Synopsis The emergence and spread of infectious diseases is a significant contributor to global amphibian declines, requiring increased surveillance and research. We assessed host–vector–parasite dynamics using a population of eastern hellbender salamanders (Cryptobranchus alleganiensis) that harbor leeches (Placobdella appalachiensis) that transmit endoparasitic blood parasites (Trypanosoma spp) to the host, with coinfection frequently occurring. We centered our study on adult males throughout their extended 8-month paternal care period because recent research indicates that nest failure caused by lack of paternal care and filial cannibalism is contributing to hellbender population declines. Recognizing the potential for parasites to modulate host physiology and behavior, we explored how infection severity influences paternal health and reproductive success. We assessed white blood cell profiles of adult male hellbenders in response to parasites, coinfection, and seasonal temperature fluctuations, while also investigating whether parasite infection or coinfection was predictive of nest success. We found that hellbenders exhibited seasonal shifts in white blood cell indices; as temperatures increased across seasons (from 5°C to 20°C), the proportion of neutrophils and eosinophils decreased (by 14% and 46%, respectively) in circulation while the proportion of lymphocytes and basophils increased (by 8% and 101%, respectively). Moreover, the proportion of neutrophil precursors increased by 80% under colder temperatures, which signifies seasonal immune cell recruitment. We demonstrated that neutrophils and eosinophils increased while lymphocytes decreased in response to leech infection. However, as leech and trypanosome infection intensity increased together, the proportion of lymphocytes increased while neutrophils and eosinophils decreased, underscoring the complex interactions between coinfection and immune responses of hellbenders that warrant future research. Despite the influence of infection and coinfection on hellbender physiology, we detected no evidence to support the hypothesis that parasites influence the likelihood of nest failure or whole-clutch filial cannibalism. In light of amphibian declines being exacerbated by climate change and disease, our study emphasizes the need to establish hematological reference values that account for physiological adaptations to seasonal fluctuations in temperature and different life history stages and to study the physiological responses of imperiled amphibian species to parasites.

Association Between Reduced Upstream Riparian Forest Cover and Impaired Development of Embryos From Eastern Hellbenders ( Cryptobranchus alleganiensis )
ABSTRACT Populations of the Eastern Hellbender, (a large‐bodied, fully aquatic salamander) inhabiting stream reaches with low catchment‐wide riparian forest cover upstream, have experienced population declines and a shift toward a geriatric population age structure. These population declines and demographic shifts might be attributed to reduced embryo viability. Reduced egg quality/viability could negatively affect recruitment and has also been known to trigger filial cannibalism in other species. Therefore, we hypothesized that in comparison to high forest cover sites, hellbender eggs collected from low forest cover sites would have a greater incidence of developmental abnormalities and lower overall viability, and that this would predict whole‐clutch cannibalism by the attending male.We collected a subset of eggs (~20–35) from 99 clutches across sites with variable upstream riparian forest cover and reared these eggs through hatching in stream water under controlled laboratory conditions. At the same time, we monitored the fate of the remaining eggs from the same clutches in the field to document the frequency of whole‐clutch filial cannibalism.We found that eggs collected from sites with lower upstream forest cover had significantly shorter embryonic development times and produced a lower percentage of viable hatchlings (hatchlings with normal development times and morphology). The average modelled viability of hatchlings was 70% higher in sites with the highest forest cover compared to our sites with the lowest forest cover. In contrast to our predictions, we did not find evidence to suggest that egg viability in the lab predicted whole‐clutch cannibalism in the field.Although forest cover was a significant predictor of egg viability and underdevelopment, substantial variance in embryonic developmental traits was unaccounted for in our models suggesting that traits associated with adults (e.g., egg and/or sperm quality) may also play a role in determining developmental outcomes. Further experiments are needed to identify what factors (e.g., egg quality, water quality) disrupt the embryonic development of hellbenders as well as the proximate stimulus that causes adult male hellbenders to eat their young.Our results emphasise the importance of restoring and protecting riparian forest cover to conserve sensitive stream species.

Synopsis The rapid environmental changes associated with the Anthropocene mean that flexible behavioral responses may be a critical determinant of animals’ resiliency to anthropogenic disturbance, particularly for species with long generation times and low vagility. One type of behavior that exemplifies this potentially important flexibility is parental care. Eggs and juvenile animals are sensitive to environmental stressors, and the ability of parents to adjust care behaviors to buffer their offspring from rapidly changing conditions may be critical to successful reproduction. In this study, we explore the role of parental care in buffering eggs from anthropogenic stressors in the long-lived, fully aquatic eastern hellbender salamander (Cryptobranchus alleganiensis alleganiensis). Using custom-designed infrared cameras installed in underwater artificial shelters in a natural stream, we describe hellbender paternal care behaviors in greater detail than has previously been possible, assess the extent to which hellbender fathers buffer their eggs from increasing levels of silt and decreasing concentrations of dissolved oxygen in nesting cavities, and describe the possible trade-offs that hellbender fathers exhibit between paternal care and self-maintenance behaviors. We found that while hellbender parents buffered their offspring from low dissolved oxygen concentrations by increasing parental care, there was an apparent trade-off between parental care and self-maintenance responses to low oxygen. Hellbender fathers did not show evidence of buffering their offspring from the effects of increasing silt or organic material in their nest cavities. We also found that filial cannibalism is a widespread behavior across nests, with almost all fathers exhibiting some cannibalism, although the extent varied widely. Our study indicates that hellbender parents may be able to reduce the impacts of declines in dissolved oxygen concentration on their offspring to a limited extent, but they may be unable to fully protect offspring from increasing silt.more » « less

Abstract Ecological and life-history variation and both interspecific and intraspecific brood parasitism contribute to diversity in egg phenotype within the same species. In this study, Barn Swallows (Hirundo rustica erythrogaster) laid eggs with high intraclutch repeatability in egg size, shape, and maculation. Despite this high intraclutch repeatability, last-laid eggs had consistently less of the eggshell covered in spots and fewer spots than earlier-laid eggs in the clutch. We examined sources of interclutch and intraclutch variation using both direct measurements and custom software (SpotEgg, NaturePatternMatch) that provide detailed information on egg characteristics, especially maculation measures. In addition to our main findings, maculation on different sides of the egg was highly repeatable; however, only shape, proportion of the eggshell maculated, and average spot size were repeatable between first and replacement clutches. Low intraclutch variation in maculation could allow females to recognize their clutch and this may be adaptive for colonial nesting species, such as the Barn Swallow. Characterizing intraspecific variation in egg size, shape, and maculation is the first step in understanding whether intraclutch variation is low enough—and interclutch variation high enough—such that eggs could serve as identity signals.more » « less

In many species, demographic assessments of population viability require an estimate of the number or proportion of breeding adults in a population that are male (the breeding sex ratio). However, this estimate is often difficult to obtain directly in species with multiple paternity when males are difficult to sample. Parentage analysis of breeding females and offspring can produce this estimate by identifying the number of unique males that contribute genetic information to (i.e., sired) a given cohort. There is an added challenge of choosing a sample design with the desired level of confidence to identify all the fathers contributing to a cohort, either at the scale of individual clutches or an entire nesting season, given limited resources. Sampling effort can be defined as the number of offspring sampled per clutch, or the number of clutches sampled per breeding season, depending on the analysis. The minimum number of samples required may depend on the proportions of eggs that different fathers fertilize in a clutch (the paternal contribution mode), the total number of fathers fertilizing a clutch, the proportion of adults available for breeding that are male (the operational sex ratio), and population size. We conducted power analyses to quantify the confidence in identifying all fathers in animal populations with multiple paternity. We simulated sampling a theoretical sea turtle population with a range of population demographics, mating systems, and sampling effort, and used the proportion of 10,000 simulations in which all fathers were identified as a proxy for confidence. At the clutch level, confidence was strongly dependent on the paternal contribution mode, and when it was skewed, it also depended on the total number of fathers contributing and the number of offspring sampled. However, sampling about one third of a clutch was sufficient to identify all fathers with high confidence for most scenarios, unless the paternal contribution mode was extremely skewed and there were many contributing fathers, such that some fathers fertilized very few eggs and were difficult to detect. At the scale of an entire nesting season, confidence was more strongly affected by the operational sex ratio, the proportion of clutches sampled, and the presence or absence of polygyny than by the lesser effects of paternal contribution mode and within-clutch sample size. Sampling fewer offspring from more clutches increased confidence compared to sampling more offspring from fewer clutches. Relaxing the minimum required proportion of fathers identified from 100% to 90% led to high confidence while sampling 50% to a maximum of 75% of clutches, depending on the mating system, even as the population size increased by an order of magnitude. Our approach and results can be widely informative for sample design as well as quantifying uncertainty in existing and future estimates of the number of breeding males in populations with multiple paternity.more » « less

The number of offspring parents rear varies considerably among closely related lineages, individuals, and even reproductive events by one individual. Constraints can come at any point, from the costs of producing a large clutch to the cost of caring for a large brood of dependent young. We report here on observations of the early phase of a reproductive cycle in a captive colony of Oophaga pumilio, a poison frog with offspring entirely dependent on maternally provisioned trophic eggs. We tracked reproductive clutches, and found that while clutch and egg size were variable, neither predicted the success of a clutch; there was also no evidence of a trade-off between clutch and egg size. Larger eggs did, however, produce larger tadpoles, and when parents transported only a subset of the brood to nurseries, the tadpoles they moved were larger than the ones they did not. Adaptive adjustment of parental investment is a key life-history trait, and a complete accounting of the way families are constructed is key to understanding the evolution of parental care, parental favoritism, and cooperation and conflict among closely related individuals.more » « less
