Abstract AimsTo determine if the bacteriophage abortive infection system ToxIN is present in foodborne Salmonella and if it protects against infection by bacteriophages specific to enteric bacteria. Methods and resultsA set of foodborne Salmonella enteritidis isolates from a 2010 eggshell outbreak was identified via BLASTN (basic local alignment search tool nucleotide) queries as harboring a close homolog of ToxIN, carried on a plasmid with putative mobilization proteins. This homolog was cloned into a plasmid vector and transformed into the laboratory strain Salmonella typhimurium LT2 and tested against a set of Salmonella-specific phages (FelixO1, S16, Sp6, LPST153, and P22 HT105/1 int-201). ToxIN reduced infection by FelixO1, S16, and LPST153 by ∼1–4 log PFU ml−1 while reducing the plaque size of Sp6. When present in LT2 and Escherichia coli MG1655, ToxIN conferred cross-genus protection against phage isolates, which infect both bacteria. Finally, the putative ToxIN plasmid was found in whole-genome sequence contigs of several Salmonella serovars, pathogenic E. coli, and other pathogenic enterobacteria. ConclusionsSalmonella and E. coli can resist infection by several phages via ToxIN under laboratory conditions; ToxIN is present in foodborne pathogens including Salmonella and Shiga-toxigenic E. coli.

ABSTRACT Seven bacteriophages were isolated from soil in Pennsylvania and Wisconsin using the hostMicrobacterium foliorum. These bacteriophages range in the number of predicted genes encoded, from 25 to 91, and are distributed across actinobacteriophage clusters EB, EC, EE, and EK.

Abstract ObjectivesCortical bone geometry is commonly used to investigate biomechanical properties of primate mandibles. However, the ontogeny of these properties is less understood. Here we investigate changes in cortical bone cross‐sectional properties throughout capuchin ontogeny and compare captive versus wild, semi‐provisioned groups. Tufted capuchins (Sapajusspp.) are known to consume relatively hard/tough foods, while untufted capuchins (Cebusspp.) exploit less mechanically challenging foods. Previous research indicates dietary differences are present early in development and adultSapajusmandibles can resist higher bending/shear/torsional loads. Materials and methodsThis study utilized microCT scans of 22Cebusand 45Sapajusfrom early infancy to adulthood from three sample populations: one captiveCebus, one captiveSapajus, and one semi‐provisioned, free‐rangingSapajus. Mandibular cross‐sectional properties were calculated at the symphysis, P3, and M1. If the tooth had not erupted, its position within the crypt was used. A series of one‐way ANOVAs were performed to assess differences between and within the sample populations. ResultsMandible robusticity increases across ontogeny for all three sample populations.Sapajuswere better able to withstand bending and torsional loading even early in ontogeny, but no difference in shear resistance was found. Semi‐provisioned, free‐rangingSapajustend to show increased abilities to resist bending and torsional loading but not shear loading compared to captiveSapajus. DiscussionThis study helps advance our understanding of the primate masticatory system development and opens the door for further studies into adaptive plasticity in shaping the masticatory apparatus of capuchins and differences in captive versus free‐ranging sample populations.more » « less

PremiseEocene floras of Patagonia document biotic response to the final separation of Gondwana. The conifer genusAraucaria, distributed worldwide during the Mesozoic, has a disjunct extant distribution between South America and Australasia. Fossils assigned to AustralasianAraucariaSect.Eutactausually are represented by isolated organs, making diagnosis difficult.Araucaria pichileufensisE.W. Berry, from the middle Eocene Río Pichileufú (RP) site in Argentine Patagonia, was originally placed in Sect.Eutactaand later reported from the early Eocene Laguna del Hunco (LH) locality. However, the relationship ofA. pichileufensisto Sect.Eutactaand the conspecificity of theAraucariamaterial among these Patagonian floras have not been tested using modern methods. MethodsWe review the type material ofA. pichileufensisalongside large (n= 192) new fossil collections ofAraucariafromLHandRP, including multi‐organ preservation of leafy branches, ovuliferous complexes, and pollen cones. We use a total evidence phylogenetic analysis to analyze relationships of the fossils to Sect.Eutacta. ResultsWe describeAraucaria huncoensissp. nov. fromLHand improve the whole‐plant concept forAraucaria pichileufensisfromRP. The two species respectively resolve in the crown and stem of Sect.Eutacta. ConclusionsOur results confirm the presence and indicate the survival of Sect.Eutactain South America during early Antarctic separation. The exceptionally complete fossils significantly predate several molecular age estimates for crownEutacta. The differentiation of twoAraucariaspecies demonstrates conifer turnover during climate change and initial South American isolation from the early to middle Eocene.more » « less

PremiseThe genusAntennariahas a complex evolutionary history due to dioecism, excessive polyploidy, and the evolution of polyploid agamic complexes. We developed microsatellite markers fromA. corymbosato investigate genetic diversity and population genetic structure inAntennariaspecies. Methods and ResultsTwenty‐four novel microsatellite markers (16 nuclear and eight chloroplast) were developed fromA. corymbosausing an enriched genomic library. Ten polymorphic nuclear markers were used to characterize genetic variation in five populations ofA. corymbosa. One to four alleles were found per locus, and the expected heterozygosity and fixation index ranged from 0.00 to 0.675 and −0.033 to 0.610, respectively. We were also able to successfully amplify these markers in five additionalAntennariaspecies. ConclusionsThese markers are promising tools to study the population genetics of sexualAntennariaspecies and to investigate interspecific gene flow, clonal diversity, and parentage ofAntennariapolyploid agamic complexes.more » « less

RationaleThe developments of new ionization technologies based on processes previously unknown to mass spectrometry (MS) have gained significant momentum. Herein we address the importance of understanding these unique ionization processes, demonstrate the new capabilities currently unmet by other methods, and outline their considerable analytical potential. MethodsTheinletandvacuumionization methods of solvent‐assisted ionization (SAI), matrix‐assisted ionization (MAI), and laserspray ionization can be used with commercial and dedicated ion sources producing ions from atmospheric or vacuum conditions for analyses of a variety of materials including drugs, lipids, and proteins introduced from well plates, pipet tips and plate surfaces with and without a laser using solid or solvent matrices. Mass spectrometers from various vendors are employed. ResultsResults are presented highlighting strengths relative to ionization methods of electrospray ionization (ESI) and matrix‐assisted laser desorption/ionization. We demonstrate the utility of multi‐ionization platforms encompassing MAI, SAI, and ESI and enabling detection of what otherwise is missed, especially when directly analyzing mixtures. Unmatched robustness is achieved with dedicated vacuum MAI sources with mechanical introduction of the sample to the sub‐atmospheric pressure (vacuumMAI). Simplicity and use of a wide array of matrices are attained using a conduit (inletionization), preferably heated, with sample introduction from atmospheric pressure. Tissue, whole blood, urine (including mouse, chicken, and human origin), bacteria strains and chemical on‐probe reactions are analyzed directly and, especially in the case ofvacuumionization, without concern of carryover or instrument contamination. ConclusionsExamples are provided highlighting the exceptional analytical capabilities associated with the novel ionization processes in MS that reduce operational complexity while increasing speed and robustness, achieving mass spectra with low background for improved sensitivity, suggesting the potential of this simple ionization technology to drive MS into areas currently underserved, such as clinical and medical applications.more » « less
