Title: A family of olfactory receptors uniquely expanded in marsupial and monotreme genomes are expressed by a T cell subset also unique to marsupials and monotremes
Olfactory receptors (OR), expressed on olfactory neurons, mediate the sense of smell. Recently, OR have also been shown to be expressed in non-olfactory tissues, including cells of the immune system. An analysis of single-cell transcriptomes of splenocytes of the gray short-tailed opossum (Monodelphis domestica) found OR are expressed on a subset of T cells, the γμ T cells, that are unique to marsupials and monotremes. A majority of opossum γμ T cells transcriptomes contain OR family 14 transcripts, specifically, from the OR14C subfamily. Amongst the mammals, the OR14 gene family is expanded in the genomes of marsupials and monotremes, and rarer or absent in placental mammals. In summary, here we demonstrate the intriguing correlation that a family of OR genes, abundant in the genomes of marsupials and monotremes, are ectopically expressed in a particular subset of T cells unique to the marsupials and monotremes.  more » « less

  1. T cells are a primary component of the vertebrate adaptive immune system. There are three mammalian T cell lineages based on their T cell receptors (TCR). The αβ T cells and γδ T cells are ancient and found broadly in vertebrates. The more recently discovered γμ T cells are uniquely mammalian and only found in marsupials and monotremes. In this study, we compare the TCRμ locus (TRM) across the genomes of two marsupials, the grey short-tailed opossum and Tasmanian devil, and one monotreme, the platypus. These analyses revealed lineage specific duplications, common to all non-eutherian mammals described. There is conserved synteny in the TRM loci of both marsupials but not in the monotreme. Our results are consistent with an ancestral cluster organization which was present in the last common mammalian ancestor which underwent lineage specific duplications and divergence among the non-eutherian mammals.

  2. The individual bones of the adult cranium of the gray short-tailed opossum, Monodelphis domestica (Wagner, 1842) are described and illustrated in multiple views based on CT scans. The author previously reported on the outer bony surfaces of the skull of Monodelphis Burnett, 1830, and the current contribution is a companion piece, paying particular attention to the inner bony surfaces (within the endocranium and nasal cavity) and the facets between individual cranial elements, including the ethmo- and frontoturbinals. Comments are provided on the internal nasal floor skeleton, which in M. domestica includes a fused conglomerate formed by the medial palatine processes of the premaxillae, the vomer, the ethmoid, the presphenoid, and the orbitosphenoids. This conglomerate includes horizontal shelves just dorsal to the hard palate, and occurs widely in marsupials but is currently unknown in monotremes and placentals.

    Among extant vertebrates, mammals are distinguished by having a chain of three auditory ossicles (the malleus, incus and stapes) that transduce sound waves and promote an increased range of audible—especially high—frequencies. By contrast, the homologous bones in early fossil mammals and relatives also functioned in chewing through their bony attachments to the lower jaw. Recent discoveries of well-preserved Mesozoic mammals have provided glimpses into the transition from the dual (masticatory and auditory) to the single auditory function for the ossicles, which is now widely accepted to have occurred at least three times in mammal evolution. Here we report a skull and postcranium that we refer to the haramiyidan Vilevolodon diplomylos (dating to the Middle Jurassic epoch (160 million years ago)) and that shows excellent preservation of the malleus, incus and ectotympanic (which supports the tympanic membrane). After comparing this fossil with other Mesozoic and extant mammals, we propose that the overlapping incudomallear articulation found in this and other Mesozoic fossils, in extant monotremes and in early ontogeny in extant marsupials and placentals is a morphology that evolved in several groups of mammals in the transition from the dual to the single function for the ossicles.

    A small piece of cartilage or bone, the element of Paaw, occurs in the tendon of the stapedius muscle in some extant marsupial and placental mammals. It has been nearly a century since the last comprehensive treatment of the distribution of the element of Paaw in mammals. The current report updates knowledge on this structure by synthesizing the subsequent literature and providing new observations of extant marsupials from the collections of the Section of Mammals, Carnegie Museum of Natural History, and two online resources for CT scanned data: DigiMorph.org and MorphoSource.org. We found an element of Paaw in some representatives of all seven extant marsupial orders: Didelphimorphia, Microbiotheria, Notoryctemorphia, Peramelemorphia, Paucituberculata, Dasyuromorphia, and Diprotodontia. In the first four orders, the element is substantial, longer than the long axis of the fenestra vestiuli (oval window), which holds the stapedial footplate; it is smaller than the long axis of the fenestra vestibuli in Paucituberculata and we do not have measures to report for the last two orders. In most marsupials examined, the element of Paaw contacts the petrosal behind the oval window, suggesting it functions as a sesamoid bone, increasing the lever arm of the stapedius muscle. Although there is some variability in the presence of the bone both between and within individual museum specimens, we interpret this as the result of preparation techniques rather than true variation. To place the element of Paaw in its anatomical context, we describe in detail the ear region and middle-ear auditory apparatus of the gray four-eyed opossum, Philander opossum (Linnaeus, 1758), a didelphid from Central and South America, based on a CT scanned specimen from Carnegie Museum of Natural History. It has an ossified element of Paaw with a volume greater than the stapes. Comparisons are made with petrosals of Didelphis marsupialis Linnaeus, 1758, and Monodelphis domestica (Wagner, 1842), also based on CT scanned specimens.

    Natural killer (NK) cells play major roles in innate immunity against viruses and cancer. Natural killer receptors (NKR) expressed by NK cells recognize foreign- or self-ligands on infected and transformed cells as well as healthy cells. NKR genes are the most rapidly evolving loci in vertebrates, and it is generally difficult to detect orthologues in different taxa. The unique exception is NKp30, an activating NKR in mammals that binds to the self-ligand B7H6. The NKp30-encoding gene, NCR3, has been found in most vertebrates including sharks, the oldest vertebrates with human-type adaptive immunity. NCR3 has a special, non-rearranging VJ-type immunoglobulin superfamily (IgSF) domain that predates the emergence of the rearranging antigen receptors. Herein we show that NCR3 loci are linked to the shark major histocompatibility complex (MHC), proving NCR3’s primordial association with the MHC. We identified eight subtypes of differentially expressed highly divergent shark NCR3 family genes. Using in situ hybridization, we detected one subtype, NS344823, to be expressed by predominantly single cells outside of splenic B cell zones. The expression by non-B cells was also confirmed by PCR in peripheral blood lymphocytes. Surprisingly, high expression of NS344823 was detected in the thymic cortex, demonstrating NS344823 expression in developing T cells. Finally, we show for the first time that shark T cells are found as single cells or in small clusters in the splenic red pulp, also unassociated with the large B cell follicles we previously identified.
