Experimental studies reveal that the anionic lipid phosphatidic acid (POPA), non-phospholipid cholesterol, and cationic lipid DOTAP inhibit the gating of voltage-sensitive potassium (Kv) channels. Here, we develop a continuum electromechanical model to investigate the interaction of these lipids with the ion channel. Our model suggests that: (i) POPA lipids may restrict the vertical motion of the voltage-sensor domain through direct electrostatic interactions; (ii) cholesterol may oppose the radial motion of the pore domain of the channel by increasing the mechanical rigidity of the membrane; and (iii) DOTAP can reduce the effect of electrostatic forces by regulating the dielectric constant at the channel–lipid interface. The electromechanical model predictions for the three lipid types match well with the experimental observations and provide mechanistic insights into lipid-dependent gating of Kv channels.

                            3D-printed machines that manipulate microscopic objects using capillary forces
                        
                    
    
            Objects that deform a liquid interface are subject to capillary forces, which can be harnessed to assemble the objects1–4. Once assembled, such structures are generally static. Here we dynamically modulate these forces to move objects in programmable two-dimensional patterns. We 3D-print devices containing channels that trap floating objects using repulsive capillary forces5,6, then move these devices vertically in a water bath. Because the channel cross-sections vary with height, the trapped objects can be steered in two dimensions. The device and interface therefore constitute a simple machine that converts vertical to lateral motion. We design machines that translate, rotate and separate multiple floating objects and that do work on submerged objects through cyclic vertical motion. We combine these elementary machines to make centimetre-scale compound machines that braid micrometre-scale filaments into prescribed topologies, including non-repeating braids. Capillary machines are distinct from mechanical, optical or fluidic micromanipulators in that a meniscus links the object to the machine. Therefore, the channel shapes need only be controlled on the scale of the capillary length (a few millimetres), even when the objects are microscopic. Consequently, such machines can be built quickly and inexpensively. This approach could be used to manipulate micrometre-scale particles or to braid microwires for high-frequency electronics.

            Time-varying fields drive the motion of magnetic particles adsorbed on liquid drops due to interfacial constraints that couple magnetic torques to capillary forces. Such magneto-capillary particle dynamics and the associated fluid flows are potentially useful for propelling drop motion, mixing drop contents, and enhancing mass transfer between phases. The design of such functions benefits from the development and validation of predictive models. Here, we apply methods of Bayesian data analysis to identify and validate a dynamical model that accurately predicts the field-driven motion of a magnetic particle adsorbed at the interface of a spherical droplet. Building on previous work, we consider candidate models that describe particle tilting at the interface, field-dependent contributions to the magnetic moment, gravitational forces, and their combinations. The analysis of each candidate is informed by particle tracking data for a magnetic Janus sphere moving in a precessing field at different frequencies and angles. We infer the uncertain parameters of each model, criticize their ability to describe and predict experimental data, and select the most probable candidate, which accounts for gravitational forces and the tilting of the Janus sphere at the interface. We show how this favored model can predict complex particle trajectories with micron-level accuracy across the range of driving fields considered. We discuss how knowledge of this “best” model can be used to design experiments that inform accurate parameter estimates or achieve desired particle trajectories.more » « less

            Some of the most important tasks of visual and motor systems involve estimating the motion of objects and tracking them over time. Such systems evolved to meet the behavioral needs of the organism in its natural environment, and may therefore be adapted to the statistics of motion it is likely to encounter. By tracking the movement of individual points in movies of natural scenes, we begin to identify common properties of natural motion across scenes. As expected, objects in natural scenes move in a persistent fashion, with velocity correlations lasting hundreds of milliseconds. More subtly, but crucially, we find that the observed velocity distributions are heavy-tailed and can be modeled as a Gaussian scale-mixture. Extending this model to the time domain leads to a dynamic scale-mixture model, consisting of a Gaussian process multiplied by a positive scalar quantity with its own independent dynamics. Dynamic scaling of velocity arises naturally as a consequence of changes in object distance from the observer, and may approximate the effects of changes in other parameters governing the motion in a given scene. This modeling and estimation framework has implications for the neurobiology of sensory and motor systems, which need to cope with these fluctuations in scale in order to represent motion efficiently and drive fast and accurate tracking behavior.more » « less

            Abstract Soft intelligent structures that are programmed to reshape and reconfigure under magnetic field can find applications such as in soft robotics and biomedical devices. Here, a new class of smart elastomeric architectures that undergo complex reconfiguration and shape change in applied magnetic fields, while floating on the surface of water, is reported. These magnetoactive soft actuators are fabricated by 3D printing with homocomposite silicone capillary ink. The ultrasoft actuators easily deform by the magnetic force exerted on carbonyl iron particles embedded in the silicone, as well as lateral capillary forces. The tensile and compressive moduli of the actuators are easily determined by their topological design through 3D printing. As a result, their responses can be engineered by the interplay of the intensity of the magnetic field gradient and the programmable moduli. 3D printing allows us to fabricate soft architectures with different actuation modes, such as isotropic/anisotropic contraction and multiple shape changes, as well as functional reconfiguration. Meshes that reconfigure in magnetic fields and respond to external stimuli by reshaping could serve as active tissue scaffolds for cell cultures and soft robots mimicking creatures that live on the surface of water.more » « less

            Abstract A key challenge underlying the design of miniature machines is encoding materials with time‐ and space‐specific functional behaviors that require little human intervention. Dissipative processes that drive materials beyond equilibrium and evolve continuously with time and location represent one promising strategy to achieve such complex functions. This work reports how internal nonequilibrium states of liquid crystal (LC) emulsion droplets undergoing chemotaxis can be used to time the delivery of a chemical agent to a targeted location. During ballistic motion, hydrodynamic shear forces dominate LC elastic interactions, dispersing microdroplet inclusions (microcargo) within double emulsion droplets. Scale‐dependent colloidal forces then hinder the escape of dispersed microcargo from the propelling droplet. Upon arrival at the targeted location, a circulatory flow of diminished strength allows the microcargo to cluster within the LC elastic environment such that hydrodynamic forces grow to exceed colloidal forces and thus trigger the escape of the microcargo. This work illustrates the utility of the approach by using microcargo that initiate polymerization upon release through the outer interface of the carrier droplet. These findings provide a platform that utilizes nonequilibrium strategies to design autonomous spatial and temporal functions into active materials.more » « less
