Title: Crystalline aliphatic polyesters from eight‐membered cyclic (di)esters
Abstract Ring‐opening polymerization (ROP) of lactones or cyclic (di)esters is a powerful method to produce well‐defined, high‐molecular‐weight (bio)degradable aliphatic polyesters. While the ROP of lactones of various ring sizes has been extensively studied, the ROP of the simplest eight‐membered lactone, 7‐heptanolactone (7‐HL), has not been reported using metal‐based catalysts. Accordingly, this contribution reports the ROP of 7‐HL via metal‐catalyzed coordinative‐insertion polymerization to the corresponding high‐molecular‐weight polyester, poly(7‐hydroxyheptanoate) (P7HHp). The resulting P7HHp is a semi‐crystalline material, with aTmof 68 °C, which is ~10 °C higher than poly(ε‐caprolactone) derived from the seven‐membered lactone. Mechanical testing showed that P7HHp is a hard and tough plastic, with elongation at break >670%. P7HHp‐based polyesters with higherTmvalues have been achieved through stereoselective copolymerization of 7‐HL with an eight‐membered cyclic diester, racemic dimethyl diolide (rac‐8DLMe), known to lead to highTmpoly(3‐hydroxyburtyrate) (P3HB). Notably, catalyst's strong kinetic preference for polymerizingrac‐8DLMeover 7‐HL in the 1/1 comonomer mixture rendered the formation of di‐block copolymer P3HB‐b‐P7HHp, showing two crystalline domains withTm1 ~ 65 °C andTm2 ~ 160 °C. Semi‐crystalline random copolymers withTmup to 164 °C have also been obtained by adjusting copolymerization conditions. Mechanical testing showed that P3HB‐b‐P7HHp can synergistically combine the high modulus of isotactic P3HB with the high ductility of P7HHp.  more » « less

Abstract Bacterial poly(3-hydroxybutyrate) (P3HB) is a perfectly isotactic, crystalline material possessing properties suitable for substituting petroleum plastics, but high costs and low volumes of its production are impractical for commodity applications. The chemical synthesis of P3HB via ring-opening polymerization (ROP) of racemicβ-butyrolactone has attracted intensive efforts since the 1960s, but not yet produced P3HB with high isotacticity and molecular weight. Here, we report a route utilizing racemic cyclic diolide (rac-DL) derived from bio-sourced succinate. With stereoselective racemic catalysts, the ROP ofrac-DL under ambient conditions produces rapidly P3HB with perfect isotacticity ([mm] > 99%), high melting temperature (Tm = 171 °C), and high molecular weight (Mn = 1.54 × 105 g mol−1,Đ = 1.01). With enantiomeric catalysts, kinetic resolution polymerizations ofrac-DL automatically stops at 50% conversion and yields enantiopure (R,R)-DL and (S,S)-DL with >99%e.e. and the corresponding poly[(S)-3HB] and poly[(R)-3HB] with highTm = 175 °C.

  2. Abstract Bacterial polyhydroxyalkanoates (PHAs) are a unique class of biodegradable polymers because of their biodegradability in ambient environments and structural diversity enabled by side‐chain groups. However, the biosynthesis of PHAs is slow and expensive, limiting their broader applications as commodity plastics. To overcome such limitation, the catalyzed chemical synthesis of bacterial PHAs has been developed, using the metal‐catalyzed stereoselective ring‐opening (co)polymerization of racemic cyclic diolides (rac‐8DLR, R=alkyl group). In this combined experimental and computational study, polymerization kinetics, stereocontrol, copolymerization characteristics, and the properties of the resulting PHAs have been examined. Most notably, stereoselective copolymerizations ofrac‐8DLMewithrac‐8DLR(R=Et, Bu) have yielded high‐molecular‐weight, crystalline isotactic PHA copolymers that are hard, ductile, and tough plastics, and exhibit polyolefin‐like thermal and mechanical properties.

Synthesis of a new mononuclear magnesium complex with a bulky bis(alkoxide) ligand environment and its reactivity in ring-opening polymerization (ROP) and ring-opening copolymerization (ROCOP) are reported. Reaction of n -butyl- sec -butylmagnesium with two equivalents of HOR (HOR = di- tert -butylphenylmethanol, HOC t Bu 2 Ph) formed Mg(OR) 2 (THF) 2 . The reaction proceeded via the Mg(OR)( sec -Bu)(THF) 2 intermediate that was independently synthesized by treating n -butyl- sec -butylmagnesium with one equivalent of HOR. Mg(OR) 2 (THF) 2 led to active albeit not well-controlled ROP of rac -lactide. In contrast, well-controlled ROCOP of epoxides with cyclic anhydrides was observed, including efficient and alternating copolymerization of phthalic anhydride with cyclohexene oxide as well as rare copolymerization of phthalic anhydride with limonene oxide and terpolymerization of phthalic anhydride with both cyclohexene oxide and limonene oxide. In addition, novel copolymerization of dihydrocoumarin with limonene oxide is described.

  4. Lactones are cyclic esters with extensive applications in materials science, medicinal chemistry, and the food and perfume industries. Nature’s strategy for the synthesis of many lactones found in natural products always relies on a single type of retrosynthetic strategy, a C−O bond disconnection. Here, we describe a set of laboratory-engineered enzymes that use a new-tonature C−C bond-forming strategy to assemble diverse lactone structures. These engineered “carbene transferases” catalyze intramolecular carbene insertions into benzylic or allylic C−H bonds, which allow for the synthesis of lactones with different ring sizes and ring scaffolds from simple starting materials. Starting from a serine-ligated cytochrome P450 variant previously engineered for other carbene-transfer activities, directed evolution generated a variant P411-LAS-5247, which exhibits a high activity for constructing a five-membered ε-lactone, lactam, and cyclic ketone products (up to 5600 total turnovers (TTN) and >99% enantiomeric excess (ee)). Further engineering led to variants P411-LAS-5249 and P411-LAS-5264, which deliver six-membered δ-lactones and seven-membered ε-lactones, respectively, overcoming the thermodynamically unfavorable ring strain associated with these products compared to the γ-lactones. This new carbene-transfer activity was further extended to the synthesis of complex lactone scaffolds based on fused, bridged, and spiro rings. The enzymatic platform developed here complements natural biosynthetic strategies for lactone assembly and expands the structural diversity of lactones accessible through C−H functionalization.

Organocatalyzed ring-opening polymerization (O-ROP) of a six-five bicyclic lactone, 4,5- trans -cyclohexyl-fused γ-butyrolactone (4,5-T6GBL), can be topologically selective or living at room temperature, depending on catalyst structure. A screening of (thio)urea [(T)U] and organic base pairs revealed unique trends in reactivity for this monomer as well as the most active catalyst pairs, which were employed as received commercially to produce relatively high molecular weight ( M n up to 106 kDa), low dispersity ( Đ = 1.04) linear poly(4,5-T6GBL) in a living fashion. The ROP using a hybrid organic/inorganic pair of TU/KOMe in neat conditions led to poly(4,5-T6GBL) with even higher molecular weight ( M n = 215 kDa, Đ = 1.04). In comparison to the metal-catalyzed system, (T)U-base pairs exhibited competitive kinetics and reached higher monomer conversions, and their reactions can be performed in air. In addition, the resulting polymers required less purification to produce materials with higher onset decomposition temperature. (T)U-base pairs were selective towards linear polymerization only, whereas triazabicyclodecene can catalyze both polymerization and (quantitative) depolymerization processes, depending on reaction conditions. Cyclic polymers with M n = 41–72 kDa were selectively formed via N-heterocyclic carbene-mediated zwitterionic O-ROP.
