    ABSTRACT Quorum sensing (QS) is a chemical communication process in which bacteria produce, release, and detect extracellular signaling molecules called autoinducers. Via combined transcriptional and posttranscriptional regulatory mechanisms, QS allows bacteria to collectively alter gene expression on a population-wide scale. Recently, the TetR family transcriptional regulator LuxT was shown to control Vibrio harveyi qrr 1, encoding the Qrr1 small RNA that functions at the core of the QS regulatory cascade. Here, we use RNA sequencing to reveal that, beyond the control of qrr 1, LuxT is a global regulator of 414 V. harveyi genes, including those involved in type III secretion, siderophore production, and aerolysin toxin biosynthesis. Importantly, LuxT directly represses swrZ , encoding a GntR family transcriptional regulator, and LuxT control of type III secretion, siderophore, and aerolysin genes occurs by two mechanisms, one that is SwrZ dependent and one that is SwrZ independent. All of these target genes specify QS-controlled behaviors that are enacted when V. harveyi is at low cell density. Thus, LuxT and SwrZ function in parallel with QS to drive particular low-cell-density behaviors. Phylogenetic analyses reveal that luxT is highly conserved among Vibrionaceae , but swrZ is less well conserved. In a test case, we find that in Aliivibrio fischeri , LuxT also represses swrZ . SwrZ is a repressor of A. fischeri siderophore production genes. Thus, LuxT repression of swrZ drives the activation of A. fischeri siderophore gene expression. Our results indicate that LuxT is a major regulator among Vibrionaceae , and in the species that also possess swrZ , LuxT functions with SwrZ to control gene expression. IMPORTANCE Bacteria precisely tune gene expression patterns to successfully react to changes that occur in the environment. Defining the mechanisms that enable bacteria to thrive in diverse and fluctuating habitats, including in host organisms, is crucial for a deep understanding of the microbial world and also for the development of effective applications to promote or combat particular bacteria. In this study, we show that a regulator called LuxT controls over 400 genes in the marine bacterium Vibrio harveyi and that LuxT is highly conserved among Vibrionaceae species, ubiquitous marine bacteria that often cause disease. We characterize the mechanisms by which LuxT controls genes involved in virulence and nutrient acquisition. We show that LuxT functions in parallel with a set of regulators of the bacterial cell-to-cell communication process called quorum sensing to promote V. harveyi behaviors at low cell density.

  2. Abstract. This paper provides an overview and demonstration of emerging float-based methods for quantifying gross primary production (GPP) and net community production (NCP) using Biogeochemical-Argo (BGC-Argo) float data. Recent publications have described GPP methods that are based on the detection of diurnal oscillations in upper-ocean oxygen or particulate organic carbon concentrations using single profilers or a composite of BGC-Argo floats. NCP methods rely on budget calculations to partition observed tracer variations into physical or biological processes occurring over timescales greater than 1 d. Presently, multi-year NCP time series are feasible at near-weekly resolution, using consecutive or simultaneous float deployments at local scales. Results, however, are sensitive to the choice of tracer used in the budget calculations and uncertainties in the budget parameterizations employed across different NCP approaches. Decadal, basin-wide GPP calculations are currently achievable using data compiled from the entire BGC-Argo array, but finer spatial and temporal resolution requires more float deployments to construct diurnal tracer curves. A projected, global BGC-Argo array of 1000 floats should be sufficient to attain annual GPP estimates at 10∘ latitudinal resolution if floats profile at off-integer intervals (e.g., 5.2 or 10.2 d). Addressing the current limitations of float-based methods should enable enhanced spatial and temporal coverage of marine GPP and NCP measurements, facilitating global-scale determinations of the carbon export potential, training of satellite primary production algorithms, and evaluations of biogeochemical numerical models. This paper aims to facilitate broader uptake of float GPP and NCP methods, as singular or combined tools, by the oceanographic community and to promote their continued development.

Abstract Most research showing that cognates are named faster than non-cognates has focused on isolated word production which might not realistically reflect cognitive demands in sentence production. Here, we explored whether cognates elicit interference by examining error rates during sentence production, and how this interference is resolved by language control mechanisms. Twenty highly proficient Spanish–English bilinguals described visual scenes with sentence structures ‘NP1-verb-NP2’ (NP = noun-phrase). Half the nouns and half the verbs were cognates and two manipulations created high control demands. Both situations that demanded higher inhibitory control pushed the cognate effect from facilitation towards interference. These findings suggest that cognates, similar to phonologically similar words within a language, can induce not only facilitation but robust interference.

This data package provides net community production (NCP) and gross oxygen production (GOP, a measure of gross primary production) for the winter, spring, and summer Northeast U.S. Shelf Long-Term Ecological Research (NES-LTER) Transect cruises in 2019. Two tables are provided: a high-frequency table with NCP rates calculated from measurements of O2/Ar made continuously by an at-sea equilibrator inlet mass spectrometer (EIMS), and a low-frequency table with both NCP and GOP rates calculated for discrete samples measured post-cruise. The GOP rates were calculated from triple O2 isotopic (TOI) ratios. These data are derived from the EIMS and TOI data for the NES-LTER Transect cruises in EDI data package knb-lter-nes.6.2.
