Honey bees are social insects that live in large groups called colonies, within structures known as hives. The young adult bees stay within the hive to build nests and care for the young, while the older bees leave the hive to forage for food. Honey bees store food and other valuable resources in their hives, so they are often targeted by predators, parasites and ‘robber’ bees from other colonies. Therefore, it is important for bees to determine whether individuals trying to enter the nest are group members or intruders. While it is known that social insects use blends of waxy chemicals called cuticular hydrocarbons to identify group members at the entrance to the colony, it is not clear how members of the same colony acquire a similar blend of cuticular hydrocarbons. Some previous work suggested that in some ant species (which are also social insects), colony members exchange cuticular hydrocarbons with each other so that all members of the colony are covered with a similar blend of chemicals. However, it was not known whether honey bees also share cuticular hydrocarbons between colony members in order to identify members of a hive. Vernier et al. used chemical, molecular and behavioral approaches to study the cuticular hydrocarbons found on honey bees. The results show that, rather than exchanging chemicals with other members of their colony, individual bees make their own blends of cuticular hydrocarbons. As a bee ages it makes different blends of cuticular hydrocarbons, and by the time it starts to leave the hive to forage it makes a blend that is specific to the colony it belongs to. The production of this final blend is influenced by the environment within the hive. Thus, the findings of Vernier et al. indicate that honey bees guarding the entrance to a hive can only identify non-colony-member forager bees as intruders, rather than any non-colony-member bee that happens upon the hive entrance. Honey bees play an essential role in pollinating many crop plants so understanding how these insects maintain their social groups may help to improve agriculture in the future. Furthermore, this work may aid our understanding of how other social insects interact in a variety of biological situations.

Automated Video Monitoring of Unmarked and Marked Honey Bees at the Hive Entrance
We present a novel system for the automatic video monitoring of honey bee foraging activity at the hive entrance. This monitoring system is built upon convolutional neural networks that perform multiple animal pose estimation without the need for marking. This precise detection of honey bee body parts is a key element of the system to provide detection of entrance and exit events at the entrance of the hive including accurate pollen detection. A detailed evaluation of the quality of the detection and a study of the effect of the parameters are presented. The complete system also integrates identification of barcode marked bees, which enables the monitoring at both aggregate and individual levels. The results obtained on multiple days of video recordings show the applicability of the approach for large-scale deployment. This is an important step forward for the understanding of complex behaviors exhibited by honey bees and the automatic assessment of colony health.

Daniel, Sloan (Ed.)Abstract Recent declines in the health of the honey bee have startled researchers and lay people alike as honey bees are agriculture’s most important pollinator. Honey bees are important pollinators of many major crops and add billions of dollars annually to the US economy through their services. One factor that may influence colony health is the microbial community. Indeed, the honey bee worker digestive tract harbors a characteristic community of bee-specific microbes, and the composition of this community is known to impact honey bee health. However, the honey bee is a superorganism, a colony of eusocial insects with overlapping generations where nestmates cooperate, building a hive, gathering and storing food, and raising brood. In contrast to what is known regarding the honey bee worker gut microbiome, less is known of the microbes associated with developing brood, with food stores, and with the rest of the built hive environment. More recently, the microbe Bombella apis was identified as associated with nectar, with developing larvae, and with honey bee queens. This bacterium is related to flower-associated microbes such as Saccharibacter floricola and other species in the genus Saccharibacter, and initial phylogenetic analyses placed it as sister to these environmental bacteria. Here, we used comparative genomics of multiple honey bee-associated strains and the nectar-associated Saccharibacter to identify genomic changes that may be associated with the ecological transition to honey bee association. We identified several genomic differences in the honey bee-associated strains, including a complete CRISPR/Cas system. Many of the changes we note here are predicted to confer upon Bombella the ability to survive in royal jelly and defend themselves against mobile elements, including phages. Our results are a first step toward identifying potential function of this microbe in the honey bee superorganism.more » « less

Abstract Honey bees are vital pollinators and can be used to monitor the landscape. Consequently, interest in mounting technologies onto bees to track foraging behaviors is increasing. The barrier to entry is steep, in part because the methodology for fastening tags to bees, and the success rates, are often missing from publications. We tested six factors suspected to influence the presence and tag retention rates of nurse honey bees after their introduction to hives, and followed bees until foraging age. We also compared reintroducing foragers to their maternal colony using the best method for nurse bees to releasing them in front of their maternal hive and allowing them to fly back unaided. Nurses were most likely to be present in the hive with their tag still attached when introduced using an introduction cage at night. Glue type was important, but may further be influenced by tag material. Foragers were most likely to be present with a tag attached if released in front of their colony. Preparation and introduction techniques influence the likelihood of tagged honey bee survival and of the tags remaining attached, which should be considered when executing honey bee tagging and tracking experiments.more » « less

Abstract Honey bees are critical pollinators in ecosystems and agriculture, but their numbers have significantly declined. Declines in pollinator populations are thought to be due to multiple factors including habitat loss, climate change, increased vulnerability to disease and parasites, and pesticide use. Neonicotinoid pesticides are agonists of insect nicotinic cholinergic receptors, and sub-lethal exposures are linked to reduced honey bee hive survival. Honey bees are highly dependent on circadian clocks to regulate critical behaviors, such as foraging orientation and navigation, time-memory for food sources, sleep, and learning/memory processes. Because circadian clock neurons in insects receive light input through cholinergic signaling we tested for effects of neonicotinoids on honey bee circadian rhythms and sleep. Neonicotinoid ingestion by feeding over several days results in neonicotinoid accumulation in the bee brain, disrupts circadian rhythmicity in many individual bees, shifts the timing of behavioral circadian rhythms in bees that remain rhythmic, and impairs sleep. Neonicotinoids and light input act synergistically to disrupt bee circadian behavior, and neonicotinoids directly stimulate wake-promoting clock neurons in the fruit fly brain. Neonicotinoids disrupt honey bee circadian rhythms and sleep, likely by aberrant stimulation of clock neurons, to potentially impair honey bee navigation, time-memory, and social communication.more » « less

Abstract BackgroundHoney bees are not only essential for pollination services, but are also economically important as a source of hive products (e.g., honey, royal jelly, pollen, wax, and propolis) that are used as foods, cosmetics, and alternative medicines. Royal jelly is a popular honey bee product with multiple potential medicinal properties. To boost royal jelly production, a long-term genetic selection program of Italian honey bees (ITBs) in China has been performed, resulting in honey bee stocks (here referred to as RJBs) that produce an order of magnitude more royal jelly than ITBs. Although multiple studies have investigated the molecular basis of increased royal jelly yields, one factor that has not been considered is the role of honey bee-associated gut microbes. ResultsBased on the behavioral, morphological, physiological, and neurological differences between RJBs and ITBs, we predicted that the gut microbiome composition of RJBs bees would differ from ITBs. To test this hypothesis, we investigated the bacterial composition of RJB and ITB workers from an urban location and RJBs from a rural location in China. Based on 16S rRNA gene profiling, we did not find any evidence that RJBs possess a unique bacterial gut community when compared to ITBs. However, we observed differences between honey bees from the urban versus rural sites. ConclusionsOur results suggest that the environmental factors rather than stock differences are more important in shaping the bacterial composition in honey bee guts. Further studies are needed to investigate if the observed differences in relative abundance of taxa between the urban and rural bees correspond to distinct functional capabilities that impact honey bee health. Because the lifestyle, diet, and other environmental variables are different in rural and urban areas, controlled studies are needed to determine which of these factors are responsible for the observed differences in gut bacterial composition between urban and rural honeybees.more » « less
