Title: An emergent temporal basis set robustly supports cerebellar time-series learning
The cerebellum is considered a “learning machine” essential for time interval estimation underlying motor coordination and other behaviors. Theoretical work has proposed that the cerebellum’s input recipient structure, the granule cell layer (GCL), performs pattern separation of inputs that facilitates learning in Purkinje cells (P-cells). However, the relationship between input reformatting and learning has remained debated, with roles emphasized for pattern separation features from sparsification to decorrelation. We took a novel approach by training a minimalist model of the cerebellar cortex to learn complex time-series data from time-varying inputs, typical during movements. The model robustly produced temporal basis sets from these inputs, and the resultant GCL output supported better learning of temporally complex target functions than mossy fibers alone. Learning was optimized at intermediate threshold levels, supporting relatively dense granule cell activity, yet the key statistical features in GCL population activity that drove learning differed from those seen previously for classification tasks. These findings advance testable hypotheses for mechanisms of temporal basis set formation and predict that moderately dense population activity optimizes learning. NEW & NOTEWORTHY During movement, mossy fiber inputs to the cerebellum relay time-varying information with strong intrinsic relationships to ongoing movement. Are such mossy fibers signals sufficient to support Purkinje signals and learning? In a model, we show how the GCL greatly improves Purkinje learning of complex, temporally dynamic signals relative to mossy fibers alone. Learning-optimized GCL population activity was moderately dense, which retained intrinsic input variance while also performing pattern separation.  more » « less

    Transcranial direct current stimulation (tDCS) of the cerebellum has rapidly raised interest but the effects of tDCS on cerebellar neurons remain unclear. Assessing the cellular response to tDCS is challenging because of the uneven, highly stratified cytoarchitecture of the cerebellum, within which cellular morphologies, physiological properties, and function vary largely across several types of neurons. In this study, we combine MRI-based segmentation of the cerebellum and a finite element model of the tDCS-induced electric field (EF) inside the cerebellum to determine the field imposed on the cerebellar neurons throughout the region. We then pair the EF with multicompartment models of the Purkinje cell (PC), deep cerebellar neuron (DCN), and granule cell (GrC) and quantify the acute response of these neurons under various orientations, physiological conditions, and sequences of presynaptic stimuli. We show that cerebellar tDCS significantly modulates the postsynaptic spiking precision of the PC, which is expressed as a change in the spike count and timing in response to presynaptic stimuli. tDCS has modest effects, instead, on the PC tonic firing at rest and on the postsynaptic activity of DCN and GrC. In Purkinje cells, anodal tDCS shortens the repolarization phase following complex spikes (-14.7 ± 6.5% of baseline value, mean ± S.D.; max: -22.7%) and promotes burstiness with longer bursts compared to resting conditions. Cathodal tDCS, instead, promotes irregular spiking by enhancing somatic excitability and significantly prolongs the repolarization after complex spikes compared to baseline (+37.0 ± 28.9%, mean ± S.D.; max: +84.3%). tDCS-induced changes to the repolarization phase and firing pattern exceed 10% of the baseline values in Purkinje cells covering up to 20% of the cerebellar cortex, with the effects being distributed along the EF direction and concentrated in the area under the electrode over the cerebellum. Altogether, the acute effects of tDCS on cerebellum mainly focus on Purkinje cells and modulate the precision of the response to synaptic stimuli, thus having the largest impact when the cerebellar cortex is active. Since the spatiotemporal precision of the PC spiking is critical to learning and coordination, our results suggest cerebellar tDCS as a viable therapeutic option for disorders involving cerebellar hyperactivity such as ataxia.

  2. Appropriate generalization of learned responses to new situations is vital for adaptive behavior. We provide a circuit-level account of generalization in the electrosensory lobe (ELL) of weakly electric mormyrid fish. Much is already known in this system about a form of learning in which motor corollary discharge signals cancel responses to the uninformative input evoked by the fish’s own electric pulses. However, for this cancellation to be useful under natural circumstances, it must generalize accurately across behavioral regimes, specifically different electric pulse rates. We show that such generalization indeed occurs in ELL neurons, and develop a circuit-level model explaining how this may be achieved. The mechanism involves regularized synaptic plasticity and an approximate matching of the temporal dynamics of motor corollary discharge and electrosensory inputs. Recordings of motor corollary discharge signals in mossy fibers and granule cells provide direct evidence for such matching.

Cerebellar granule cells (GrCs) are usually regarded as a uniform cell type that collectively expands the coding space of the cerebellum by integrating diverse combinations of mossy fiber inputs. Accordingly, stable molecularly or physiologically defined GrC subtypes within a single cerebellar region have not been reported. The only known cellular property that distinguishes otherwise homogeneous GrCs is the correspondence between GrC birth timing and the depth of the molecular layer to which their axons project. To determine the role birth timing plays in GrC wiring and function, we developed genetic strategies to access early- and late-born GrCs. We initiated retrograde monosynaptic rabies virus tracing from control (birth timing unrestricted), early-born, and late-born GrCs, revealing the different patterns of mossy fiber input to GrCs in vermis lobule 6 and simplex, as well as to early- and late-born GrCs of vermis lobule 6: sensory and motor nuclei provide more input to early-born GrCs, while basal pontine and cerebellar nuclei provide more input to late-born GrCs. In vivo multidepth two-photon Ca 2+ imaging of axons of early- and late-born GrCs revealed representations of diverse task variables and stimuli by both populations, with modest differences in the proportions encoding movement, reward anticipation, and reward consumption. Our results suggest neither organized parallel processing nor completely random organization of mossy fiber→GrC circuitry but instead a moderate influence of birth timing on GrC wiring and encoding. Our imaging data also provide evidence that GrCs can represent generalized responses to aversive stimuli, in addition to recently described reward representations.

  4. Neural dynamics can reflect intrinsic dynamics or dynamic inputs, such as sensory inputs or inputs from other brain regions. To avoid misinterpreting temporally structured inputs as intrinsic dynamics, dynamical models of neural activity should account for measured inputs. However, incorporating measured inputs remains elusive in joint dynamical modeling of neural-behavioral data, which is important for studying neural computations of behavior. We first show how training dynamical models of neural activity while considering behavior but not input or input but not behavior may lead to misinterpretations. We then develop an analytical learning method for linear dynamical models that simultaneously accounts for neural activity, behavior, and measured inputs. The method provides the capability to prioritize the learning of intrinsic behaviorally relevant neural dynamics and dissociate them from both other intrinsic dynamics and measured input dynamics. In data from a simulated brain with fixed intrinsic dynamics that performs different tasks, the method correctly finds the same intrinsic dynamics regardless of the task while other methods can be influenced by the task. In neural datasets from three subjects performing two different motor tasks with task instruction sensory inputs, the method reveals low-dimensional intrinsic neural dynamics that are missed by other methods and are more predictive of behavior and/or neural activity. The method also uniquely finds that the intrinsic behaviorally relevant neural dynamics are largely similar across the different subjects and tasks, whereas the overall neural dynamics are not. These input-driven dynamical models of neural-behavioral data can uncover intrinsic dynamics that may otherwise be missed.

  5. The goal of odor source separation and identification from real-world data presents a challenging problem. Both individual odors of potential interest and multisource odor scenes constitute linear combinations of analytes present at different concentrations. The mixing of these analytes can exert nonlinear and even nonmonotonic effects on cross-responsive chemosensors, effectively occluding diagnostic activity patterns across the array. Neuromorphic algorithms, inspired by specific computational strategies of the mammalian olfactory system, have been trained to rapidly learn and reconstruct arbitrary odor source signatures in the presence of background interference. However, such networks perform best when tuned to the statistics of well-behaved inputs, normalized and predictable in their activity distributions. Deployment of chemosensor arrays in the wild exposes these networks to disruptive effects that exceed these tolerances. To address the problems inherent to chemosensory signal conditioning and representation learning, the olfactory bulb deploys an array of strategies: (1) shunting inhibition in the glomerular layer implements divisive normalization, contributing to concentration-invariant representations; (2) feedforward gain diversification (synaptic weight heterogeneity) regularizes spiking activity in the external plexiform layer (mitral and granule cells), enabling the network to handle unregulated inputs; (3) gamma-band oscillations segment activity into packets, enabling a spike phase code and iterative denoising; (4) excitatory and inhibitory spike timing dependent learning rules induce hierarchical attraction basins, enabling the network to map its highly complex inputs to regions of a lower dimensional manifold; (5) neurogenesis in the granule cell layer enables lifelong learning and prevents order effects (regularizing the learned synaptic weight distribution over the span of training). Here, we integrate these motifs into a single neuromorphic model, bringing together prior OB-inspired model architectures. In a series of simulation experiments including real-world data from a chemosensor array, we demonstrate the network’s ability to learn and detect complex odorants in variable environments despite unpredictable noise distributions.
