Title: Enhancing chitosan-zein films with Bergenia ciliata extracts toward smart food packaging
Abstract This research aims to develop chitosan-zein protein films supplemented withBergenia ciliata(Bc) extract, a traditionally important medical herb of Himalayan origin. The film’s physicochemical, mechanical, antioxidant, and antimicrobial properties were systematically explored. The opacity of chitosan film increased from 2.42 ± 0.97 to 10.32 ± 1.44 upon introducing zein (Z) protein in chitosan (Cs) in a 1:2 ratio (w/w); conferring enhanced UV-blocking attributes. IncorporatingB. ciliataextracts in the chitosan-zein film (Cs-Z-Bc) under optimized conditions further increased the opacity to 16.27 ± 1.03 without compromising the tensile strength. The α-diphenyl-β-picrylhydrazyl scavenging activity of the Cs-Z-Bc film was found to be 97.07 ± 1.09%. Additionally, these optimized films displayed significant antimicrobial efficacy, with zones of inhibition of 11.4 mm measured for gram-positive strains likeC. subtilisandS. aureusand 11.2 mm and 11.1 mm forE. coliandK. pneumoniae(gram-negative) bacterial strains. The film also showed excellent biodegradable properties. The shelf life study of Himalayan cheese was significantly increased when wrapped with the film. These findings suggested thatB. ciliataextract-fortified chitosan-zein films can be an excellent food packaging material.  more » « less

    This work presents an energy efficient technique for fabricating flexible thermoelectric generators while using printable ink. We have fabricated thermoelectric composite thick films using two different mesh sizes of n-type bismuth particles, various binder to thermoelectric material weight ratios, and two different pressures, 200 MPa and 300 MPa, in order to optimize the thermoelectric properties of the composite films. The use of chitosan dissolved in dimethylsulfoxide with less than 0.2 wt. % of chitosan, the first time chitosan has been used in this process, was sufficient for fabricating TE inks and composite films. Low temperature curing processes, along with uniaxial pressure, were used to evaporate the solvent from the drop-casted inks. This combination reduced the temperature needed compared to traditional curing processes while simultaneously increasing the packing density of the film by removing the pores and voids in the chitosan-bismuth composite film. Microstructural analysis of the composite films reveals low amounts of voids and pores when pressed at sufficiently high pressures. The highest performing composite film was obtained with the weight ratio of 1:2000 binder to bismuth, 100-mesh particle size, and 300 MPa of pressure. The best performing bismuth chitosan composite film that was pressed at 300 MPa had a power factor of 4009 ± 391 μW/m K2 with high electrical conductivity of 7337 ± 522 S/cm. The measured thermal conductivity of this same sample was 4.4 ± 0.8 W/m K and the corresponding figure of merit was 0.27 at room temperature.

  2. Titanium dioxide (TiO2)/nitrogen-doped graphene (NG) nanocomposite is prepared via a solvent-free hydrothermal reaction. The resulting TiO2/NG materials exhibit a reduction of the band gap energy compared to pristine TiO2 from 3.27 eV to 2.69 eV. These materials are characterized by scanning transmission electron microscopy (STEM), energy dispersive X-ray spectroscopy (EDX), and X-ray photoelectron spectroscopy (XPS). To prepare biopolymer films with photocatalytic properties, TiO2 and NG are mixed with biodegradable chitosan and spin-coated on a silicon wafer. Film roughness and thickness are evaluated by atomic force microscopy (AFM). These films are then tested for ciprofloxacin photodegradation by irradiating with visible light. In comparison to the TiO2/chitosan films, the addition of NG substantially enhances photodegradation efficiency by up to 34% upon the addition of 5% w/w of NG. Furthermore, this film is shown to be a good substrate for biomarker detection using laser desorption ionization mass spectrometry (LDI-MS). In summary, this nanocomposite-biopolymer film provides good photocatalytic activity towards ciprofloxacin degradation and enhances the ionization efficiency of peptide biomarkers in LDI-MS owing to high efficiency of laser absorption/desorption. This nanocomposite film might be useful for environmental-related and medical application.

    Diabetic patients are especially susceptible to chronic wounds of the skin, which can lead to serious complications. Sodium citrate is one potential therapeutic molecule for the topical treatment of diabetic ulcers, but its viability requires the assistance of a biomaterial matrix. In this study, nanofibers and thin films fabricated from natural corn zein protein are explored as a drug delivery vehicle for the topical drug delivery of sodium citrate. Corn zein is cheap and abundant in nature, and easily extracted with high purity, while nanofibers are frequently cited as ideal drug carriers due to their high surface area and high porosity. To further reduce costs, the 1-D nanofibers in this study were fabricated through an air jet-spinning method rather than the conventional electrospinning method. Thin films were also created as a comparative 2-D material. Corn zein composite nanofibers and thin films with different concentration of sodium citrate (1–30%) were analyzed through FTIR, DSC, TGA, and SEM. Results reveal that nanofibers are a much more effective vehicle than films, with the ability to interact with sodium citrate. Thermal analysis results show a stable material with low degradation, while FTIR reveals strong control over the protein secondary structures and hold of citrate. These tunable properties and morphologies allow the fibers to provide a sustained release of citrate and then revert to their structure prior to citrate loading. A statistical analysis via t-test confirmed a significant difference between fiber and film drug release. A biocompatibility study also confirms that cells are much more tolerant of the porous nanofiber structure than the nonporous protein films, and lower percentages of sodium citrate (1–5%) were outperformed to higher percentages (15–30%). This study demonstrated that protein-based nanofiber materials have high potential as vehicles for the delivery of topical diabetic drugs.

  4. Abstract Group BStreptococcus(GBS) is an encapsulated Gram‐positive bacterial pathogen that causes severe perinatal infections. Human milk oligosaccharides (HMOs) are short‐chain sugars that have recently been shown to possess antimicrobial and anti‐biofilm activity against a variety of bacterial pathogens, including GBS. We have expanded these studies to demonstrate that HMOs can inhibit and dismantle biofilm in both invasive and colonizing strains of GBS. A cohort of 30 diverse strains of GBS were analyzed for susceptibility to HMO‐dependent biofilm inhibition or destruction. HMOs were significantly effective at inhibiting biofilm in capsular‐type‐ and sequence‐type‐specific fashion, with significant efficacy in CpsIb, CpsII, CpsIII, CpsV, and CpsVI strains as well as ST‐1, ST‐12, ST‐19, and ST‐23 strains. Interestingly, CpsIa as well as ST‐7 and ST‐17 were not susceptible to the anti‐biofilm activity of HMOs, underscoring the strain‐specific effects of these important antimicrobial molecules against the perinatal pathogenStreptococcus agalactiae.

    The development of innovative antimicrobial materials is crucial in thwarting infectious diseases caused by microbes, as drug-resistant pathogens are increasing in both number and capacity to detoxify the antimicrobial drugs used today. An ideal antimicrobial material should inhibit a wide variety of bacteria in a short period of time, be less or not toxic to normal cells, and the fabrication or synthesis process should be cheap and easy. We report a one-step microwave-assisted hydrothermal synthesis of mixed composite CuxFeyOz (Fe2O3/Cu2O/CuO/CuFe2O) nanoparticles (NPs) as an excellent antimicrobial material. The 1 mg/mL CuxFeyOz NPs with the composition 36% CuFeO2, 28% Cu2O and 36% Fe2O3 have a general antimicrobial activity greater than 5 log reduction within 4 h against nine important human pathogenic bacteria (including drug-resistant bacteria as well as Gram-positive and Gram-negative strains). For example, they induced a >9 log reduction in Escherichia coli B viability after 15 min of incubation, and an ~8 log reduction in multidrug-resistant Klebsiella pneumoniae after 4 h incubation. Cytotoxicity tests against mouse fibroblast cells showed about 74% viability when exposed to 1 mg/mL CuxFeyOz NPs for 24 h, compared to the 20% viability for 1 mg/mL pure Cu2O NPs synthesized by the same method. These results show that the CuxFeyOz composite NPs are a highly efficient, low-toxicity and cheap antimicrobial material that has promising potential for applications in medical and food safety.
