Although domestic dogs were the first domesticated species, the nature of dog domestication remains a topic of ongoing debate. In particular, brain and behavior changes associated with different stages of the domestication process have been difficult to disambiguate. Most modern Western breed dogs possess highly derived physical and behavioral traits because of intense artificial selection for appearance and function within the past 200 years. In contrast, premodern dogs, including primitive/ancient breeds, village dogs, and New Guinea Singing Dogs, have undergone less intensive artificial selection and retain more ancestral characteristics. Consequently, comparisons between modern and premodern dogs can shed light on brain and behavior changes that have occurred recently in the domestication process. Here, we addressed this question using a voxel-based morphometry analysis of structural MRI images from 72 modern breed dogs and 13 premodern dogs (32 females). Modern breed dogs show widespread expansions of neocortex and reductions in the amygdala and other subcortical regions. Furthermore, cortical measurements significantly predicted individual variation in trainability, while amygdala measurements significantly predicted fear scores. These results contrast with the long-standing view that domestication consistently involves reduction in brain size and cognitive capacity. Rather, our results suggest that recent artificial selection has targeted higher-order brain regions in modern breed dogs, perhaps to facilitate behavioral flexibility and close interaction and cooperation with humans.

Influence of early life adversity and breed on aggression and fear in dogs
Abstract Among the animals on this planet, dogs are uniquely adapted for life with humans, a status that exposes them to risks of human-mediated traumatic experiences. At the same time, some lineages of dogs have undergone artificial selection for behavioral phenotypes that might increase risk or resilience to stress exposure, providing an opportunity to examine interactions between heritable and acquired traits. In a large-scale study (N = 4,497), English-speaking dog guardians reported on their dogs’ life histories, current living environments, and provided observer ratings of dog behavior using the Canine Behavior Assessment and Research Questionnaire (C-BARQ). Our analysis revealed that adverse experiences in the first six months of life, such as abuse and relinquishment, were significantly associated with increased aggression and fearfulness in adulthood, even when accounting for factors such as acquisition source, sex, and neuter status. Additionally, effects of adversity on fearful and aggressive behavior systematically varied at the breed level, suggesting heritable factors for risk and resilience for developing particular phenotypes. Our findings establish that breed ancestry and individual experience interact to show fear and aggressive behavior in pet dogs, confirming that socioemotional behavior is shaped by gene-environment interactions.

Underlying observed morphological differences, there must also be differences at other levels of organization that could lead to elucidating aging rates and life span disparities between wild and domestic canids. Furthermore, small-breed dogs live significantly longer lives than large-breed dogs, while having higher mass-specific metabolic rates and faster growth rates. At the cellular level, a clear mechanism underlying whole animal traits has not been fully elucidated, although oxidative stress has been implicated as a potential culprit of the disparate life spans of domestic dogs. We used plasma and red blood cells from known aged domestic dogs and wild canids, and measured several oxidative stress variables: total antioxidant capacity (TAC), lipid damage, and enzymatic activities of catalase, superoxide dismutase, and glutathione peroxidase (GPx). We used phylogenetically informed general linear mixed models and nonphylogenetically corrected linear regression analysis. We found that lipid damage increases with age in domestic dogs, whereas TAC increases with age and TAC and GPx activity increases as a function of age/maximum life span in wild canids, which may partly explain longer potential life spans in wolves. As body mass increases, TAC and GPx activity increase in wild canids, but not domestic dogs, highlighting that artificial selection may have decreased antioxidant capacity in domestic dogs. We found that small-breed dogs have significantly higher circulating lipid damage compared with large-breed dogs, concomitant to their high mass-specific metabolism and higher growth rates, but in opposition to their long life spans.more » « less

Does the head tilt observed in many domesticated dogs index lateralized language processing? To answer this question, the present study evaluated household dogs responding to four conditions in which owners provided an increasing number of communicative cues. These cues ranged from no communicative/affective cues to rich affective cues coupled with dog-directed speech. Dogs’ facial responses were first coded manually using the Dog Facial Action Coding System (DogFACS), followed by an in-depth investigation of head tilt behavior, in which AI-based automated analysis of head tilt and audio analysis of acoustic features extracted from communicative cues were implemented. In a sample of 103 dogs representing seven breed groups and mixed-breed dogs, we found significant differences in the number of head tilts occurring between conditions, with the most communicative (last) condition eliciting the most head tilts. There were also significant differences in the direction of the head tilts and between sex groups. Dogs were more likely to tilt their heads to the right, and neutered male dogs were more likely to tilt their heads than spayed female dogs. The right-tilt bias is consistent with left-hemisphere language processing in humans, with males processing language in a more lateralized manner, and females processing language more bilaterally—a pattern also observed in humans. Understanding the canine brain is important to both evolutionary research through a comparative lens, and in understanding our interspecies relationship.more » « less

A group of diseases have been shown to correlate with a phenomenon called microbiome dysbiosis, where the bacterial species composition of the gut becomes abnormal. The gut microbiome of an animal is influenced by many factors including diet, exposures to bacteria during post-gestational growth, lifestyle, and disease status. Studies also show that host genetics can affect microbiome composition. We sought to test whether host genetic background is associated with gut microbiome composition in the Norwegian Lundehund dog, a highly inbred breed with an effective population size of 13 individuals. The Lundehund has a high rate of a protein-losing enteropathy in the small intestine that is often reported as Lundehund syndrome, which negatively affects longevity and life-quality. An outcrossing project with the Buhund, Norrbottenspets, and Icelandic sheepdog was recently established to reintroduce genetic diversity to the Lundehund and improve its health. To assess whether there was an association between host genetic diversity and the microbiome composition, we sampled the fecal microbiomes of 75 dogs of the parental (Lundehund), F1 (Lundehund x Buhund), and F2 (F1 x Lundehund) generations. We found significant variation in microbiome composition from the parental Lundehund generation compared to the outcross progeny. The variation observed in purebred Lundehunds corresponded to dysbiosis as seen by a highly variable microbiome composition with an elevated Firmicutes to Bacteroidetes ratio and an increase in the prevalence of Streptococcus bovis/Streptococcus equinus complex, a known pathobiont that can cause several diseases. We tracked several other environmental factors including diet, the presence of a cat in the household, living in a farm and the use of probiotics, but we did not find evidence of an effect of these on microbiome composition and alpha diversity. In conclusion, we found an association between host genetics and gut microbiome composition, which in turn may be associated with the high incidence of Lundehund syndrome in the purebred parental dogs.more » « less

Abstract Joint intentionality, the mutual understanding of shared goals or actions to partake in a common task, is considered an essential building block of theory of mind in humans. Domesticated dogs are unusually adept at comprehending human social cues and cooperating with humans, making it possible that they possess behavioral signatures of joint intentionality in interactions with humans. Horschler and colleagues (Anim Behav 183: 159–168, 2022) examined joint intentionality in a service dog population, finding that upon interruption of a joint experience, dogs preferentially re-engaged their former partner over a passive bystander, a behavior argued to be a signature of joint intentionality in human children. In the current study, we aimed to replicate and extend these results in pet dogs. One familiar person played with the dog and then abruptly stopped. We examined if dogs would preferentially re-engage the player instead of a familiar bystander who was also present. Consistent with the findings of Horschler and colleagues (Anim Behav 183: 159–168, 2022), pet dogs preferentially gazed toward and offered the toy to the player significantly more than the familiar bystander. However, no difference was observed in physical contact. These findings provide preliminary evidence for behavioral signatures of joint intentionality in pet dogs, but future work is needed to understand whether this phenomenon extends to other contexts.more » « less
