ABSTRACT Organisms may simultaneously face thermal, desiccation and nutritional stress under climate change. Understanding the effects arising from the interactions among these stressors is relevant for predicting organisms' responses to climate change and for developing effective conservation strategies. Using both dynamic and static protocols, we assessed for the first time how sublethal desiccation exposure (at 16.7%, 50.0% and 83.3% of LD50) impacts the heat tolerance of foragers from two social bee species found on the Greek island of Lesbos: the managed European honey bee, Apis mellifera, and the wild, ground-nesting sweat bee Lasioglossum malachurum. In addition, we explored how a short-term starvation period (24 h), followed by a moderate sublethal desiccation exposure (50% of LD50), influences honey bee heat tolerance. We found that neither the critical thermal maximum (CTmax) nor the time to heat stupor was significantly impacted by sublethal desiccation exposure in either species. Similarly, starvation followed by moderate sublethal desiccation did not affect the average CTmax estimate, but it did increase its variance. Our results suggest that sublethal exposure to these environmental stressors may not always lead to significant changes in bees' heat tolerance or increase vulnerability to rapid temperature changes during extreme weather events, such as heat waves. However, the increase in CTmax variance suggests greater variability in individual responses to temperature stress under climate change, which may impact colony-level performance. The ability to withstand desiccation may be impacted by unmeasured hypoxic conditions and the overall effect of these stressors on solitary species remains to be assessed.

            The European honey bee, Apis mellifera L., is the single most valuable managed pollinator in the world. Poor colony health or unusually high colony losses of managed honey bees result from a myriad of stressors, which are more harmful in combination. Climate change is expected to accentuate the effects of these stressors, but the physiological and behavioral responses of honey bees to elevated temperatures while under simultaneous influence of one or more stressors remain largely unknown. Here we test the hypothesis that exposure to acute, sublethal doses of neonicotinoid insecticides reduce thermal tolerance in honey bees. We administered to bees oral doses of imidacloprid and acetamiprid at 1/5, 1/20, and1/100 of LD50 and measured their heat tolerance 4 h post- feeding, using both dynamic and static protocols. Contrary to our expectations, acute exposure to sublethal doses of both insecticides resulted in higher thermal tolerance and greater survival rates of bees. Bees that ingested the higher doses of insecticides displayed a critical thermal maximum from 2˚C to 5 ˚C greater than that of the control group, and 67%–87% reduction in mortality. Our study suggests a resilience of honey bees to high temperatures when other stressors are present, which is consistent with studies in other insects. We discuss the implications of these results and hypothesize that this compensatory effect is likely due to induction of heat shock proteins by the insecticides, which provides temporary protection from elevated temperatures

            Honey bees (Apis mellifera) are an agriculturally important pollinator species that live in easily managed social groups (i.e., colonies). Unfortunately, annual losses of honey bee colonies in many parts of the world have reached unsustainable levels. Multiple abiotic and biotic stressors, including viruses, are associated with individual honey bee and colony mortality. Honey bees have evolved several antiviral defense mechanisms including conserved immune pathways (e.g., Toll, Imd, JAK/STAT) and dsRNA-triggered responses including RNA interference and a non-sequence specific dsRNA-mediated response. In addition, transcriptome analyses of virus-infected honey bees implicate an antiviral role of stress response pathways, including the heat shock response. Herein, we demonstrate that the heat shock response is antiviral in honey bees. Specifically, heat-shocked honey bees (i.e., 42 °C for 4 h) had reduced levels of the model virus, Sindbis-GFP, compared with bees maintained at a constant temperature. Virus-infection and/or heat shock resulted in differential expression of six heat shock protein encoding genes and three immune genes, many of which are positively correlated. The heat shock protein encoding and immune gene transcriptional responses observed in virus-infected bees were not completely recapitulated by administration of double stranded RNA (dsRNA), a virus-associated molecular pattern, indicating that additional virus–host interactions are involved in triggering antiviral stress response pathways.more » « less

            Across diverse taxa, sublethal exposure to abiotic stressors early in life can lead to benefits such as increased stress tolerance upon repeat exposure. This phenomenon, known as hormetic priming, is largely unexplored in early life stages of marine invertebrates, which are increasingly threatened by anthropogenic climate change. To investigate this phenomenon, larvae of the sea anemone and model marine invertebrateNematostella vectensiswere exposed to control (18 °C) or elevated (24 °C, 30 °C, 35 °C, or 39 °C) temperatures for 1 h at 3 days post-fertilization (DPF), followed by return to control temperatures (18 °C). The animals were then assessed for growth, development, metabolic rates, and heat tolerance at 4, 7, and 11 DPF. Priming at intermediately elevated temperatures (24 °C, 30 °C, or 35 °C) augmented growth and development compared to controls or priming at 39 °C. Indeed, priming at 39 °C hampered developmental progression, with around 40% of larvae still in the planula stage at 11 DPF, in contrast to 0% for all other groups. Total protein content, a proxy for biomass, and respiration rates were not significantly affected by priming, suggesting metabolic resilience. Heat tolerance was quantified with acute heat stress exposures, and was significantly higher for animals primed at intermediate temperatures (24 °C, 30 °C, or 35 °C) compared to controls or those primed at 39 °C at all time points. To investigate a possible molecular mechanism for the observed changes in heat tolerance, the expression of heat shock protein 70 (HSP70) was quantified at 11 DPF. Expression of HSP70 significantly increased with increasing priming temperature, with the presence of a doublet band for larvae primed at 39 °C, suggesting persistent negative effects of priming on protein homeostasis. Interestingly, primed larvae in a second cohort cultured to 6 weeks post-fertilization continued to display hormetic growth responses, whereas benefits for heat tolerance were lost; in contrast, negative effects of short-term exposure to extreme heat stress (39 °C) persisted. These results demonstrate that some dose-dependent effects of priming waned over time while others persisted, resulting in heterogeneity in organismal performance across ontogeny following priming. Overall, these findings suggest that heat priming may augment the climate resilience of marine invertebrate early life stagesviathe modulation of key developmental and physiological phenotypes, while also affirming the need to limit further anthropogenic ocean warming.more » « less

            Anthropogenic climate change is projected to affect marine ecosystems by challenging the environmental tolerance of individuals. Marine fishes may be particularly vulnerable to emergent climate stressors during early life stages. Here we focus on embryos of Pacific herring(Clupea pallasii), an important forage fish species widely distributed across the North Pacific. Embryos were reared under a range of temperatures (10-16°C) crossed with twopCO2levels (600 and 2000μatm) to investigate effects on metabolism and survival. We further tested how elevatedpCO2affects critical thermal tolerance (CTmax) by challenging embryos to short-term temperature fluctuations. Experiments were repeated on embryos collected from winter and spring spawning populations to determine if spawning phenology corresponds with different limits of environmental tolerance in offspring. We found that embryos could withstand acute exposure to 20°C regardless of spawning population or incubation treatment, but that survival was greatly reduced after 2-3 hours at 25°C. We found thatpCO2had limited effects onCTmax. The survival of embryos reared under chronically warm conditions (12°, 14°, or 16°C) was significantly lower relative to 10°C treatments in both populations. Oxygen consumption rates (MO2) were also higher at elevated temperatures andpCO2levels. However, heart contraction measurements made 48 hours afterCTmaxexposure revealed a greater increase in heart rate in embryos reared at 10°C compared to 16°C, suggesting acclimation at higher incubation temperatures. Our results indicate that Pacific herring are generally tolerant ofpCO2but are vulnerable to acute temperature stress. Importantly, spring-spawning embryos did not clearly exhibit a higher tolerance to heat stress compared to winter offspring.more » « less

            Climate change poses a threat to organisms across the world, with cold-adapted species such as bumble bees (Bombus spp.) at particularly high risk. Understanding how organisms respond to extreme heat events associated with climate change as well as the factors that increase resilience or prime organisms for future stress can inform conservation actions. We investigated the effects of heat stress within different contexts (duration, periodicity, with and without access to food, and in the laboratory versus field) on bumble bee (Bombus impatiens) survival and heat tolerance. We found that both prolonged (5 h) heat stress and nutrition limitation were negatively correlated with worker bee survival and thermal tolerance. However, the effects of these acute stressors were not long lasting (no difference in thermal tolerance among treatment groups after 24 h). Additionally, intermittent heat stress, which more closely simulates the forager behavior of leaving and returning to the nest, was not negatively correlated with worker thermal tolerance. Thus, short respites may allow foragers to recover from thermal stress. Moreover, these results suggest there is no priming effect resulting from short- or long-duration exposure to heat – bees remained equally sensitive to heat in subsequent exposures. In field-caught bumble bees, foragers collected during warmer versus cooler conditions exhibited similar thermal tolerance after being allowed to recover in the lab for 16 h. These studies offer insight into the impacts of a key bumble bee stressor and highlight the importance of recovery duration, stressor periodicity and context on bumble bee thermal tolerance outcomes.more » « less
