Title: Trails to Research: an Inquiry-Based Course Using Zebrafish To Provide Research Experience to Tribal College Students
ABSTRACT Embryonic development is fascinating to follow and highly engaging and, therefore, lends itself for undergraduate students’ first steps in experimental science. We developed the “Trails to Research” inquiry-based course, which exposes students to life science research using zebrafish as model organism. Zebrafish are ideal in the classroom: they are easy to maintain, their embryos develop rapidly, and they are easily manipulated. Further, they lend themselves to teach about embryo development and experimental design. We developed the course for undergraduates at 2-year colleges and, therefore, for students with little or no research experience. In this 5-day intensive course (which is taught during summers as a stand-alone course), students design treatment experiments for zebrafish embryos with known teratogens and with substances they select. The course comprises three modules that overlap over the 5 days: (i) introduction to developmental biology, model organisms, toxicology, and experimental design, (ii) zebrafish embryo experimental setup, and (iii) collecting, analyzing, and presenting data. Student learning was significant in the areas of experimental design, working with model systems, working with zebrafish embryos, using laboratory equipment, and presenting the results of their experiments using effective methods.  more » « less

  1. Open-ended laboratory projects increase student success and retention in the sciences. However, developing organismal-based research projects is a challenge for students with restricted laboratory access, such as those attending courses remotely. Here I describe the use of image analysis of zebrafish neural development for authentic research projects in an introductory biology laboratory course. Zebrafish are a vertebrate model that produce large numbers of externally and rapidly developing embryos. Because zebrafish larvae are transparent, fluorescent reporters marking nervous system structures can be imaged over time and analyzed by undergraduate scientists. In the pilot of this project, remote first-year college students independently developed biological questions based on an image collection comparing zebrafish mutants and wild-type siblings. Students created and mastered techniques to analyze position, organization, and other morphological features of developing neurons and glia in the images to directly test their biological questions. At the end of the course, students communicated their project results in journal article format and oral presentations. Students were able to hone skills in organismal observation and data collection while studying remotely, and they reported excitement at applying lecture-based knowledge to their own independent questions. This module can be adapted by other instructors for both students on- and off-campus to teach principles of neural development, data collection, data analysis, and scientific communication.

  2. Undergraduate instructional biology laboratories are typically taught within two paradigms. Some labs focus on protocols and techniques delivered in “cookbook” format with defined experimental outcomes. There is increasing momentum to alternatively employ student-driven, open-ended, and discovery-based strategies, oftenviacourse-based undergraduate research experiences (CUREs) using crowd-sourcing initiatives. A fraction of students also participate in funded research in faculty research labs, where they have opportunities to work on projects designed to expand the frontiers of human knowledge. These experiences are widely recognized as valuable but are not scalable, as most institutions have many more undergraduates than research lab positions. We sought to address this gap through our department’s curriculum by creating an opportunity for students to participate in the real-world research process within a laboratory course. We conceived, developed, and delivered an authentic, guided research experience to students in an upper-level molecular biology laboratory course. We refer to this model as a “research program-linked CURE.” The research questions come directly from a faculty member’s research lab and evolve along with that research program. Students study post-transcriptional regulation in mycobacteria. We use current molecular biology methodologies to test hypotheses like “UTRs affect RNA and protein expression levels,” “there is functional redundancy among RNA helicases,” and “carbon starvation alters mRNA 5′ end chemistries.” We conducted standard assessments and developed a customized “Skills and Concepts Inventory” survey to gauge how well the course met our student learning outcomes. We report the results of our assessments and describe challenges addressed during development and execution of the course, including organizing activities to fit within an instructional lab, balancing breadth with depth, and maintaining authenticity while giving students the experience of obtaining interpretable and novel results. Our data suggest student learning was enhanced through this truly authentic research approach. Further, students were able to perceive they were participants and contributors within an active research paradigm. Students reported increases in their self-identification as scientists, and a positive impact on their career trajectories. An additional benefit was reciprocation back to the funded research laboratory, by funneling course alumni, results, materials, and protocols.

  3. Course-based undergraduate research experiences (CUREs) can have many positive effects on students’ learning and sense of self-efficacy. We have developed a networked CURE between four different institutions designed for courses in analytical chemistry that focuses on the process of adapting published solution-phase colorimetric assays into microfluidic paper analytical devices (μPAD) assays. We used a backward design process to develop 5 scaffolded learning outcomes: (1) identify and assess relevant literature sources, (2) propose a viable experimental plan to answer a well-defined scientific question based on literature information and experimental results, (3) apply appropriate methods of data analysis to interpret experimental results, (4) evaluate multiple pieces of experimental data to support conclusions, and (5) contribute to a team by working collaboratively toward common goals. Students begin the project by completing a literature search assignment to identify a published colorimetric assay they plan to adapt. They then write a proposal which identifies their analyte, sample of interest, and the figures of merit required for successful sample analysis using their μPAD. During the 3–5 weeks of laboratory work, students conduct their experiments, and each week evaluate the significance of their data and propose an experimental plan for the upcoming week. At the end of the μCURE project, students present their results in a joint, asynchronous poster session. Student artifacts are assessed for evidence of particular skills using rubrics from the Enhancing Learning by Improving Process Skills in STEM (ELIPSS) Project. Scores on the rubrics indicate partial to full attainment of each of the five learning outcomes

  4. The Project-Based Scientific Research is a new interdisciplinary course developed by the National Science Foundation (NSF - IUSE) funded STEM center at _______ State University. The implementation of this new course was one of the major three goals for this five year grant to strengthen the STEM undergraduate research community at ______ State University by helping undergraduates who are interested in hands-on and/or scientific research. The course is designed to introduce undergraduate junior and senior science, engineering technology and math students to the vibrant world of real research; to build foundational skills for research; to help STEM students meet potential mentors whose research labs they might join with the goal of gaining experimental research experience while on campus. On top of course content and requirements the following goals are aimed for the student and faculty mentors to strengthen the research community; (1) helping undergraduate students who are interested in research connect with faculty partners who are committed to mentoring undergraduates in research, (2) to guide students in reading through papers that introduce the type of research being carried out in a faculty partners lab, (3) to guide students in drafting a mini-review of 5 papers relevant to that research, (4) to guide students in identifying and writing up a research proposal which they will complete in the lab of the faculty partner. The learning objectives for the students in this course are summarized as; (a) by the end of this course, all students build a foundational understanding of the principles of STEM research through the exploration and discussion of important historical interdisciplinary projects; (b) interact with faculty researchers who perform projects across STEM disciplines; (c) be able to describe the similarities and differences between experimental and theoretical STEM research; (d) explore and present several possibilities for future research topics; (e) design and present a research prospectus, complete with a review of some of the relevant literature; (f) and be prepared to continue a research project with a chosen faculty mentor or mentors. First year, six academic departments out of eight participated this new course by offering a cross-listed course for their students under one major course taught by one of the PIs at the STEM Center. All the details such as challenges faced, outcomes, resources used, faculty involved, student and faculty feedback etc. for this course will be shared with academia in the paper.

  5. The Association of American Colleges and Universities identifies undergraduate research experiences as a high impact practice for increasing student success and retention in STEM majors. Most undergraduate research opportunities for community college engineering students involve partnerships with universities and typically take the form of paid summer experiences. Course-based Undergraduate Research Experiences (CUREs) offer an alternative model with potential for significant expansion of research opportunities for students. This approach weaves research into the courses students are already required to complete for their degrees. CUREs are an equitable approach for introducing students to research because they do not demand extracurricular financial and/or time commitments beyond what students must already commit to for their courses. This paper describes an adaptable model for implementing a CURE in an introductory engineering design and computing course that features applications of low-cost microcontrollers. Students work toward course learning outcomes focused on computer programming, engineering design processes, and effective teamwork in the context of multi-term research and development efforts to design, build, and test devices for other CUREs in science lab courses as well as for other applications at the college or with community partners. Students choose from a menu of projects each term, with a typical course offering involving four to six different projects running simultaneously. Each team identifies a focused design and development scope of work within the larger context of the project they are interested in. They give weekly progress reports and gather input from their customers. The work culminates in a prototype and final report to document their work for student teams who will carry it forward in future terms. We assessed the impact of the experience on students’ beliefs about science and engineering, STEM confidence, and career aspirations using a nationally normed survey for CUREs in STEM and report results from five terms of offering this course. We find statistically significant pre-post gains on two-thirds of the survey items relating to students’ understanding of the research process and confidence in their STEM abilities. The pre-post gains are generally comparable to those reported by others who used the same survey to assess the impact of a summer research experience for community college students. These findings indicate that the benefits of student participation in this CURE model are comparable to the benefits students see by participation in summer research programs.
