Title: Stereoselective Approaches to β-Linked 2-Deoxy Sugars
This review presents a survey of recent developments in the synthesis of β-linked 2-deoxy sugars. Approaches ranging from catalysis to de novo synthesis are described, with a focus on methods developed in the last 10 years. Where relevant, the application of these technologies to synthesis and mechanistic information is discussed. Finally, it concludes with an examination of the scope and limitations of these technologies, as well as examinations about where the field should head next.  more » « less

  1. Abstract Tropical forests are increasingly threatened by deforestation and degradation, impacting carbon storage, climate regulations and biodiversity. Restoring these ecosystems is crucial for environmental sustainability, yet monitoring these efforts poses significant challenges. Secondary forests are in a constant state of flux, with growth depending on multiple factors.Remote sensing technologies offer cost‐effective, scalable and transferable solutions, advancing forest restoration monitoring towards more accurate, efficient and real‐time data analysis and interpretation. This review provides a comprehensive evaluation of the current state and advancements in remote sensing technologies applied to monitoring tropical forest restoration.Synthesis and applications: This review brings together the state of the art of remote sensing technologies, such as very‐high‐resolution RGB imagery, multi‐ and hyperspectral imaging, lidar, radar and thermal‐infrared technologies and their applicability in monitoring forest restoration. In conclusion, this review emphasizes the potential of remote sensing technologies, coupled with advanced computational techniques, to enhance global efforts towards effective and sustainable forest restoration monitoring.

Abstract In recent years, microfluidic technologies have emerged as a powerful approach for the advanced synthesis and rapid optimization of various solution‐processed nanomaterials, including semiconductor quantum dots and nanoplatelets, and metal plasmonic and reticular framework nanoparticles. These fluidic systems offer access to previously unattainable measurements and synthesis conditions at unparalleled efficiencies and sampling rates. Despite these advantages, microfluidic systems have yet to be extensively adopted by the colloidal nanomaterial community. To help bridge the gap, this progress report details the basic principles of microfluidic reactor design and performance, as well as the current state of online diagnostics and autonomous robotic experimentation strategies, toward the size, shape, and composition‐controlled synthesis of various colloidal nanomaterials. By discussing the application of fluidic platforms in recent high‐priority colloidal nanomaterial studies and their potential for integration with rapidly emerging artificial intelligence‐based decision‐making strategies, this report seeks to encourage interdisciplinary collaborations between microfluidic reactor engineers and colloidal nanomaterial chemists. Full convergence of these two research efforts offers significantly expedited and enhanced nanomaterial discovery, optimization, and manufacturing.

  3. Understanding the preferences for new and future transportation technologies is important to ensure an efficient and equitable future transportation system. A survey was conducted of Americans’ preferences for several such technologies. Americans are concerned about vehicle range and charging station availability for electric vehicles (EVs) and hesitant about autonomous vehicle (AV) safety. Opinions about many transportation technologies, such as vertical takeoff and landing (i.e., air taxis), shared parking, and air-drone delivery are mixed. These less familiar technologies require continued tracking of preferences. A 55% increase is estimated in the probability of an individual choosing a battery electric vehicle (BEV) pickup truck if its fuel economy increases by about 9%. This result supports a market for BEV pickup trucks currently under development by many automakers. The preference for vehicle autonomation appears to depend on the use case. Driving task automation is preferred by residents of low-density, car-dependent areas where long commutes are common. In contrast, automated parking technologies are favored by those living in denser communities. Intermittent bus lanes are favored by those living in high population density areas, but not among those in areas with high shares of zero-vehicle households. These results provide indications of where to direct future research in the field.

  4. Long oligodeoxynucleotides (ODNs) are segments of DNAs having over one hundred nucleotides (nt). They are typically assembled using enzymatic methods such as PCR and ligation from shorter 20 to 60 nt ODNs produced by automated de novo chemical synthesis. While these methods have made many projects in areas such as synthetic biology and protein engineering possible, they have various drawbacks. For example, they cannot produce genes and genomes with long repeats and have difficulty to produce sequences containing stable secondary structures. Here, we report a direct de novo chemical synthesis of 400 nt ODNs, and their isolation from the complex reaction mixture using the catching-by-polymerization (CBP) method. To determine the authenticity of the ODNs, 399 and 401 nt ODNs were synthesized and purified with CBP. The two were joined together using Gibson assembly to give the 800 nt green fluorescent protein (GFP) gene construct. The sequence of the construct was verified via Sanger sequencing. To demonstrate the potential use of the long ODN synthesis method, the GFP gene was expressed inE. coli. The long ODN synthesis and isolation method presented here provides a pathway to the production of genes and genomes containing long repeats or stable secondary structures that cannot be produced or are highly challenging to produce using existing technologies.

  5. Abstract The diversity of plant natural products presents a rich resource for accelerating drug discovery and addressing pressing human health issues. However, the challenges in accessing and cultivating source species, as well as metabolite structural complexity, and general low abundance present considerable hurdles in developing plant-derived therapeutics. Advances in high-throughput sequencing, genome assembly, gene synthesis, analytical technologies, and synthetic biology approaches, now enable us to efficiently identify and engineer enzymes and metabolic pathways for producing natural and new-to-nature therapeutics and drug candidates. This review highlights challenges and progress in plant natural product discovery and engineering by example of recent breakthroughs in identifying the missing enzymes involved in the biosynthesis of the anti-cancer agent Taxol®. These enzyme resources offer new avenues for the bio-manufacture and semi-synthesis of an old blockbuster drug.
