Mono-hydroxychlorins are uncommon macrocycles that have only been synthetically realized by modifying porphyrin rings using the harsh oxidizing agent OsO4. We show here that a more directed delivery of the mono-hydroxychlorin may be concomitantly obtained from the oxidation of porphyrinogen using the mild conditions of the high dilution Lindsey porphyrin forming reaction where water content is minimized by using dry CHCl3within the environment of a glovebox. We now report the direct synthesis of 17,18-dihydro-18-hydroxy-5,10,15,20-tetrakis-(4-fluoro,2,6-dimethylphenyl)-porphyrin (2H-TFChl-[Formula: see text]OH) together with the corresponding freebase porphyrin TFP. The TFP has been metalated with FeBr2and MgBr2•OEt2resulting in metalloporphyrins Fe(III)TFP(Cl) and Mg(II)-TFP which have been structurally characterized by single-crystal X-ray crystallography. We find that the excited state properties of the mono-hydroxychlorin are similar to that of its parent TFP and Mg(II)TFP porphyrin congeners. Excited state deactivation by vibronic coupling to the high energy O-H oscillator is circumvented with the hydroxyl group remote to the 18[Formula: see text]-electron framework of the chlorin ring. These results reveal that strong H-bonding groups may be introduced on the periphery of the chlorin ring while maintaining the light-gathering properties that lie at the heart of photosynthesis of the chlorin ring.

One-flask synthesis of zinc metalated mono - and di -hydroxychlorins
Lewis acid catalyzed condensation of pyrrole and 4-fluoro-2,6-dimethylbenzaldehyde followed by chemical oxidation afforded the corresponding chlorin along with the parent porphyrin. The subsequent metalation of the porphyrin-chlorin mixture in the presence of Zn(OAc)2•2H2O afforded Zn monoand di-hydroxychlorins in addition to the Zn porphyrin in a one-flask synthesis. This new direct hydroxylation reaction eliminates the need for highly toxic OsO4and H2S that are traditionally used for the generation of hydroxy chlorins. In addition to the full characterization of the zinc chlorins, we present cyclic voltammograms, steady-state absorption, and emission profiles of this rarely available class of compounds. Our findings show that Zn mono- and di-hydroxychlorins are stable compounds that possess exceptionally long triplet excited states in solution, making them promising candidates for photodynamic therapy.

The title chlorin, 2 Ph H 2 , hydrogen-bonded to dimethylaminopyridine (DMAP), C 44 H 32 N 4 O 2 ·C 7 H 10 N 2 , and its corresponding zinc(II) complex, 2 Ph Zn , axially coordinated to ethylenediamine (EDA), [Zn(C 44 H 30 N 4 O 2 )]·C 2 H 8 N 2 , were isolated and crystallized by adventitious reduction of the corresponding osmate esters by DMAP and EDA, respectively. Known since 1996 and, inter alia , used for the preparation of a wide range of (planar and non-planar) chlorin analogues (so-called pyrrole-modified porphyrins), their conformational analyses in the solid state are important benchmarks. Both macrocycles are only modestly distorted from planarity and both are slightly more non-planar than the corresponding dimethoxy-derivative, but less planar than a free-base meso -pentafluorophenyl-based osmate ester. NSD analyses provide quantitative and qualitative analyses of the distortion modes. One origin of the non-planarity is presumably the avoidance of the eclipsed configuration of the two vic–cis diols on the pyrroline moiety; the resulting deformation of the pyrroline translates in some cases into the macrocycle. The structure of 2 Ph H 2 features voids making up ca 26% of the unit-cell volume filled with highly disordered solvate molecules (chloroform and hexanes). 2 Ph Zn crystallized with a 13.6 (4)% occupied solvate methanol molecule.more » « less

In the centrosymmetric title complexes, di-μ-acetato-bis({N,N-dimethyl-2-[phenyl(pyridin-2-yl)methylidene]hydrazine-1-carbothioamidato}zinc(II)), [Zn2(C15H15N4S)2(C2H3O2)2] (I), and di-μ-acetato-bis({N-ethyl-2-[phenyl(pyridin-2-yl)methylidene]hydrazine-1-carbothioamidato}zinc(II)), [Zn2(C16H17N4S)2(C2H3O2)2] (II), the zinc ions are chelated by theN,N,S-tridentate ligands and bridged by pairs of acetate ions. The acetate ion in (I) is disordered over two orientations in a 0.756 (6):0.244 (6) ratio, leading to different zinc coordination modes for the major (5-coordinate) and minor (6-coordinate) disorder components. Geometrical indices [τ5= 0.32 and 0.30 for (I) (major component) and (II), respectively] suggest the zinc coordination in these phases to be distorted square pyramidal. This study forms part of our aim to discern the mechanism of metal binding in these chelators, their specificity and selectivity, and to gain insight into the role of cellular zinc in physiological processes such as infection, immunity and cancer.more » « less

Reactions of the bicompartmental bis(phenolato) compound 6,6′-methylenebis(2-((bis(pyridin-2-ylmethyl)amino)methyl)-4-chlorophenol)hemihydrate (H 2 L ½H 2 O) with 3d metal( ii ) ions afforded novel fully structurally characterized bridged acetato dinuclear complexes [Mn 2 (HL)(μ 1,2 -OAc) 2 ]PF 6 (1) [Zn 2 (HL)(μ 1,2 -OAc)(H 2 O) 0.75 (MeOH) 0.25 ](PF 6 ) 2 ·0.45(H 2 O) (5) and [Cd 2 (HL)(μ 1,1,2 -OAc)(OAc)(H 2 O)]PF 6 ·H 2 O (6) as well as the polymeric bridged-azido tetranuclear catena -[Cu 4 (HL) 2 (μ 1,1 -N 3 ) 2 (μ 1,3 -N 3 ) 2 ](NO 3 ) 2 ·5H 2 O (4). The complex [Cu 4 (HL) 2 (ClO 4 ) 3 (H 2 O) 5 ](ClO 4 ) 3 ·5H 2 O (2) was partially characterized. In addition, three more dinuclear complexes [Cu 2 (H 2 L)(NO 3 ) 2 (H 2 O) 2 ](NO 3 ) 2 (3), [Cu 2 (HL)(OAc)(CH 3 OH)](PF 6 ) 2 (7) and [Cu 2 (HL)(NCS) 2 ]NO 3 ·2H 2 O (8) were also isolated. All complexes were characterized by CHN elemental analysis, IR and UV-Vis spectroscopy, ESI-MS, conductivity measurements and X-ray single crystal crystallography for compounds 1, 4, 5 and 6, where the bis(phenolato) ligand displayed different deprotonation (H 2 L, HL − and L 2− ). The magnetic susceptibility measurements over the temperature range 2–300 K revealed very weak antiferromagnetic coupling in dimanganese( ii ) 1 ( J = −1.64(1) cm −1 ) and almost negligible magnetic interaction in dicopper( ii ) 2 ( J = 0(3) cm −1 ). In the azido catena -[Cu 4 (HL) 2 (μ 1,1 -N 3 ) 2 (μ 1,3 -N 3 ) 2 ](NO 3 ) 2 ·5H 2 O (4) complex, the J value of −133(3) cm −1 was obtained upon moderate-to-strong antiferromagnetic coupling through the di-μ 1,3 -N 3 -bridged dicopper( ii ) unit with no magnetic interaction between the two copper( ii ) ions in the di-μ 1,1 -N 3 -bridged unit.more » « less

Reaction of nickel and zinc triflates with the tridentate leucoverdazyl 1-isopropyl-3,5-di (2′-pyridyl)-6-oxo-2H-tetrazine (dipyvdH) and triethylamine resulted in the neutral coordination compounds M(dipyvd)2(M = Ni,Zn). In acetonitrile, both compounds undergo two one electron oxidation processes, Zn (dipyvd)2 at −0.28 V and −0.12 V and Ni(dipyvd)2 at −0.32 V and −0.15 V vs ferrocene/ferricenium. Oxidations are ligand based resulting in an intermediate mixed valence species and a cationic bis(verdazyl) compound respectively. Oxidation of the ligand changes a localized, antiaromatic, non-planar 8π electron anion to a planar, delocalized 7π electron radical. The change in ligand structure results in an increase in the octahedral ligand field splitting from 10,500 cm–1to ∼13,000 cm–1, suggesting an increase in the pi acceptor character of the ligand. In the mixed valence species, spectroscopic data suggests minimal interaction between ligands mediated by the metal center; i.e., these are class I-II systems in the Robin-Day classification.more » « less
