Abstract 5 by reversible imine condensation of cup‐shaped aldehyde2 (i.e., basket) with trivalent aromatic amine4 . This nanosized [4+4] cage (V =5 nm3,M w=6150 Da) has shape of a truncated tetrahedron with four baskets at its vertices and four aromatic amines forming the faces. Importantly,tris ‐aldehyde basket2 and aliphatictris ‐amine7 undergo condensation to give small [1+1] cage6 . The imine metathesis of6 and aromatictris ‐amine4 into CBC5 was optimized to bias the equilibrium favouring6 . Addition of tribromoacetic acid (TBA) as a chemical fuel perturbs this equilibrium to result in the transient formation of CBC5 , with subsequent consumption of TBA via decarboxylation driving the system back to the starting state. -
Abstract We describe the preparation, dynamic, assembly characteristics of vase‐shaped basket
1 3−along with its ability to form an inclusion complex with anticancer drug mitoxantrone in abiotic and biotic systems. This novel cavitand has a deep nonpolar pocket consisting of three naphthalimide sides fused to a bicyclic platform at the bottom while carrying polar glycines at the top. The results of1H Nuclear Magnetic Resonance (NMR),1H NMR Chemical Exchange Saturation Transfer (CEST), Calorimetry, Hybrid Replica Exchange Molecular Dynamics (REMD), and Microcrystal Electron Diffraction (MicroED) measurements are in line with1 forming dimer [1 2]6−, to be in equilibrium with monomers1 (R )3−(relaxed) and1 (S )3−(squeezed). Through simultaneous line‐shape analysis of1H NMR data, kinetic and thermodynamic parameters characterizing these equilibria were quantified. Basket1 (R )3−includes anticancer drug mitoxantrone (MTO 2+) in its pocket to give stable binary complex [MTO ⊂1 ]−(K d=2.1 μM) that can be precipitated in vitro with UV light or pH as stimuli. Both in vitro and in vivo studies showed that the basket is nontoxic, while at a higher proportion with respect to MTO it reduced its cytotoxicity in vitro. With well‐characterized internal dynamics and dimerization, the ability to include mitoxantrone, and biocompatibility, the stage is set to develop sequestering agents from deep‐cavity baskets. -
Abstract Simple synthetic routes to regioselectively deuterated tris[2‐(dimethylamino)ethyl]amine (Me6TREN) variants are described. Imine formation with formaldehyde‐
d2 from tris(2‐aminoethyl)amine (TREN) and subsequent reductions with NaBD4afforded N[CH2CH2N(CD3)2]3ord 18‐Me6TREN in 79 % yield. A trisubstitution protocol from 2‐bromo‐N,N ‐dimethylacetamide and ammonium carbonate and subsequent reduction of the N(CH2CONMe2)3intermediate by lithium aluminum deuteride has afforded N[CH2CD2N(CH3)2]3or (d 6‐arm)‐Me6TREN in three steps and 52 % overall yield. A similar protocol from 2‐bromo‐N,N ‐dimethyl‐d 2‐ acetamide, obtained in two steps fromd 4‐acetic acid, with reduction of the N(CD2CONMe2)3intermediate by lithium aluminum hydride has afforded N[CD2CH2N(CH3)2]3or (d 6‐cap)‐Me6TREN in four steps and 13 % overall yield from CD3COOD. -

Abstract New stimulus‐responsive scaffolds are of interest as constituents of hierarchical supramolecular ensembles. 1,3,5–2,4,6‐Functionalized, facially segregated benzene moieties have a time‐honored role as building blocks for host molecules. However, their user as switchable motifs in the construction of multi‐component supramolecular structures remains poorly explored. Here, we report a molecular cage
1 , which consists of a bent anthracene dimer3 paired with 1,3,5‐tris(aminomethyl)‐2,4,6‐triethylbenzene2 . As the result of the pH‐inducedababab ↔bababa isomerization of the constituent‐functionalized benzene units derived from2 , this cage can reversibly convert between an open state and a closed form, both in solution and in the solid state. Cage1 was used to create stimuli‐responsive hierarchical superstructures, namely Russian doll‐like complexes with [K⊂18‐crown‐6⊂1 ]+and [K⊂cryptand‐222⊂1 ]+. The reversible assembly and disassembly of these superstructures could be induced by switching cage1 from its open to closed form. The present study thus provides an unusual example where pH‐triggered conformation motion within a cage‐like scaffold is used to control the formation and disassociation of hierarchical ensembles. -
Abstract Herein we combine the subcomponent self‐assembly and integrative self‐sorting techniques with the well‐established platinum(II)‐pyridine coordination‐driven self‐assembly to report the quantitative synthesis and spectroscopic characterization of a heterometallic CuI‐PtIIquadrilateral
QL that is formed from a total of twelve molecular components from four unique species, including 5‐(pyridin‐4‐ylethynyl)picolinaldehyde (1 ),p ‐toluidine (2 ), [Cu(CH3CN4](PF6) (3 ), andcis ‐Pt(PEt3)2(OTf)2(4 ), in a 4:4:2:2 ratio. Despite the many different entities potentially forming from these four precursor units, the clean formation ofQL is mainly guided by the different coordination preferences of the metal ions3 and4 , and the design criteria inherent in compounds1 and2 .
