Title: Evaluation of Endoplasmic Reticulum Targeting Isoquinol‐Based BODIPY Dyes and Their Properties as Molecular Rotors
Abstract

A series of six new isoquinol‐based BODIPY dyes have been synthesized by a two‐step method. Unlike traditional synthetic methods, synthesis did not require the use of oxidizing agents or solvent. Spectroscopic, electrochemical and DFT calculations indicate that the properties associated with the six dyes are very similar with little influence of themeso‐substituents. Viscosity studies identified three of the new dyes having properties of molecular rotors for applications in the detection of changes in cellular viscosity. Incubation of the dyes with the HEK cell line show no toxicity up to 10 μM over a 72 h. period. The dyes show the ability to localize within the endoplasmic reticulum (ER) of cells. With two of the dyes showing favorable comparisons to commercially available ER dyes.

  1. Abstract

    Phenolic acids are a class of poorly water‐soluble organic compounds with antioxidant properties that contain a carboxylic acid and a phenol group, common in plants. We used hydroxypropyl‐β‐cyclodextrin (HP‐β‐CD) as a supramolecular host to sense phenolic acids in water near physiological pH. The analytes’ complexation was monitored through changes in the optical absorption and emission properties of a series of six organic dyes, acting as indicators in a displacement assay, that were selected from screening a panel of 17 candidates. Binding constants between these dyes and the HP‐β‐CD were in the 102–104range. We showed that the nuanced differences in the dyes’ optical signatures associated with this displacement process, when analyzed and summarized using multivariate analysis algorithms such as principal component analysis (PCA), can be used to successfully discriminate among six phenolic acids, and to identify six unknown samples of such acids, down to a 0.02 mM concentration.

    Near‐infrared (NIR) fluorescent dyes with favorable photophysical properties are highly useful for bioimaging, but such dyes are still rare. The development of a unique class of NIR dyes via modifying the rhodol scaffold with fused tetrahydroquinoxaline rings is described. These new dyes showed large Stokes shifts (>110 nm). Among them, WR3, WR4, WR5, and WR6 displayed high fluorescence quantum yields and excellent photostability in aqueous solutions. Moreover, their fluorescence properties were tunable by easy modifications on the phenolic hydroxy group. Based on WR6, two NIR fluorescent turn‐on probes, WSP‐NIR and SeSP‐NIR, were devised for the detection of H2S. The probe SeSP‐NIR was applied in visualizing intracellular H2S. These dyes are expected to be useful fluorophore scaffolds in the development of new NIR probes for bioimaging.

  4. Abstract

    Development of latent prints employing cyanoacrylate ester (CA) can be a multistep process including CA fuming and subsequent fluorescent staining to produce fingerprints of sufficient contrast for comparison work. To enable a single‐step CA fuming—staining process, a selection of fluorophores have been developed as sublimation dyes in CA fuming. A greater array of such luminescent sublimation dyes would allow users greater flexibility in selecting a particular dye—CA combination to best suit their processing needs. Toward this end, six benzoic acid derivatives were evaluated for use as luminescent sublimation dyes under elementary CA fuming conditions using a single non‐porous surface type and an inexpensive handheld UV lamp for excitation. Two benzoic acid derivatives, 2‐hydroxybenzoic acid (salicylic acid) and 2‐aminobenzoic acid (anthranilic acid), were identified as new potential luminescent sublimation dyes with stained fingerprints excited at 254 nm. The fluorescence intensity and stability of prints produced via the sublimation of CA with 2‐hydroxybenzoic acid and 2‐aminobenzoic acid were evaluated over approximately six weeks using image and statistical analysis.

  5. Abstract

    Dibenzothiophene 5,5‐dioxide (DBTOO) derivatives have recently been shown to processes utility as fluorescent cell dyes. In an effort to extend the functionality of DBTOO‐based dyes to include the visualization of cellular membranes, two lipophilic DBTOO were synthesized and their ability to incorporate into the plasma membrane of HeLa cells was examined by fluorescent microscopy. The photophysical properties of the two new DBTOO derivatives were determined and both have good fluorescent quantum yields and a visible blue emission. Due to agreeable wavelengths of excitation and emission, a standard 4′,6‐diamindino‐2‐phenylindole (DAPI) filter set worked well with these dyes. After co‐staining, it was confirmed that both DBTOO dyes localized in the plasma membrane. The quality of the overlap was quantified using Pearson correlation coefficient, which indicated a strong overlap between the DBTOO dyes and the standard plasma membrane dye. The novel dyes also displayed relatively low toxicity to the HeLa cells with IC50between 10 and 100 µm. Thus, this work reports a new use of DBTOO derivatives as fluorescent microscopy stains.
