Salmonella enterica serovar Typhimurium must adapt to rapid environmental shifts, including those encountered upon entry and during replication to survive within macrophages during pathogenesis. Despite extensive RNA-seq-based investigations, questions remain regarding the range, timing and magnitude of response dynamics. Here we constructed a comprehensive GFP-reporter strain library representing 2,901 computationally identified Salmonella promoter regions to study time-resolved Salmonella transcriptional responses. Promoter activity was measured during in vitro growth and during intracellular infection of RAW 264.7 macrophages. Using bulk measurements and single-cell imaging, we uncovered condition-specific transcriptional regulation and population-level heterogeneity in SPI2-related promoter activity. We also discovered previously unidentified transcriptional activity from 234 promoters. These analyses revealed metabolic shifts including requirements for mntS expression to support manganese homeostasis and expression of Entner–Doudoroff pathway-associated genes to support growth within macrophages. Abstract The purpose of this computational study was to investigate the effects of neonate-focused clinical delivery maneuvers on brachial plexus (BP) during shoulder dystocia. During shoulder dystocia, the anterior shoulder of the neonate is obstructed behind the symphysis pubis of the maternal pelvis, postdelivery of the neonate's head. This is managed by a series of clinical delivery maneuvers. The goal of this study was to simulate these delivery maneuvers and study their effects on neonatal BP strain. Using madymo models of a maternal pelvis and a 90th-percentile neonate, various delivery maneuvers and positions were simulated including the lithotomy position alone of the maternal pelvis, delivery with the application of various suprapubic pressures (SPPs), neonate in an oblique position, and during posterior arm delivery maneuver. The resulting BP strain (%) along with the required maternal delivery force was reported in these independently simulated scenarios. The lithotomy position alone served as the baseline. Each of the successive maneuvers reported a decrease in the required delivery force and resulting neonatal BP strain. As the applied SPP force increased (three scenarios simulated), the required maternal delivery force and neonatal BP strain decreased. A further decrease in both delivery force and neonatal BP strain was observed in the oblique position, with the lowest delivery force and neonatal BP strain reported during the posterior arm delivery maneuver. Data obtained from the improved computational models in this study enhance our understanding of the effects of clinical maneuvers on neonatal BP strain during complicated birthing scenarios such as shoulder dystocia.more » « less

Declining Arctic sea ice is increasing polar bear land use. Polar bears on land are thought to minimize activity to conserve energy. Here, we measure the daily energy expenditure (DEE), diet, behavior, movement, and body composition changes of 20 different polar bears on land over 19–23 days from August to September (2019–2022) in Manitoba, Canada. Polar bears on land exhibited a 5.2-fold range in DEE and 19-fold range in activity, from hibernation-like DEEs to levels approaching active bears on the sea ice, including three individuals that made energetically demanding swims totaling 54–175 km. Bears consumed berries, vegetation, birds, bones, antlers, seal, and beluga. Beyond compensating for elevated DEE, there was little benefit from terrestrial foraging toward prolonging the predicted time to starvation, as 19 of 20 bears lost mass (0.4–1.7 kg•day−1). Although polar bears on land exhibit remarkable behavioral plasticity, our findings reinforce the risk of starvation, particularly in subadults, with forecasted increases in the onshore period.more » « less

Abstract The Green Bank 820 MHz pulsar survey covers ≃173 deg2in the Cygnus X region of the Galaxy, centered onl= 84.°5 andb= 1.°5. Significant star formation is present in this region, and lines of sight pass through three arms of the Galaxy (Orion–Cygnus, Perseus, and an outer arm). Using the Green Bank Telescope, we recorded 200 MHz of bandwidth for 4.5 minutes at 81.92μs resolution for each of 3457 observed survey pointings during 2016 and 2017, covering about two-thirds of the total area. We searched these data for pulsars and report the discovery of six new pulsars—PSRs J2016+3820, J2016+4231, J2019+3810, J2035+3538, J2035+3655, and J2041+4551—and the codiscovery of PSR J2057+4701. PSR J2035+3655 is in a short (4.5 hr) binary orbit; we report the full binary solution and weakly constrain the mass of the pulsar via a marginal (2σ) detection of the Shapiro delay. We also searched the survey data for known pulsars to estimate the survey’s sensitivity and measured 820 MHz pulse widths and flux density for 20 detected sources. For sources that were also detected in the Green Bank North Celestial Cap survey at 350 MHz, we measure scattering parameters and compare to expectations for the region. With these results, we revisit the population estimates that motivated this survey and consider the impact of the survey’s yield on their underlying models. We note an apparent underestimate in dispersion measure predictions from typical Galactic electron density models in the survey region, and discuss future observation strategies.more » « less

In this work, we expand on the XENON1T nuclear recoil searches to study the individual signals of dark matter interactions from operators up to dimension eight in a chiral effective field theory (ChEFT) and a model of inelastic dark matter (iDM). We analyze data from two science runs of the XENON1T detector totaling exposure. For these analyses, we extended the region of interest from to to enhance our sensitivity for signals that peak at nonzero energies. We show that the data are consistent with the background-only hypothesis, with a small background overfluctuation observed peaking between 20 and , resulting in a maximum local discovery significance of for the ChEFT channel for a dark matter particle of and for an iDM particle of with a mass splitting of . For each model, we report 90% confidence level upper limits. We also report upper limits on three benchmark models of dark matter interaction using ChEFT where we investigate the effect of isospin-breaking interactions. We observe rate-driven cancellations in regions of the isospin-breaking couplings, leading to up to 6 orders of magnitude weaker upper limits with respect to the isospin-conserving case. This paper details the first application of a software tagging algorithm to reduce radon-induced backgrounds in liquid noble element time projection chambers, such as XENON1T and XENONnT. The convection velocity field in XENON1T was mapped out using and events, and the rms convection speed was measured to be . Given this velocity field, background events can be tagged when they are followed by and decays, or preceded by decays. This was achieved by evolving a point cloud in the direction of a measured convection velocity field, and searching for and decays or decays within a volume defined by the point cloud. In XENON1T, this tagging system achieved a background reduction of with an exposure loss of , despite the timescales of convection being smaller than the relevant decay times. We show that the performance can be improved in XENONnT, and that the performance of such a software-tagging approach can be expected to be further improved in a diffusion-limited scenario. Finally, a similar method might be useful to tag the cosmogenic background, which is relevant to the search for neutrinoless double-beta decay. Abstract The precision in reconstructing events detected in a dual-phase time projection chamber depends on an homogeneous and well understood electric field within the liquid target. In the XENONnT TPC the field homogeneity is achieved through a double-array field cage, consisting of two nested arrays of field shaping rings connected by an easily accessible resistor chain. Rather than being connected to the gate electrode, the topmost field shaping ring is independently biased, adding a degree of freedom to tune the electric field during operation. Two-dimensional finite element simulations were used to optimize the field cage, as well as its operation. Simulation results were compared to$${}^{83\textrm{m}}\hbox {Kr }$$ calibration data. This comparison indicates an accumulation of charge on the panels of the TPC which is constant over time, as no evolution of the reconstructed position distribution of events is observed. The simulated electric field was then used to correct the charge signal for the field dependence of the charge yield. This correction resolves the inconsistent measurement of the drift electron lifetime when using different calibrations sources and different field cage tuning voltages.more » « less

The impact of educators in informal science learning sites (ISLS) remains understudied from the perspective of youth visitors. Less is known about whether engagement with educators differs based on the age and gender of both visitor and educator. Here, visitors (5–17 years old) to six ISLS in the United States and United Kingdom (n¼488, female n¼244) were surveyed following an interaction with either a youth (14–18 -years old) or adult educator (19þ years old). For participants who reported lower interest in the exhibit, more educator engagement was related to greater self-reported learning. Younger children and adolescents reported more engagement with an adult educator, whereas engagement in middle childhood did not differ based on educator age. Participants in middle childhood showed a trend toward answering more conceptual knowledge questions correctly following an interaction with a youth educator. Together, these findings emphasize the promise of tailoring educator experiences to visitor demographics.more » « less

Abstract The multi-staged XENON program at INFN Laboratori Nazionali del Gran Sasso aims to detect dark matter with two-phase liquid xenon time projection chambers of increasing size and sensitivity. The XENONnT experiment is the latest detector in the program, planned to be an upgrade of its predecessor XENON1T. It features an active target of 5.9 tonnes of cryogenic liquid xenon (8.5 tonnes total mass in cryostat). The experiment is expected to extend the sensitivity to WIMP dark matter by more than an order of magnitude compared to XENON1T, thanks to the larger active mass and the significantly reduced background, improved by novel systems such as a radon removal plant and a neutron veto. This article describes the XENONnT experiment and its sub-systems in detail and reports on the detector performance during the first science run.more » « less
