Three cyclopentadienylmolybdenum(II) propionyl complexes featuring triarylphosphine ligands with different para substituents, namely, dicarbonyl(η 5 -cyclopentadienyl)propionyl(triphenylphosphane-κ P )molybdenum(II), [Mo(C 5 H 5 )(C 3 H 5 O)(C 18 H 15 P)(CO) 2 ], ( 1 ), dicarbonyl(η 5 -cyclopentadienyl)propionyl[tris(4-fluorophenyl)phosphane-κ P ]molybdenum(II), [Mo(C 5 H 5 )(C 3 H 5 O)(C 18 H 12 F 3 P)(CO) 2 ], ( 2 ), and dicarbonyl(η 5 -cyclopentadienyl)propionyl[tris(4-methoxyphenyl)phosphane-κ P ]molybdenum(II) dichloromethane solvate, [Mo(C 5 H 5 )(C 3 H 5 O)(C 21 H 21 O 3 P)(CO) 2 ]·CH 2 Cl 2 , ( 3 ), have been prepared from the corresponding ethyl complexes via phosphine-induced migratory insertion. These complexes exhibit four-legged piano-stool geometries with molecular structures quite similar to each other and to related acetyl complexes. The extended structures of the three complexes differ somewhat, with the para substituent of the triarylphosphine of ( 2 ) (fluoro) or ( 3 ) (methoxy) engaging in non-classical C—H...F or C—H...O hydrogen-bonding interactions. The structure of ( 3 ) exhibits modest disorder in the position of one Cl atom of the dichloromethane solvent, which was modeled with two sites showing approximately equivalent occupancies [0.532 (15) and 0.478 (15)].more » « less

PURPOSE Lehmann et al have identified four molecular subtypes of triple-negative breast cancer (TNBC)—basal-like (BL) 1, BL2, mesenchymal (M), and luminal androgen receptor—and an immunomodulatory (IM) gene expression signature modifier. Our group previously showed that the response of TNBC to neoadjuvant systemic chemotherapy (NST) differs by molecular subtype, but whether NST affects the subtype was unknown. Here, we tested the hypothesis that in patients without pathologic complete response, TNBC subtypes can change after NST. Moreover, in cases with the changed subtype, we determined whether epithelial-to-mesenchymal transition (EMT) had occurred. MATERIALS AND METHODS From the Pan-Pacific TNBC Consortium data set containing TNBC patient samples from four countries, we examined 64 formalin-fixed, paraffin-embedded pairs of matched pre- and post-NST tumor samples. The TNBC subtype was determined using the TNBCtype-IM assay. We analyzed a partial EMT gene expression scoring metric using mRNA data. RESULTS Of the 64 matched pairs, 36 (56%) showed a change in the TNBC subtype after NST. The most frequent change was from BL1 to M subtypes (38%). No tumors changed from M to BL1. The IM signature was positive in 14 (22%) patients before NST and eight (12.5%) patients after NST. The EMT score increased after NST in 28 (78%) of the 36 patients with the changed subtype ( v 39% of the 28 patients without change; P = .002254). CONCLUSION We report, to our knowledge, for the first time that the TNBC molecular subtype and IM signature frequently change after NST. Our results also suggest that EMT is promoted by NST. Our findings may lead to innovative adjuvant therapy strategies in TNBC cases with residual tumor after NST.more » « less

Direct numerical simulation (DNS) of a transitional boundary layer over a plate with an elliptical leading edge. Navier-Stokes was discretized on a curvilinear grid and solved using a finite volume DNS code. A fractional-step algorithm was adopted, and the spatial discretization was a staggered volume-flux formulation. The viscous terms were integrated in time implicitly using the Crank-Nicolson and the advections terms were treated explicitly using the Adams-Bashforth. Pressure was treated using implicit Euler in the δp-form. The pressure equation was Fourier transformed in the span, and the resulting Helmholtz equation was solved for every spanwise wavenumber using two-dimensional multi-grid. After the simulation has reached a statistical stationary state, 4701 frames of data, which includes the 3 components of the velocity vector and the pressure, are generated and written in files that can be accessed directly by the database (FileDB system). Since the grid is staggered, data at the wall are not stored in the database. However, JHTDB provides values in the region between the wall and the first grid point, y∈[0, 0.0036], using 4th-order Lagrange polynomial inter- and extrapolation. Abstract A cobalt silylene (Co=Si) linkage enables a distinct metal/ligand cooperative activation of an organic azide, where nitrene transfer occurs to and from the Co⋅⋅⋅Si linkage without ligand dissociation from the 18‐electron cobalt center. This process utilizes the orthogonal binding affinities of the silicon and cobalt sites to avoid CO poisoning that would otherwise inhibit reactivity, leading to significantly improved catalytic isocyanate generation compared with related systems. The dual‐site approach demonstrates the potential of metal/main‐group bonds to access new and efficient catalytic pathways.more » « less
