Abstract Dormant propagules can provide a rapid colonization source for temporary aquatic habitats and set the trajectory for community dynamics, yet the egg banks of stormwater management systems have received little attention. We asked which species hatched from the sediment of drainage ditches in Champaign County, IL, and found bdelloid rotifers and ostracods (Heterocypris incongruens) to be the most common taxa. These sites also are colonized by mosquitoes, and we established laboratory experiments to examine interspecific interactions between common co‐occurring taxa.Culex restuanslarvae were reared in the presence or absence ofH. incongruensat two intra‐ and interspecific densities (20 or 40 total individuals) and their survivorship to adulthood, development time to adulthood, adult body size, and sex ratio were determined. Survival forCx. restuanswas significantly lower at high larval density than at low larval density in both treatments.Culex restuanslarvae reared in the presence ofH. incongruenshad a shorter development time to adulthood and emerged as larger adults compared to those reared in the absence ofH. incongruens. The sex ratios in theH. incongruenstreatments were female‐biased whereas those in theCulex‐only treatments were male‐biased. These differences may have epidemiological implications, as only female mosquitoes serve as disease vectors. Our results emphasize the importance of understanding interspecific interactions in influencing larval mosquito development traits.

Rearing and Maintaining a Culex Colony in the Laboratory
After overcoming the significant obstacle of getting adultCulexmosquitoes to reproduce and blood feed in the laboratory, maintaining a laboratory colony is much more achievable. However, great care and attention to detail are still required to ensure that the larvae have adequate food without being overwhelmed by bacterial growth. Additionally, achieving the appropriate densities of larvae and pupae is essential, as overcrowding delays development, prevents pupae from successfully emerging as adults, and/or reduces adult fecundity and alters sex ratios. Finally, adult mosquitoes should have constant access to H2O and nearly constant access to sugar sources to ensure that both males and females have adequate nutrition and can produce the maximum number of offspring. Here, we describe our methods for maintaining the Buckeye strain ofCulex pipiensand how other researchers might modify them to suit their specific needs.

Culexlarvae are well adapted to growing and developing in containers, and therefore collecting and rearing field-collectedCulexto adulthood in the laboratory is relatively straightforward. What is substantially more challenging is simulating natural conditions that encourageCulexadults to mate, blood feed, and reproduce in laboratory settings. In our experience, this is the most difficult hurdle to overcome when establishing new laboratory colonies. Here, we detail how to collectCulexeggs from the field and establish a colony in the laboratory. Culexmosquitoes transmit several pathogens to humans and animals, including viruses that cause West Nile fever and St. Louis encephalitis and filarial nematodes that cause canine heartworm and elephantiasis. Additionally, these mosquitoes have a cosmopolitan distribution and provide interesting models for understanding population genetics, overwintering dormancy, disease transmission, and other important and ecological questions. However, unlikeAedesmosquitoes that produce eggs that can be stored for weeks at a time, no obvious “stopping” point exists in the development ofCulexmosquitoes. Therefore, these mosquitoes require nearly continuous care and attention. Here, we describe some general considerations when rearing laboratory colonies ofCulexmosquitoes. We highlight different methods so that readers may choose what works best for their experimental needs and laboratory infrastructure. We hope that this information will enable additional scientists to conduct laboratory research on these important disease vectors.more » « less

Abstract Understanding the development process of male and female mosquitoes provides important basic information for sterile insect release programmes and is important for improving other vector control strategies. However, little is known about the molecular mechanisms that distinguish male from female‐specific developmental processes in this species. We used IlluminaRNA‐seq to identify sex‐specific genes during pupal and adult stages. One hundred and forty‐seven genes were expressed only in pupal males, 56 genes were expressed in adult males and another 82 genes were commonly expressed in both male samples. In addition, 26 genes were expressed only in the pupal females, 163 genes were found in the adult females and only one gene was expressed in both female samples. A further quantitative real‐time PCR validation of selected genes from the RNA sequencing (RNA‐seq) analysis confirmed upregulation of those genes in a sex‐specific manner, including: fibrinogen and fibronectin, a zinc finger protein, phospholipase A(2) and a serine protein for female pupae; venom allergen 3, a perlecan, testis‐specific serine/threonine‐protein kinase 1, testis‐specific serine/threonine‐protein kinase 6 and cytochrome c‐2 for male pupae; a salivary protein, D7 protein precursor, trypsin 7 precursor, D7 protein and nanos for female adults; and tetraspanin F139, cytosol aminopeptidase, testis‐specific serine/threonine‐protein kinase 1, a testis‐specific serine/threonine‐protein kinase 6 and a C‐type lectin for male adults. These findings provide insight into the development and physiology ofCulexmosquitoes, which will help in the development of more effective control methods for these disease vectors.more » « less

Abstract In parasitoid systems, resource competition can significantly impact developmental outcomes. This study investigates how larval competition and host characteristics influence development in the acoustic parasitoid fly Ormia ochracea, using the house cricket Acheta domesticus as a host. We experimentally manipulated larval load (1 vs. 2 larvae per host) and recorded host sex and size to assess their effects on pupation and eclosion (adult hatching) success, as well as pupal and adult fly size. While double infestations increased total yield (0.78 vs. 0.54 flies per host), larvae developing without competition exhibited higher relative pupation and eclosion success and produced larger pupae and adult flies, indicating greater individual fitness. Although female host crickets yielded larger pupae, resource competition was the dominant factor shaping developmental outcomes. These results highlight the trade-offs between reproductive yield and offspring fitness driven by resource competition and validate the commercially available A. domesticus as a viable host.more » « less
