Title: High Stereocontrol in the Preparation of Silyl-Protected γ-Substituted Enoldiazoacetates
A robust and efficient synthesis of triisopropylsilyl (TIPS)-protected γ-substituted enoldiazoacetates with excellent Z stereocontrol by using lithium bis(trimethylsilyl)azanide (LiHMDS) as a base and TIPSOTf as a silyl transfer reagent is reported. Despite their increased size compared to previously tert-butyldimethylsilyl (TBS)-protected γ-unsubstituted enoldiazoacetates, a high product yield with exceptional stereocontrol has been achieved in copper-catalyzed [3+3] cycloaddition reaction with nitrones by using a chiral indeno bisoxazoline ligand.  more » « less

    Chiral copper( i ) catalysts are preferred over chiral dirhodium( ii ) catalysts for [3 + 3]-cycloaddition reactions of γ-alkyl-substituted enoldiazoacetates compounds with nitrones. Using the In-SaBox ligand these reactions effectively produce cis -3,6-dihydro-1,2-oxazine derivatives under mild conditions in high yield and with exceptional stereocontrol, and enantioselectivity increases with the size of the γ-substituent. Mechanistic studies show that cycloaddition occurs solely through the formation of ( Z )-γ-substituted metallo-enolcarbene intermediates that are catalytically gennerated from both ( Z )- and ( E )-γ-substituted enoldiazoactates via donor–acceptor cyclopropene intermediates.

This review describes catalytic asymmetric cycloaddition reactions of silyl-protected enoldiazo compounds for the construction of highly functionalized carbo- and heterocycles which possess one or more chiral center(s). The enoldiazo compound or its derivative, donor–acceptor cyclopropene, form electrophilic vinylogous metal carbene intermediates that combine stepwise with nucleophilic dipolar reactants to form products from [3 + 1]-, [3 + 2]-, [3 + 3]-, [3 + 4]-, and [3 + 5]-cycloaddition, generally in high yield and with exceptional stereocontrol and regioselectivity.

  3. Abstract We report a general, regioselective, and metal free γ‐fluorination of α,β‐unsaturated carbonyls via silyl dienol ethers that are readily prepared from simple ketones and aldehydes. The transformation displays broad scope including 27 cyclic and acyclic siloxydienes providing γ‐fluoro compounds in 28–91 % yield. Notably, the reported conditions are also suitable for the synthesis of challenging tertiary fluorides. The regioselectivity of the reaction was studied on a series of acyclic siloxydienes and was observed to be sensitive to the conformational flexibility of the substrate. Diversification of the γ‐fluorocarbonyls demonstrates the promise of fluorine as a stereocontrol element.

ABSTRACT This work investigates effects of poly(γ‐butyrolactone) (PγBL) with different initiation and termination chain ends on five types of materials properties, including thermal stability, thermal transitions, thermal recyclability, hydrolytic degradation, and dynamic mechanical behavior. Four different chain‐end‐capped polymers with similar molecular weights, BnO‐[C(=O)(CH2)3O]n‐R, R = C(=O)Me, C(=O)CH=CH2, C(=O)Ph, and SiMe2CMe3, along with a series of uncapped polymers R′O‐[C(=O)(CH2)3O]n‐H (R′ = Bn, Ph2CHCH2) withMnranging from low (4.95 kg mol−1) to high (83.2 kg mol−1), have been synthesized. The termination chain end R showed a large effect on polymer decomposition temperature and hydrolytic degradation, relative to H. Overall, for those properties sensitive to the chain ends, chain‐end capping renders R‐protected linear PγBL behaving much like cyclic PγBL. © 2018 Wiley Periodicals, Inc. J. Polym.   5. Abstract We report an asymmetric synthesis of the (3R,5R)-γ-hydroxypiperazic acid (γ-OHPiz) residue encountered in several bioactive nonribosomal peptides. Our strategy relies on a diastereoselective enolate hydroxylation reaction and electrophilic N-amination to provide the acyclic γ-OHPiz precursor. This orthogonally protected α-hydrazino acid intermediate is amenable to late-stage diazinane ring formation following incorporation into a peptide chain. We determined the N-terminal amide rotamer propensity of the γ-OHPiz residue and showed that the γ-OH substituent enhances trans-amide bias relative to piperazic acid.
