the association between milk outcomes (fat, protein, lactose, and energy) and anemia, regression models were constructed with and without adjustment for maternal age, parity, and time (days) postpartum. The same models were constructed using hemoglobin as a continuous predictor in lieu of dichotomous anemia to explore the role of hemoglobin levels and anemia severity in predicting milk outcomes. ResultsThe group comparison indicated significantly higher milk protein and lower milk fat for anemic mothers relative to nonanemic counterparts. After adjustment for maternal age, parity, and time postpartum, maternal anemia was associated with significantly higher milk protein (P = 0.001) and significantly lower milk fat (P = 0.025). Hemoglobin had a significant inverse relationship with milk protein (P = 0.017) and a marginally significant positive relationship with milk fat (P = 0.060) after adjusting for the maternal variables. Neither anemia nor hemoglobin was significant in predicting lactose or milk energy. ConclusionsMaternal anemia and hemoglobin concentration may be associated with complex changes in milk macronutrients. Future research should clarify the impact of maternal anemia on a range of breast milk components while accounting for other maternal characteristics.

The evolutionary ecology of breastmilk folate among Ariaal agro-pastoralists in Kenya
Objective: Folate in breastmilk has important implications for offspring health and survival given the essential role of this vitamin in DNA synthesis, epigenetic functions, and amino acid metabolism. Yet, little is understood about the variation of folate in breastmilk and transfer across the postpartum year and beyond. Published studies tend to be limited to milk during days/weeks postpartum, and none applied an evolutionary perspective of parental investment. Methods: A secondary analysis of the data and specimens from 200 breastfeeding mothers within 1.5 years postpartum in food-insecure northern Kenya was conducted. ELISA determined folate-binding protein (FOLR1) in cryogenically archived breastmilk and maternal blood specimens, originally collected in 2006. Maternal folate was defined as blood serum FOLR1 multiplied by –1 because elevated FOLR1 is associated with folate deficiency. The concentration of milk FOLR1 was evaluated in relation to maternal folate and 1) infant sex (Trivers-Willard hypothesis), 2) time postpartum and parity (maternal residual reproductive value) using regression models adjusted for covariates. Results indicated: 1) no Trivers-Willard effect; 2) support for time postpartum but not for parity. Maternal folate and time postpartum inversely predicted milk FOLR1. There was an interaction between these variables (p<0.05). Maternal folate improved over time at a varying rate while milk FOLR1 decreased at a relatively steady rate. This inverse relationship became stronger as time advanced. Conclusion: The priority shift from the investment in current offspring toward maternal soma and potential future offspring in this study provides empirical support for the evolutionary hypothesis of parental investment and parent-offspring conflict. Orangutans have a 7.6 year average inter-birth interval, the longest of any mammal. Suckling occurs throughout that interval, until the birth of the next offspring, but it is unclear how important milk consumption is during that period, as we cannot assess the actual intake amount. Measurement of stable carbon isotope ratios (d13C), stable nitrogen isotope ratios (d15N) and nitrogen content of feces (%N) provide evidence of the transition between breast milk to solid food. Here we present pilot data on these isotopic ratios from matched fecal samples of mothers and offspring (n=43), collected from wild orangutans in Gunung Palung National Park, Borneo, Indonesia. We found that the youngest infant (2.3 yrs) had the highest d15N values overall, indicative of a higher percentage of animal products (milk) in the diet. Older juveniles (5.8 yrs) did not consistently show higher d15N than their mothers. This may indicate variation in suckling frequency or the amount of breast milk consumed per suckling session. Adolescents (10-13 yrs) showed significantly (GLMM, p< 0.007) lower d15N than samples taken from the same day on their mothers. This is surprising given that they were eating similar diets. We thus compare the isotopic signature of the plants consumed to examine the sources of this variation. We conclude that analysis of fecal samples collected from wild orangutans can be used to assess the relative importance of breast milk in the diet, but caution that isotopic excretion may also be effected by differences in the isotopic content of the diet. null (Ed.)Bornean orangutans' extended life history may be an adaptation to their forest habitat, characterized by dramatically fluctuating fruit availability. We hypothesize that juveniles rest and are carried more when fruit availability is low, spending less time in developmentally important behavior including play and independent travel to conserve energy. We test this using generalized linear mixed models and data derived from 976 follows of infant and juvenile orangutans collected during periods of varying fruit availability. Age (p<0.001), but not fruit availability (p>0.05) significantly affected offsprings’ time being carried (β=-8,1) and resting (β=-2.6, p<0.001). Younger individuals spent more time clinging and resting regardless of fruit availability. Fruit availability and offspring age interact to affect the proportion of time juveniles play (β=1.5, p<0.001) and travel (β=-0.8, p<0.001). Fruit availability impacts younger juveniles’ play behavior more dramatically than older juveniles: younger juveniles play more when fruit availability is high than when it is low, while older juveniles exhibit little variation in time spent playing depending on fruit availability. Juveniles aged 6-8 travel more when fruit availability is medium and high, juveniles aged 2-5 travel slightly less when fruit availability is high, and juveniles under 2 rarely travel independently. We also examined relationships between fruit availability, offspring age, and maternal travel distance using 2065 mother-offspring follows, demonstrating that females travel shorter distances with offspring under four, and when fruit availability is low. Thus, orangutans shift activity in response to fruit availability and throughout development, buffering young orangutans against energy depletion but suppressing developmentally important activities. Synopsis The ability to provision offspring with milk is a significant adaptive feature of mammals that allows for considerable maternal regulation of offspring beyond gestation, as milk provides complete nutrition for developing neonates. For mothers, lactation is a period of marked increases in energetic and nutritive demands to support milk synthesis; because of this considerable increase in demand imposed on multiple physiological systems, lactation is particularly susceptible to the effects of chronic stress. Here, we present work that explores the impact of chronic stress during lactation on maternal lactation performance (i.e., milk quality and quantity) and the expression of key milk synthesis genes in mammary tissue using a Sprague–Dawley rat model. We induced chronic stress using a well-established, ethologically relevant novel male intruder paradigm for 10 consecutive days during the postpartum period. We hypothesized that the increased energetic burden of mounting a chronic stress response during lactation would decrease lactation performance. Specifically, we predicted that chronic exposure to this social stressor would decrease either milk quality (i.e., composition of proximate components and energy density) or quantity. We also predicted that changes in proximate composition (i.e., lipid, lactose, and protein concentrations) would be associated with changes in gene expression levels of milk synthesis genes. Our results supported our hypothesis that chronic stress impairs lactation performance. Relative to the controls, chronically stressed rats had lower milk yields. We also found that milk quality was decreased; milk from chronically stressed mothers had lower lipid concentration and lower energy density, though protein and lactose concentrations were not different between treatment groups. Although there was a change in proximate composition, chronic stress did not impact mammary gland expression of key milk synthesis genes. Together, this work demonstrates that exposure to a chronic stressor impacts lactation performance, which in turn has the potential to impact offspring development via maternal effects.more » « less

The Trivers–Willard hypothesis states that mothers should adjust their offspring sex ratio according to their own condition and the environment they face during breeding. Past tests of this hypothesis have focused on how natural variation in weather, food availability, or predation pressure shapes sex allocation trade-offs. However, anthropogenic activities, such as urbanization, can alter all of the above characteristics presenting animals with novel challenges in optimizing their brood sex ratio. Previous research has examined how urban living influences individual body condition in several bird taxa, but few have explored subsequent impacts on secondary offspring sex ratio. One likely mediator of the link between environmental conditions, parental condition, and sex ratios is corticosterone (CORT), the primary glucocorticoid in birds. Research on CORT’s influence on sex ratios has focused solely on maternal CORT. However, for species with biparental care, paternal CORT or the similarity of maternal and paternal phenotypes may also help ensure that offspring demand matches parental care quality. To test these hypotheses, we explore offspring secondary sex ratios in European starlings (Sturnus vulgaris). We did not find an effect of site or parental body condition on the production of the more costly sex (males). Instead, we found preliminary evidence suggesting that the similarity of maternal and paternal CORT levels within a breeding pair may increase the likelihood of successfully fledging sons. Maternal and paternal CORT were not significant predictors of secondary sex ratio, suggesting that parental similarity, rather than parental CORT alone, could play a role in shaping secondary offspring sex ratios, but additional work is needed to support this pattern. Starlings are considered an urban-adapted species, making them a compelling model for future studies of the relationship between urbanization, parental body condition, and sex ratios.more » « less
