Abstract The Diaprepes root weevil (DRW), Diaprepes abbreviatus, is a broadly polyphagous invasive pest of agriculture in the southern United States and the Caribbean. Its genome was sequenced, assembled, and annotated to study genomic correlates of specialized plant-feeding and invasiveness and to facilitate the development of new methods for DRW control. The 1.69 Gb D. abbreviatus genome assembly was distributed across 653 contigs, with an N50 of 7.8 Mb and the largest contig of 62 Mb. Most of the genome was comprised of repetitive sequences, with 66.17% in transposable elements, 5.75% in macrosatellites, and 2.06% in microsatellites. Most expected orthologous genes were present and fully assembled, with 99.5% of BUSCO genes present and 1.5% duplicated. One hundred and nine contigs (27.19 Mb) were identified as putative fragments of the X and Y sex chromosomes, and homology assessment with other beetle X chromosomes indicated a possible sex chromosome turnover event. Genome annotation identified 18,412 genes, including 43 putative horizontally transferred (HT) loci. Notably, 258 genes were identified from gene families known to encode plant cell wall degrading enzymes and invertases, including carbohydrate esterases, polysaccharide lyases, and glycoside hydrolases (GH). GH genes were unusually numerous, with 239 putative genes representing 19 GH families. Interestingly, several other beetle species with large numbers of GH genes are (like D. abbreviatus) successful invasive pests of agriculture or forestry.

The complete genome sequences of three species of Mountain Avens (Dryas, Rosaceae)
We present the whole genome sequences of Dryas alaskensis, D. ajanensis, and D. integrifolia from plants collected from interior Alaska. We performed deep Illumina sequencing of a single leaf of each voucher. The sequence reads were then de novo assembled and conserved regions across all preassemblies were used to join contigs in a finishing step. The raw and assembled data is publicly available via Genbank.

An ionic metal–organic-framework (MOF) containing nanoscale channels was readily assembled from ditopic 4′-pyridyl-2,2′:6′,2′′-terpyridine (pytpy) and a simple iron( ii ) salt. X-ray structural analysis revealed a two-dimensional grid-like framework assembled by classic octahedral (pytpy) 2 Fe II cations as linkers (with pytpy as a new ditopic pyridyl ligand) and octa-coordinate FeCl 2 centers as nodes. The layer-by-layer assembly of the 2-D framework resulted in the formation of 3-D porous materials consisting of nano-scale channels. The charges of the cationic framework were balanced with anionic Cl 3 FeOFeCl 3 in its void channels. The new Fe-based MOF material was employed as a precatalyst for syn -selective hydroboration of alkynes under mild, solvent-free conditions in the presence of an activator, leading to the synthesis of a range of trans -alkenylboronates in good yields. The larger scale applicability and recyclability of the new MOF catalyst was further explored. This represents a rare example of an ionic MOF material that can be utilized in hydroboration catalysis.more » « less

Sachs, M (Ed.)Abstract Aegilops tauschii is the donor of the D subgenome of hexaploid wheat and a valuable genetic resource for wheat improvement. Several reference-quality genome sequences have been reported for A. tauschii accession AL8/78. A new genome sequence assembly (Aet v6.0) built from long Pacific Biosciences HiFi reads and employing an optical genome map constructed with a new technology is reported here for this accession. The N50 contig length of 31.81 Mb greatly exceeded that of the previous AL8/78 genome sequence assembly (Aet v5.0). Of 1,254 super-scaffolds, 92, comprising 98% of the total super-scaffold length, were anchored on a high-resolution genetic map, and pseudomolecules were assembled. The number of gaps in the pseudomolecules was reduced from 52,910 in Aet v5.0 to 351 in Aet v6.0. Gene models were transferred from the Aet v5.0 assembly into the Aet v6.0 assembly. A total of 40,447 putative orthologous gene pairs were identified between the Aet v6.0 and Chinese Spring wheat IWGSC RefSer v2.1 D-subgenome pseudomolecules. Orthologous gene pairs were used to compare the structure of the A. tauschii and wheat D-subgenome pseudomolecules. A total of 223 structural differences were identified. They included 44 large differences in sequence orientation and 25 differences in sequence location. A technique for discriminating between assembly errors and real structural variation between closely related genomes is suggested.more » « less

Chiral nanostructures have been attracting extensive interest in recent years primarily because of the unique materials properties that can be exploited for diverse applications. In this study, gold Janus nanoparticles, with hexanethiolates and 3-mercapto-1,2-propanediol segregated on the two hemispheres of the metal cores (dia. 2.7 ± 0.4 nm), self-assembled into vesicle-like, hollow nanostructures in both water and organic media, and exhibited apparent plasmonic circular dichroism (PCD) absorption in the visible range. This was in contrast to individual Janus nanoparticles, bulk-exchange nanoparticles where the two ligands were homogeneously mixed on the nanoparticle surface, or nanoparticles capped with only one kind of ligand. The PCD signals were found to become intensified with increasing coverage of the 3-mercapto-1,2-propanediol ligands on the nanoparticle surface. This was accounted for by the dipolar property of the structurally asymmetrical Janus nanoparticles, and theoretical simulations based on first principles calculations showed that when the nanoparticle dipoles self-assembled onto the surface of a hollow sphere, a vertex was formed which gave rise to the unique chiral characteristics. The resulting chiral nanoparticle vesicles could be exploited for the separation of optical enantiomers, as manifested in the selective identification and separation of d -alanine from the l -isomer.more » « less
