Title: Effects of repeated intragastric administrations with heat-inactivated Mycobacterium aurum DSM 33539 on the stress-induced aggravation of dextran sulfate sodium (DSS) colitis in C57BL/6N mice
Stress-protective effects have been reported forM. vaccaeNCTC 11659 andM. vaccaeATCC 15483T. However, it remains to be investigated whether also closely related rapidly growing environmental saprophytic non-tuberculous mycobacteria (NTM) species have protective effects against the negative consequences of chronic psychosocial stress. Therefore, the aim of the current study was to assess whether repeated i.g. administrations of a heat-inactivated preparation ofMycobacterium aurumDSM 33539 prior to 19 days of chronic subordinate colony housing (CSC) are able to ameliorate the negative effects of this preclinically validated mouse model for chronic psychosocial stress on subsequent dextran sulfate sodium (DSS) colitis in male C57BL/6N mice. The results of the present study show that repeated i.g. administrations ofM. aurumDSM 33539 have stabilizing effects on the composition of the gut microbiome, indicated by the findings thatM. aurumDSM 33539 prevented CSC-induced increases in the relative abundances of the colitogenic phyla Desulfobacterota and Deferribacterota. Indeed, the relative abundance of Deferribacterota on day 19 was strongly correlated with histological damage to the colon. In line with the latter,M. aurumDSM 33539 was further protective against the aggravating effects of stress on subsequent DSS colitis. Collectively, our findings confirm and extend previous findings from our group and suggest that the stress-protective effects reported forM. vaccaeNCTC 11659 andM. vaccaeATCC 15483Tare generalizable also to other NTM species.  more » « less

    ABSTRACT Inflammatory bowel diseases (IBDs) are devastating conditions of the gastrointestinal tract with limited treatments, and dietary intervention may be effective and affordable for managing symptoms. Glucosinolate compounds are highly concentrated in broccoli sprouts, especially glucoraphanin (GLR), and can be metabolized by certain mammalian gut bacteria into anti-inflammatory isothiocyanates, such as sulforaphane. Gut microbiota exhibit biogeographic patterns, but it is unknown if colitis alters these or whether the location of glucoraphanin-metabolizing bacteria affects anti-inflammatory benefits. We fed specific pathogen-free C57BL/6 mice either a control diet or a 10% steamed broccoli sprout diet and gave a three-cycle regimen of 2.5% dextran sodium sulfate (DSS) in drinking water over a 34-day experiment to simulate chronic, relapsing ulcerative colitis (UC). We monitored body weight, fecal characteristics, lipocalin, serum cytokines, and bacterial communities from the luminal- and mucosal-associated populations in the jejunum, cecum, and colon. Mice fed the broccoli sprout diet with DSS treatment performed better than mice fed the control diet with DSS, and had significantly more weight gain, lower Disease Activity Index scores, lower plasma lipocalin and proinflammatory cytokines, and higher bacterial richness in all gut locations. Bacterial communities were assorted by gut location but were more homogenous across locations in the control diet + DSS mice. Importantly, our results showed that broccoli sprout feeding abrogated the effects of DSS on gut microbiota, as bacterial richness and biogeography were similar between mice receiving broccoli sprouts with and without DSS. Collectively, these results support the protective effect of steamed broccoli sprouts against dysbiosis and colitis induced by DSS. IMPORTANCEEvaluating bacterial communities across different locations in the gut provides a greater insight than fecal samples alone and provides an additional metric by which to evaluate beneficial host-microbe interactions. Here, we show that 10% steamed broccoli sprouts in the diet protects mice from the negative effects of dextran sodium sulfate-induced colitis, that colitis erases biogeographic patterns of bacterial communities in the gut, and that the cecum is not likely to be a significant contributor to colonic bacteria of interest in the DSS mouse model of ulcerative colitis. Mice fed the broccoli sprout diet during colitis performed better than mice fed the control diet while receiving DSS. The identification of accessible dietary components and concentrations that help maintain and correct the gut microbiome may provide universal and equitable approaches to IBD prevention and recovery, and broccoli sprouts represent a promising strategy.

  2. Introduction Stress in relation to the Coronavirus disease 19 pandemic (i.e., COVID-19, COVID stress) may be linked with poor sleep quality. The association between stress that is specific to the COVID-19 pandemic and sleep quality has been understudied, particularly in racially diverse people across the adult lifespan. Here, we investigated self-reported sleep quality in relation to COVID stress and factors that may protect against experiencing poor sleep quality from high COVID stress, including social support and religiosity. Method We recruited non-Hispanic Black ( n = 73) and non-Hispanic White ( n = 178) participants across the adult lifespan (18–76 years) using an online, cross-sectional design during the COVID-19 pandemic (March 2021–June 2021). We asked participants to report information regarding demographics (age, race/ethnicity, years of education), sleep (sleep quality, sleep habits), and positive (social support, religious activities) and negative (events of discrimination, depression, general stress, COVID stress) psychosocial factors. Results Across age and racial groups, better sleep habits were associated with better sleep quality, and higher COVID stress was linked to poorer sleep quality. Black participants reported higher quality sleep than White participants ( p = 0.006). They also endorsed greater private and internal religiosity ( p’s < 0.001). Across racial groups, moderation analyses revealed a protective effect of religiosity against poor sleep ( p’s < 0.006). Specifically, individuals with high religious activity and high COVID stress did not experience poor sleep quality, but individuals with low religious activity and high COVID stress demonstrated poor sleep quality. These results remained significant when controlling for general stress. Discussion Protective factors, such as religiosity, may mitigate the negative associations between high COVID stress and poor sleep quality.

  3. ABSTRACT ObjectiveInflammation may be an integral physiological mechanism through which discrimination impacts cardiovascular health and contributes to racial health disparities. Limited research has examined psychosocial factors that protect against the negative effects of discrimination on inflammation. Perceived control is a promising possible protective factor, given that it has been shown to moderate the relationship between other psychosocial stressors and physiological outcomes. This study thus tested whether systemic inflammation mediated the link between discrimination and cardiovascular health and whether perceived control moderated this relationship. MethodsData for this project included 347 non-Hispanic/Latinx Black adults (mean [standard deviation] age = 51.64 [11.24] years; 33% female) taken from the Midlife in the United States study. Perceived control and daily discrimination were assessed via self-report, and inflammation was measured via circulating levels of C-reactive protein (CRP), interleukin-6 (IL-6), fibrinogen, and tumor necrosis factor α. Cardiovascular health was measured by morbidity of cardiovascular conditions: heart disease, hypertension, and/or stroke. ResultsCRP (indirect effect:b =0.004, 95% confidence interval [CI] = 0.001–0.007) and fibrinogen (indirect effect:b =0.002, 95% CI = 0.0003–0.005) mediated the link between discrimination and cardiovascular conditions. Perceived control moderated the relationship between discrimination and CRP (F(1, 293) = 4.58, ΔR2= 0.013,b= −0.02, SE = 0.01,p= .033). CRP mediated the link between discrimination and cardiovascular conditions only for those who reported low levels of perceived control (Index = −0.003, 95% CI = −0.007 to −0.0001). ConclusionFindings provide empirical evidence of inflammation as a mechanism linking discrimination to cardiovascular conditions among Black Americans. Additionally, perceived control may be protective. Findings could suggest beliefs about control as a potential intervention target to help reduce the negative effects of discrimination on cardiovascular health among Black Americans.

A Gram-stain-negative, strictly anaerobic, non-motile, rod-shaped bacterium, designated SFB93T, was isolated from the intertidal sediments of South San Francisco Bay, located near Palo Alto, CA, USA. SFB93Twas capable of acetylenotrophic and diazotrophic growth, grew at 22–37 °C, pH 6.3–8.5 and in the presence of 10–45 g l−1NaCl. Phylogenetic analyses based on 16S rRNA gene sequencing showed that SFB93Trepresented a member of the genusSyntrophotaleawith highest 16S rRNA gene sequence similarities toSyntrophotalea acetylenicaDSM 3246T(96.6 %),Syntrophotalea carbinolicaDSM 2380T(96.5 %), andSyntrophotalea venetianaDSM 2394T(96.7 %). Genome sequencing revealed a genome size of 3.22 Mbp and a DNA G+C content of 53.4 %. SFB93Thad low genome-wide average nucleotide identity (81–87.5 %) and <70 % digital DNA–DNA hybridization value with other members of the genusSyntrophotalea. The phylogenetic position of SFB93Twithin the familySyntrophotaleaceaeand as a novel member of the genusSyntrophotaleawas confirmed via phylogenetic reconstruction based on concatenated alignments of 92 bacterial core genes. On the basis of the results of phenotypic, genotypic and phylogenetic analyses, a novel species,Syntrophotalea acetylenivoranssp. nov., is proposed, with SFB93T(=DSM 106009T=JCM 33327T=ATCC TSD-118T) as the type strain.

Abstract ObjectivesThe COVID‐19 pandemic in South Africa introduced new societal adversities and mental health threats in a country where one in three individuals are expected to develop a psychiatric condition sometime in their life. Scientists have suggested that psychosocial stress and trauma during childhood may increase one's vulnerability to the mental health consequences of future stressors—a process known as stress sensitization. This prospective analysis assessed whether childhood adversity experienced among South African children across the first 18 years of life, coinciding with the post‐apartheid transition, exacerbates the mental health impacts of psychosocial stress experienced during the 2019 coronavirus (COVID‐19) pandemic (ca. 2020–2021). Materials and MethodsData came from 88 adults who participated in a follow‐up study of a longitudinal birth cohort study in Soweto, South Africa. Childhood adversity and COVID‐19 psychosocial stress were assessed as primary predictors of adult PTSD risk, and an interaction term between childhood adversity and COVID‐19 stress was calculated to evaluate the potential effect of stress sensitization. ResultsFifty‐six percent of adults exhibited moderate‐to‐severe PTSD symptoms. Greater childhood adversity and higher COVID‐19 psychosocial stress independently predicted worse post‐traumatic stress disorder symptoms in adults. Adults who reported greater childhood adversity exhibited non‐significantly worse PTSD symptoms from COVID‐19 psychosocial stress. DiscussionThese results highlight the deleterious mental health effects of both childhood trauma and COVID‐19 psychosocial stress in our sample and emphasize the need for greater and more accessible mental health support as the pandemic progresses in South Africa.
