We present a mathematical model of lophotrichous bacteria, motivated by Pseudomonas putida, which swim through fluid by rotating a cluster of multiple flagella extended from near one pole of the cell body. Although the flagella rotate individually, they are typically bundled together, enabling the bacterium to exhibit three primary modes of motility: push, pull, and wrapping. One key determinant of these modes is the coordination between motor torque and rotational direction of motors. The computational variations in this coordination reveal a wide spectrum of dynamical motion regimes, which are modulated by hydrodynamic interactions between flagellar filaments. These dynamic modes can be categorized into two groups based on the collective behavior of flagella, i.e., bundled and unbundled configurations. For some of these configurations, experimental examples from fluorescence microscopy recordings of swimming P. putida cells are also presented. Furthermore, we analyze the characteristics of stable bundles, such as push and pull, and investigate the dependence of swimming behaviors on the elastic properties of the flagella.more » « less

Plasmids are extrachromosomal DNA molecules that can be transferred vertically as well as horizontally between bacteria. They have been shown to code for numerous bacterial phenotypes, the most concerning of which is resistance to commonly used antibiotics. Plasmid-mediated transfer of drug resistance genes has notably led to the rapid spread of resistant strains, which now presents a human health crisis. While this rapid evolution of bacteria due to horizontal plasmid transfer has historically dominated this research field, increasing attention is being paid to the (co-)evolutionary interactions that shape the long-term population dynamics of bacteria and plasmids.more » « less

Chemotaxis is the ability of certain microscopic organisms to sense and swim towards beneficial or away from detrimental chemicals in their surroundings. Identifying this behavior is important for understanding the relationships between species and their environments in the natural world. Predicting migration of an entire population from known characteristics of individual microorganisms is a key contribution, but can be a laborious process and requires watching and waiting for visual evidence of the process on a large population scale. Sonification offers a novel solution to this problem by allowing the observer to tap into our auditory sensory system to process information. In this project, we developed and assessed a proof-of-concept sonification tool as a high throughput, real-time screening tool for chemotaxis in populations of swimming bacteria. The tool operates by reporting the y-axis position of bacteria that appear in the microscope image as microsecond duration pitched notes, giving the user a sense of the average location of the population. In this paper, we present how it has been used as a chemotaxis assay and as a tool to locate traveling waves of bacteria as they pass through the field of view in order to capture data at specific timepoints, which is used to analyze individual swimming patterns of microorganisms within the wave.more » « less
