Title: Discovery of a new gall-inducing species, Aciurina luminaria (Insecta, Diptera, Tephritidae) via multi-trait integrative taxonomy
Integrative taxonomic practices that combine multiple lines of evidence for species delimitation greatly improve our understanding of intra- and inter-species variation and biodiversity. However, extended phenotypes remain underutilized despite their potential as a species-specific set of extracorporeal morphological and life history traits. Primarily relying on variations in wing patterns has caused taxonomic confusion in the genusAciurina, which are gall-inducing flies on Asteraceae plants in western North America. However, species display distinct gall morphologies that can be crucial for species identification. Here we investigate a unique gall morphotype in New Mexico and Colorado that was previously described as a variant of that induced byAciurina bigeloviae(Cockerell, 1890). Our analysis has discovered several consistent features that distinguish it from galls ofA. bigeloviae. A comprehensive description ofAciurina luminariaBaine,sp. nov.and its gall is provided through integrative taxonomic study of gall morphology, host plant ecology, wing morphometrics, and reduced-representation genome sequencing.  more » « less

  1. Four species of the genusPanaspis–P. cabindae,P. wahlbergi,P. maculicollisandP. mocamedensis– are currently known from Angola. The analysis of recently collected specimens from Serra da Neve Inselberg, an isolated mountain located in northern Namibe Province, revealed unexpected taxonomic diversity in the group. Using an integrative taxonomy approach based on morphological and DNA sequence data, with both mitochondrial (16S) and nuclear (RAG-1) genes, we were able to distinguish two distinct populations, described here as two new species,Panaspis ericaesp. nov.andP. mundavambosp. nov.Both species are assumed to be endemic to the inselberg. This reinforces our notion of southwestern Angola as a hotspot of skink diversity, and highlights the urgent need for the conservation of Serra da Neve.

  2. Solenogastres is a group of mollusks with evolutionary and ecological importance. Nevertheless, their diversity is underestimated and knowledge about the distribution of the approximately 300 formally described species is limited. Factors that contribute to this include their small size and frequent misidentification by non-specialists. Recent deep-sea explorations have resulted in the collection of numerous specimens through effective methods such as epibenthic sledges. However, this is a costly, labor-intensive, and destructive methodology. In contrast, Autonomous Reef Monitoring Structures (ARMS) offer a novel, non-destructive approach, by providing a substrate for benthic organism colonization. This study is the first to describe Solenogastres collected using ARMS, demonstrating that they are an effective tool for biodiversity assessment and characterizing rare marine invertebrates. Following an integrative taxonomic approach, two new solenogaster species are described:Dondersia tweedtaeFarris, Olson & Kocot,sp. nov.(Dondersiidae) andEleutheromenia bullescensCobo,sp. nov.(Pruvotinidae). The diagnosis of the family Dondersiidae is amended and the necessity of reassessing the validity of the current diagnostic characters for Pruvotinidae, and its classification is emphasized. The two newly described species exhibit distinct external characteristics;D. tweedtaesp. nov.has a striking pink color with a bright yellow dorsal keel andE. bullescenssp. nov.has a unique, discontinuous dorsal keel with nearly spherical protrusions. The presence of cnidocytes in the digestive systems of both species indicate that they feed on cnidarians. It is hypothesized that, like in some nudibranchs, their coloration and body features reflect defensive adaptations related to their diet. This study shows that while habitus alone is typically insufficient for accurate identification in solenogasters, it can sometimes simplify the process. For this, live observations and photographs are essential.

  3. Summary Agrobacterium tumefaciens, the causal agent of plant crown gall disease, has been widely used to genetically transform many plant species. The inter‐kingdom gene transfer capability madeAgrobacteriuman essential tool and model system to study the mechanism of exporting and integrating a segment of bacterial DNA into the plant genome. However, many biological processes such asAgrobacterium‐host recognition and interaction are still elusive. To accelerate the understanding of this important plant pathogen and further improve its capacity in plant genetic engineering, we adopted a CRISPR RNA‐guided integrase system forAgrobacteriumgenome engineering. In this work, we demonstrate thatINsertion ofTransposableElements byGuideRNA–AssistedTargEting (INTEGRATE) can efficiently generate DNA insertions to enable targeted gene knockouts. In addition, in conjunction with Cre‐loxPrecombination system, we achieved precise deletions of large DNA fragments. This work provides new genetic engineering strategies forAgrobacteriumspecies and their gene functional analyses.

This revision is based on sampling efforts over the past three decades in the southern Appalachian Mountains which have providedNesticus(Araneae, Nesticidae) collections of approximately 2100 adult specimens from more than 475 unique collecting events. Using a “morphology first” framework we examined recently collected specimens plus museum material to formulate morphology-based species hypotheses for putative new taxa (discovery phase). Using sequence capture of nuclear ultraconserved elements (UCEs) we analyzed 801 nuclear loci to validate new (and prior) morphology-based species hypotheses (validation phase) and reconstructed a robust backbone phylogeny including all described and new species. Sanger sequencing and UCE-bycatch were also used to gather mitochondrial data for more than 240 specimens. Based on our integrative taxonomic framework ten newNesticusspecies are herein described, includingN. binfordaesp. nov.,N. bondisp. nov.,N. caneisp. nov.,N. cherokeensissp. nov.,N. dellingerisp. nov.,N. dykemanaesp. nov.,N. jemisinaesp. nov.,N. lowderisp. nov.,N. roanensissp. nov., andN. templetonisp. nov.Previously unknown males are also described forN. bishopiGertsch, 1984,N. crosbyiGertsch, 1984, andN. silvanusGertsch, 1984, as well as the previously unknown female forN. mimusGertsch, 1984. Based on combined evidenceN. cooperiGertsch, 1984 is placed in synonymy withN. reclususGertsch, 1984. Overall, the montane radiation of AppalachianNesticusreveals a general lack of species sympatry and compelling biogeographic patterns. Several regionalNesticustaxa are rare, microendemic habitat specialists that deserve conservation attention and detailed future monitoring as conservation sentinels.

The rarely encountered spider genusHexurellaGertsch & Platnick, 1979 includes some of the smallest mygalomorph spiders in the world, with four poorly known taxa from central and southeastern montane Arizona, southern California, and northern Baja California Norte. At time of description the genus was known from fewer than 20 individuals, with sparse natural history information suggesting a vagrant, web-building, litter-dwelling natural history. Here the first published taxonomic and natural history information for this taxon is provided in more than 50 years, working from extensive new geographic sampling, consideration of male and female morphology, and sequence capture-based nuclear phylogenomics and mitogenomics. Several new species are easily diagnosed based on distinctive male morphologies, while a complex of populations from central and northern Arizona required an integrative combination of genomic algorithmic species delimitation analyses and morphological study. Four new species are described, includingH. ephedrasp. nov.,H. uwiiltilsp. nov.,H. xericasp. nov., andH. zassp. nov.Females ofH. encinaGertsch & Platnick, 1979 are also described for the first time. It is predicted that additional new species will ultimately be found in the mountains of central and northwestern Arizona, northern mainland Mexico, and the Mojave Desert of California.
