Title: Neuromuscular actuation of biohybrid motile bots
The integration of muscle cells with soft robotics in recent years has led to the development of biohybrid machines capable of untethered locomotion. A major frontier that currently remains unexplored is neuronal actuation and control of such muscle-powered biohybrid machines. As a step toward this goal, we present here a biohybrid swimmer driven by on-board neuromuscular units. The body of the swimmer consists of a free-standing soft scaffold, skeletal muscle tissue, and optogenetic stem cell-derived neural cluster containing motor neurons. Myoblasts embedded in extracellular matrix self-organize into a muscle tissue guided by the geometry of the scaffold, and the resulting muscle tissue is cocultured in situ with a neural cluster. Motor neurons then extend neurites selectively toward the muscle and innervate it, developing functional neuromuscular units. Based on this initial construct, we computationally designed, optimized, and implemented light-sensitive flagellar swimmers actuated by these neuromuscular units. Cyclic muscle contractions, induced by neural stimulation, drive time-irreversible flagellar dynamics, thereby providing thrust for untethered forward locomotion of the swimmer. Overall, this work demonstrates an example of a biohybrid robot implementing neuromuscular actuation and illustrates a path toward the forward design and control of neuron-enabled biohybrid machines.  more » « less

  1. The intriguing opportunities enabled by the use of living components in biological machines have spurred the development of a variety of muscle‐powered biohybrid robots in recent years. Among them, several generations of tissue‐engineered biohybrid walkers have been established as reliable platforms to study untethered locomotion. However, despite these advances, such technology is not mature yet, and major challenges remain. Herein, steps are taken to address two of them: the lack of systematic design approaches, common to biohybrid robotics in general, and in the case of biohybrid walkers specifically, the lack of maneuverability. A dual‐ring biobot is presented which is computationally designed and selected to exhibit robust forward motion and rotational steering. This dual‐ring biobot consists of two independent muscle actuators and a four‐legged scaffold asymmetric in the fore/aft direction. The integration of multiple muscles within its body architecture, combined with differential electrical stimulation, allows the robot to maneuver. The dual‐ring robot design is then fabricated and experimentally tested, confirming computational predictions and turning abilities. Overall, a design approach based on modeling, simulation, and fabrication exemplified in this versatile robot represents a route to efficiently engineer complex biological machines with adaptive functionalities.

Abstract Advancing biologically driven soft robotics and actuators will involve employing different scaffold geometries and cellular constructs to enable a controllable emergence for increased production of force. By using hydrogel scaffolds and muscle tissue, soft biological robotic actuators that are capable of motility have been successfully engineered with varying morphologies. Having the flexibility of altering geometry while ensuring tissue viability can enable advancing functional output from these machines through the implementation of new construction concepts and fabrication approaches. This study reports a forward engineering approach to computationally design the next generation of biological machines via direct numerical simulations. This was subsequently followed by fabrication and characterization of high force producing biological machines. These biological machines show millinewton forces capable of driving locomotion at speeds above 0.5 mm s−1. It is important to note that these results are predicted by computational simulations, ultimately showing excellent agreement of the predictive models and experimental results, further providing the ability to forward design future generations of these biological machines. This study aims to develop the building blocks and modular technologies capable of scaling force and complexity of these devices for applications toward solving real world problems in medicine, environment, and manufacturing.

  3. Soft robotics enriches the robotic functionalities by engineering soft materials and electronics toward enhanced compliance, adaptivity, and friendly human machine. This decade has witnessed extraordinary progresses and benefits in scaling down soft robotics to small scale for a wide range of potential and promising applications, including medical and surgical soft robots, wearable and rehabilitation robots, and unconstructed environments exploration. This perspective highlights recent research efforts in miniature soft robotics in a brief and comprehensive way in terms of actuation, powering, designs, fabrication, control, and applications in four sections. Section 2 discusses the key aspects of materials selection and structural designs for small‐scale tethered and untethered actuation and powering, including fluidic actuation, stimuli‐responsive actuation, and soft living biohybrid materials, as well as structural forms from 1D to 3D. Section 3 discusses the advanced manufacturing techniques at small scales for fabricating miniature soft robots, including lithography, mechanical self‐assembly, additive manufacturing, tissue engineering, and other fabrication methods. Section 4 discusses the control systems used in miniature robots, including off‐board/onboard controls and artificial intelligence‐based controls. Section 5 discusses their potential broad applications in healthcare, small‐scale objects manipulating and processing, and environmental monitoring. Finally, outlooks on the challenges and opportunities are discussed.

  4. Fluid‐driven artificial muscles exhibit a behavior similar to biological muscles which makes them attractive as soft actuators for wearable assistive robots. However, state‐of‐the‐art fluidic systems typically face challenges to meet the multifaceted needs of soft wearable robots. First, soft robots are usually constrained to tethered pressure sources or bulky configurations based on flow control valves for delivery and control of high assistive forces. Second, although some soft robots exhibit untethered operation, they are significantly limited to low force capabilities. Herein, an electrohydraulic actuation system that enables both untethered and high‐force soft wearable robots is presented. This solution is achieved through a twofold design approach. First, a simplified direct‐drive actuation paradigm composed of motor, gear‐pump, and hydraulic artificial muscle (HAM) is proposed, which allows for a compact and lightweight (1.6 kg) valveless design. Second, a fluidic engine composed of a high‐torque motor with a custom‐designed gear pump is created, which is capable of generating high pressure (up to 0.75 MPa) to drive the HAM in delivering high forces (580 N). Experimental results show that the developed fluidic engine significantly outperforms state‐of‐the‐art systems in mechanical efficiency and suggest opportunities for effective deployment in soft wearable robots for human assistance.

  5. Aquatic insects have developed versatile locomotion mechanisms that have served as a source of inspiration for decades in the development of small‐scale swimming robots. However, despite recent advances in the field, efficient, untethered, and integrated powering, actuation, and control of small‐scale robots remains a challenge due to the out‐of‐equilibrium and dissipative nature of the driving physical and chemical phenomena. Here, we have designed small‐scale, bioinspired aquatic locomotors with programmable deterministic trajectories that integrate self‐propelled chemical motors and photoresponsive shape‐morphing structures. A Marangoni motor system is developed integrating structural protein networks that self‐regulate the release of chemical fuel with photochemical liquid crystal network (LCN) actuators that change their shape and deform in and out of the surface of water. While the diffusion of fuel from the motor system regulates the propulsion, the dissipative photochemical deformation of LCNs provides locomotors with control over the directionality of motion at the air‐water interface. This approach gives access to five different but interchangeable modes of locomotion within a single swimming robot via morphing of the soft structure. The proposed design, which mimics the mechanisms of surface gliding and posture change of semiaquatic insects such as water treaders, offers solutions for autonomous swimming soft robots via untethered and orthogonal power and control.
