A growing interest in fungi that occur within symptom-less plants and lichens (endophytes) has uncovered previously uncharacterized species in diverse biomes worldwide. In many temperate and boreal forests, endophytic Coniochaeta (Sacc.) Cooke ( Coniochaetaceae , Coniochaetales, Sordariomycetes , Ascomycota ) are commonly isolated on standard media, but rarely are characterized. We examined 26 isolates of Coniochaeta housed at the Gilbertson Mycological Herbarium. The isolates were collected from healthy photosynthetic tissues of conifers, angiosperms, mosses and lichens in Canada, Sweden and the United States. Their barcode sequences (nuclear ribosomal internal transcribed spacer and 5.8S; ITS rDNA) were ≤97% similar to any documented species available through GenBank. Phylogenetic analyses based on two loci (ITS rDNA and translation elongation factor 1-alpha) indicated that two isolates represented Coniochaeta cymbiformispora , broadening the ecological niche and geographic range of a species known previously from burned soil in Japan. The remaining 24 endophytes represented three previously undescribed species that we characterize here: Coniochaeta elegans sp. nov., Coniochaeta montana sp. nov. and Coniochaeta nivea sp. nov. Each has a wide host range, including lichens, bryophytes and vascular plants. C. elegans sp. nov. and C. nivea sp. nov. have wide geographic ranges. C. montana sp. nov. occurs in the Madrean biome of Arizona (USA), where it is sympatric with the other species described here. All three species display protease, chitinase and cellulase activity in vitro . Overall, this study provides insight into the ecological and evolutionary diversity of Coniochaeta and suggests that these strains may be amenable for studies of traits relevant to a horizontally transmitted, symbiotic lifestyle.

This study describes the worker and queen castes of the Neotropical ponerine Corrieopone nouragues gen. nov. , sp. nov. , an ant from the tropical rainforest in French Guiana. Worker morphology of the taxon is compared with those of other Ponerinae and the similarities between them are discussed, refining the definition of character states for some diagnostic characters at the generic level, providing an identification key to the Neotropical genera, and making some adjustments to the taxonomic framework within the subfamily. Descriptions, diagnosis, character discussion, identification key, and glossary are illustrated with more than 300 images and line drawings. Open science is supported by providing access to measurement data for specimens of the new genus, a matrix of character states for all ponerine taxa evaluated in this study, and specimen data for all examined material. The new or revived combinations presented here are Pachycondyla procidua Emery, comb. rev. , Neoponera curiosa (Mackay and Mackay), comb. nov. , Leptogenys butteli (Forel), comb. nov. , and Bothroponera escherichi (Forel), comb. nov. (Ed.)Premise of research: In our modern flora, the Cycadales represent one of the oldest-known gymnosperm clades, with their evolutionary roots tracing back to the late Paleozoic. Their radiation and wide distribution in the Mesozoic are well documented by numerous fossils. In contrast, the fossil record of late Paleozoic forms is restricted to a few specimens, thus leaving open questions on the origin and early evolution of cycads. A petrified stem with cycadalean affinity has been found in Saxony (central-eastern Germany) in Holocene gravel deposits of the Zwickauer Mulde river as a result of fluvial transportation from its original lower Permian locality. Methodology: The specimen was sectioned transversely, radially, and tangentially. Its provenance was clarified by comparing fossil woods of various localities in the upstream catchment area. Samples of these localities were exposed to ultraviolet light of different wavelengths, revealing distinctive fluorescence patterns related to specific geochemical conditions during the fossilization process. Plant anatomical studies were performed by macroscopic and microscopic documentation, including photo scanning, microscopic photography, and anatomical measurements. Pivotal results: Cycadodendron galtieri gen. nov. et sp. nov. originates from the Chemnitz Fossil Lagerstätte, an autochthonous floral assemblage of early Permian age (291 Ma; Sakmarian–Artinskian) that has been buried and conserved by pyroclastics in a geological instant. Several anatomical characteristics provide evidence for its cycadalean affinity: (1) a wide pith with numerous scattered medullary bundles, (2) a pith-peripheral vascular system with endarch primary xylem bundles, (3) at least nine successive vascular segments with secondary xylem and phloem, (4) araucarioid-type pitting of secondary xylem tracheids, and (5) primary rays and medullary bundles traversing the vascular segments. Conclusions: Cycadodendron galtieri gen. nov. et sp. nov. provides insights into stem anatomical characteristics of the oldest-known cycads, for example, revealing that polyxyly was an early-derived feature in cycad evolution. Its provenance reveals that the arborescent but generally small taxon was part of intramontane forested landscapes thriving on a well-drained mineral substrate in an alluvial plain setting and experiencing seasonally dry paleoclimate.more » « less

null (Ed.)The anaerobic gut fungi (AGF; phylum Neocallimastigomycota ) reside in the alimentary tracts of herbivores. Multiple novel, yet-uncultured AGF taxa have recently been identified in culture-independent diversity surveys. Here, we report on the isolation and characterization of the first representative of the RH5 lineage from faecal samples of a wild blackbuck (Indian Antelope, Antilope cervicapra ) from Sutton County, Texas, USA. The isolates displayed medium sized (2–4 mm) compact circular colonies on agar roll tubes and thin loose biofilm-like growth in liquid medium. Microscopic examination revealed monoflagellated zoospores and polycentric thalli with highly branched nucleated filamentous rhizomycelium, a growth pattern encountered in a minority of described AGF genera so far. The obtained isolates are characterized by formation of spherical vesicles at the hyphal tips from which multiple sporangia formed either directly on the spherical vesicles or at the end of sporangiophores. Phylogenetic analysis using the D1/D2 regions of the large ribosomal subunit (D1/D2 LSU) and the ribosomal internal transcribed spacer 1 (ITS1) revealed sequence similarities of 93.5 and 81.3%, respectively, to the closest cultured relatives ( Orpinomyces joyonii strain D3A (D1/D2 LSU) and Joblinomyces apicalis strain GFH681 (ITS1). Substrate utilization experiments using the type strain (BB-3 T ) demonstrated growth capabilities on a wide range of mono-, oligo- and polysaccharides, including glucose, xylose, mannose, fructose, cellobiose, sucrose, maltose, trehalose, lactose, cellulose, xylan, starch and raffinose. We propose accommodating these novel isolates in a new genus and species, for which the name Paucimyces polynucleatus gen. nov., sp. nov. is proposed.more » « less
