Aromatic electron-deficient heterocycles, such as pyridines, are found in many biologically relevant structures, including those with medicinal applications. Methods for their substitution can streamline the synthesis of valuable molecules and allow access to unexplored chemical space. However, enantioselective methods for these derivatizations remain lacking, especially at remote stereocenters. Here, we present a photoenzymatic reaction for the reductive coupling of electron-deficient heterocycles with alkenes using flavin-dependent “ene”-reductases. This transformation results in the generation of a γ-stereocenter with high enantioselectivity. We propose that this light-driven transformation proceeds via excitation of a transient enzyme–substrate complex, enabling the enzyme to access the reductive potential needed for radical initiation when the substrates are bound in the active site. This work represents a stereoselective method for synthesizing derivatives of pyridine and similar heterocycles and an expansion of the substrate capabilities of “ene”-reductases in chemical synthesis.

Photodriven Sm-Catalyzed Asymmetric Ketyl-Olefin Coupling
SmI2 is a privileged, single-electron reductant employed in the synthesis of diverse products. Though traditionally used in (super)stoichiometric amounts, SmI2 has recently been employed catalytically with viable turnover strategies. Enantioselective reductive cross-coupling reactions mediated by SmI2 are scarce and asymmetric methods that use catalytic quantities of SmI2 are unknown. Herein, we report an enantioselective ketyl-olefin coupling, employing a SmI2-derived catalyst with a widely available chiral pyridine-bis(oxazoline) (PyBOX) ligand and a commercially available Ir photocatalyst. A combination of optical, electrochemical, and computational data reveals that a Sm-stabilized PyBOX radical is the reactive species responsible for initiating the reductive coupling step.

Ni-catalyzed asymmetric reductive cross-coupling reactions provide rapid and modular access to enantioenriched building blocks from simple electrophile precursors. Reductive coupling reactions that can diverge through a common organometallic intermediate to two distinct families of enantioenriched products are particularly versatile but underdeveloped. Here, we describe the development of a bis(oxazoline) ligand that enables the desymmetrization of meso-anhydrides. When secondary benzylic electrophiles are employed, doubly stereoselective acyl cross-coupling proceeds to give ketone products with catalyst control over three newly formed stereogenic centers. Alternatively, the use of primary alkyl halides in the presence of an additional halogen atom transfer catalyst results in decarbonylative alkylation to give enantioenriched β-alkyl acids. Analysis of reaction rates for a range of both catalysts and substrates supports the notion that tuning the different electrophile activation steps with the two catalysts is required for enhanced reaction performance. These studies illustrate how reaction design can diverge a common Ni-acyl intermediate to either acyl or decarbonylative coupling products and highlight how dual ligand systems can be used to engage unactivated alkyl halides in Ni-catalyzed asymmetric reductive coupling.more » « less

This article describes the first enantioselective synthesis of the Tasmanian marine alkaloid (+)-cylindricine B. The concise construction of the compound hinged on dearomative retrosynthetic logic combined with a tactical advance in the generation of congested, cyclic, alpha-tertiary amine centers. The scope of this key coupling reaction was explored in addition to providing a synthetic application for Cu-catalyzed enantioselective dearomatization of N-acyl-pyridiniums. The synthesis proceeds in five or six steps from commercially available starting materials.more » « less

Abstract Catalyst‐controlled divergent transformations represent a vital strategy for the efficient production of structurally diverse molecules from the same readily available precursors. Herein, we report the palladium/phosphine‐catalyst‐driven chemo‐ and regiodivergent allyl‐propargyl cross‐coupling of propargyl carbonates with allyl boronates. Density functional theory (DFT) calculations reveal that the bidentate phosphine ligand plays a crucial role in determining the relative favorability of possible reductive elimination modes from the ƞ3‐allyl‐ƞ1‐allenyl palladium intermediate species. Specifically, MeO‐BIPHEP facilitates the 1,1′‐reductive elimination to form the 1,4‐enallenes; in contrast, XantPhos favors the formation of 1,5‐enynes through a 3,3′‐reductive elimination pathway.more » « less

The first transition metal mediated reductive coupling of diazoesters through the terminal nitrogens is reported. The resulting tetrazene-bridged bis(diazenylacetate) serves as a novel dinucleating ligand to iron(III). DFT calculations suggest that the reductive coupling takes place via a κ 2 intermediate, which induces significant radical character on the terminal nitrogen.more » « less
