Title: One‐pot fabrication of robust interpenetrating hydrogels via orthogonal click reactions
ABSTRACT

Interpenetrating polymer network (IPN) hydrogels have been fabricated through a facile one‐pot approach from tetra/bifunctional telechelic macromonomers with epoxy, amine, azide, and alkyne groups by orthogonal double click reactions: epoxy‐amine reaction and copper‐catalyzed azide‐alkyne cycloaddition. Both the crosslinked networks are simultaneously constructed in water from the biocompatible poly (ethylene glycol)‐based macromonomers. The crosslinking density of each network was finely tuned by the macromonomer structure, permitting control of network molecular weights between crosslinks of the final gels. Compared to corresponding single network gels, the IPN gels containing both tightly and loosely crosslinked networks exhibited superior mechanical properties with shear moduli above 15 kPa and fracture stresses over 40 MPa. The synthetic versatility of this one‐pot approach will further establish design principles for the next generation of robust hydrogel materials. © 2016 Wiley Periodicals, Inc. J. Polym.   1. ABSTRACT

    Crosslinked polyhydroxyurethane (PHU) networks synthesized from difunctional six‐membered cyclic carbonates and triamines are reprocessable at elevated temperatures through transcarbamoylation reactions. Here we study the structural effects on reprocessability and stress relaxation in crosslinked PHUs. Crosslinked PHUs derived frombis(five‐membered cyclic carbonates) are shown to decompose at temperatures needed for reprocessing, likely via initial reversion of the PHU linkage and subsequent side reactions of the liberated amine and cyclic carbonate. Therefore, several six‐membered cyclic carbonate‐based PHUs with varying polymer backbones and crosslink densities were synthesized. These networks show large differences in the Arrhenius activation energy of stress relaxation (from 99 to 136 kJ/mol) that depend on the network structure, suggesting that transcarbamoylation reactions may be highly affected by both chemical and mechanical effects. Furthermore, all crosslinked PHUs derived from six‐membered cyclic carbonates show mechanical properties typical of thermoset polymers, but recovered as much as 80% of their as‐synthesized tensile properties after elevated temperature compression molding. These studies provide significant insight into factors affecting the reprocessability of PHUs and inform design criteria for the future synthesis of sustainable and repairable crosslinked PHUs. © 2017 Wiley Periodicals, Inc.   2. ABSTRACT

    Chemical crosslinkers are commonly used to stabilize both natural and synthetic macromolecules, while providing opportunities to install functionality and modulate polymer architecture. Here, we introduce the aromatic cyclopropenium cation as a tri‐functional crosslinker of secondary amine‐containing polymers. The one‐step crosslinking reaction is rapid and requires no subsequent purification. When dispersed in aqueous media, the crosslinked polymers form spherical nanoparticles with highly positive charge that is maintained even in alkaline conditions. This synthetic strategy will enable the incorporation of cyclopropenium into a wide variety of macromolecules. © 2018 Wiley Periodicals, Inc. J. Polym. Sci., Part A: Polym.   3. ABSTRACT

    1H NMR and SEC analyses are used to investigate the overall efficiency of Copper Catalyzed Azide Alkyne Cycloaddition (CuAAC) “click” coupling reactions between alkyne‐ and azide‐terminated polymers using polystyrene as a model. Quantitative convolution modeling of the entire molecular weight distribution is applied to characterize the outcomes of the functional polymer synthesis reactions (i.e., by atom transfer radical polymerization), as well as the CuAAC coupling reaction. Incomplete functionality of the azide‐terminated polystyrene (∼92%) proves to be the largest factor compromising the efficacy of the CuAAC coupling reaction and is attributed primarily to the loss of terminal bromide functionality during its synthesis. The efficiency of the SN2 reaction converting bromide to azide was found to be about 99%. After taking into account the influence of non‐functional polymer, we find that, under the reaction conditions used, the efficiency of the CuAAC coupling reaction determined from both techniques is about 94%. These inefficiencies compromise the fidelity and potential utility of CuAAC coupling reactions for the synthesis of hierarchically structured polymers. While CuAAC efficiency is expected to depend on the specific reaction conditions used, the framework described for determining reaction efficiency does provide a means for ultimately optimizing the reaction conditions for CuAAC coupling reactions. © 2017 Wiley Periodicals, Inc.   4. Abstract

    In this study, an in situ forming corneal stromal substitute based on collagen type I crosslinked by bio‐orthogonal strain‐promoted azide–alkyne cycloaddition (SPAAC) is presented. The crosslinked collagen gel has greater transparency compared to non‐crosslinked collagen gels. The mechanical properties of the gels are controlled by changing functional group ratios and conjugated collagen concentrations. Higher concentrations of conjugated collagen yield enhances mechanical properties, where the storage modulus increases from 42.39 ± 8.95 to 112.03 ± 3.94 Pa after SPAAC crosslinking. Encapsulated corneal keratocytes grow within the SPAAC‐crosslinked gels and corneal keratinocytes are supported on top of the gel surfaces. SPAAC‐crosslinked gels support more favorable and stable keratinocyte morphology on their surface compared to non‐crosslinked gels likely as a result of more optimal substrate stiffness, gel integrity, and resistance to degradation. SPAAC‐crosslinked collagen gels with and without encapsulated keratocytes applied to rabbit corneas in an organ culture model after keratectomy exhibit surface epithelialization with multilayered morphology. The novel in situ forming gel is a promising candidate for lamellar and defect reconstruction of corneal stromal tissue.

  5. Abstract

    The preparation of functionalized graft copolymers utilizing a combination of ATRP, ROMP, thio‐bromo, and Huisgen‐type click chemistries is described. The construction of these polymeric architectures hinges on the use of a norbornene‐based α‐bromo ester that can act as an ATRP initiator and ROMP monomer (a so‐calledinimer), as well as a thio‐bromo click partner. This allows for the use of a “grafting‐through” ROMP approach of ATRP‐based macromonomers that can undergo a post‐polymerization thio‐bromo click modification. Additionally, this material can undergo further modifications using archetypal copper‐catalyzed azide/alkyne click reaction as well as metal‐free strain‐promoted azide/alkyne click reactions.
