Salicylic acid (SA) is an important plant hormone with a critical role in plant defense against pathogen infection. Despite extensive research over the past 30 year or so, SA biosynthesis and its complex roles in plant defense are still not fully understood. Even though earlier biochemical studies suggested that plants synthesize SA from cinnamate produced by phenylalanine ammonia lyase (PAL), genetic analysis has indicated that in Arabidopsis, the bulk of SA is synthesized from isochorismate (IC) produced by IC synthase (ICS). Recent studies have further established the enzymes responsible for the conversion of IC to SA in Arabidopsis. However, it remains unclear whether other plants also rely on the ICS pathway for SA biosynthesis. SA induces defense genes against biotrophic pathogens, but represses genes involved in growth for balancing defense and growth to a great extent through crosstalk with the growth-promoting plant hormone auxin. Important progress has been made recently in understanding how SA attenuates plant growth by regulating the biosynthesis, transport, and signaling of auxin. In this review, we summarize recent progress in the biosynthesis and the broad roles of SA in regulating plant growth during defense responses. Further understanding of SA production and its regulation of both defense and growth will be critical for developing better knowledge to improve the disease resistance and fitness of crops.

Lignans of Sesame (Sesamum indicum L.): A Comprehensive Review
Major lignans of sesame sesamin and sesamolin are benzodioxol--substituted furofurans. Sesamol, sesaminol, its epimers, and episesamin are transformation products found in processed products. Synthetic routes to all lignans are known but only sesamol is synthesized industrially. Biosynthesis of furofuran lignans begins with the dimerization of coniferyl alcohol, followed by the formation of dioxoles, oxidation, and glycosylation. Most genes of the lignan pathway in sesame have been identified but the inheritance of lignan content is poorly understood. Health-promoting properties make lignans attractive components of functional food. Lignans enhance the efficiency of insecticides and possess antifeedant activity, but their biological function in plants remains hypothetical. In this work, extensive literature including historical texts is reviewed, controversial issues are critically examined, and errors perpetuated in literature are corrected. The following aspects are covered: chemical properties and transformations of lignans; analysis, purification, and total synthesis; occurrence in Seseamum indicum and related plants; biosynthesis and genetics; biological activities; health-promoting properties; and biological functions. Finally, the improvement of lignan content in sesame seeds by breeding and biotechnology and the potential of hairy roots for manufacturing lignans in vitro are outlined.

Abstract Phyllobilins are open-chain products of the biological degradation of chlorophyll a in higher plants. Recent studies reveal that phyllobilins exert anti-oxidative and anti-inflammatory properties, as well as activities against cancer cells, that contribute to the human health benefits of numerous plants. In general, phyllobilins have been overlooked in phytochemical analyses, and – more importantly – in the analyses of medicinal plant extracts. Nevertheless, over the past three decades, > 70 phyllobilins have been identified upon examination of more than 30 plant species. Eight distinct chromophoric classes of phyllobilins are known: phyllolumibilins (PluBs), phylloleucobilins (PleBs), phylloxanthobilins (PxBs), and phylloroseobilins (PrBs)–each in type-I or type-II groups. Here, we present a database of absorption and fluorescence spectra that has been compiled of 73 phyllobilins to facilitate identification in phytochemical analyses. The spectra are provided in digital form and can be viewed and downloaded at www.photochemcad.com. The present review describes the plant origin, molecular structure, and absorption and fluorescence features of the 73 phyllobilins, along with an overview of key medicinal properties. The review should provide an enabling tool for the community for the straightforward identification of phyllobilins in plant extracts, and the foundation for deeper understanding of these ubiquitous but underexamined plant-derived micronutrients for human health.more » « less

Web applications are governed by privacy policies, but developers lack practical abstractions to ensure that their code actually abides by these policies. This leads to frequent oversights, bugs, and costly privacy violations. Sesame is a practical framework for end-to-end privacy policy enforcement. Sesame wraps data in policy containers that associate data with policies that govern its use. Policy containers force developers to use privacy regions when operating on the data, and Sesame combines sandboxing and a novel static analysis to prevent privacy regions from leaking data. Sesame enforces a policy check before externalizing data, and it supports custom I/O via reviewed, signed code. Experience with four web applications shows that Sesame’s automated guarantees cover 95% of application code, with the remaining 5% needing manual review. Sesame achieves this with reasonable application developer effort and imposes 3–10% performance overhead (10–55% with sandboxes).more » « less

Products derived from Cannabis sativa L. have gained increased interest and popularity. As these products become common amongst the public, the health and potential therapeutic values associated with hemp have become a premier focus of research. While the psychoactive and medicinal properties of Cannabis products have been extensively highlighted in the literature, the antibacterial properties of cannabidiol (CBD) have not been explored in depth. This research serves to examine the antibacterial potential of CBD against Salmonella newington and S. typhimurium. In this study, we observed bacterial response to CBD exposure through biological assays, bacterial kinetics, and fluorescence microscopy. Additionally, comparative studies between CBD and ampicillin were conducted against S. typhimurium and S. newington to determine comparative efficacy. Furthermore, we observed potential resistance development of our Salmonella spp. against CBD treatment.more » « less

Glucosylceramides impact cellulose deposition and cellulose synthase complex motility in ArabidopsisAbstract Cellulose is an abundant component of plant cell wall matrices, and this para-crystalline polysaccharide is synthesized at the plasma membrane by motile Cellulose Synthase Complexes (CSCs). However, the factors that control CSC activity and motility are not fully resolved. In a targeted chemical screen, we identified the alkylated nojirimycin analog N-Dodecyl Deoxynojirimycin (ND-DNJ) as a small molecule that severely impacts Arabidopsis seedling growth. Previous work suggests that ND-DNJ-related compounds inhibit the biosynthesis of glucosylceramides (GlcCers), a class of glycosphingolipid associated with plant membranes. Our work uncovered major changes in the sphingolipidome of plants treated with ND-DNJ, including reductions in GlcCer abundance and altered acyl chain length distributions. Crystalline cellulose content was also reduced in ND-DNJ-treated plants as well as plants treated with the known GlcCer biosynthesis inhibitor N-[2-hydroxy-1-(4-morpholinylmethyl)-2-phenyl ethyl]-decanamide (PDMP) or plants containing a genetic disruption in GLUCOSYLCERAMIDE SYNTHASE (GCS), the enzyme responsible for sphingolipid glucosylation that results in GlcCer synthesis. Live-cell imaging revealed that CSC speed distributions were reduced upon treatment with ND-DNJ or PDMP, further suggesting an important relationship between glycosylated sphingolipid composition and CSC motility across the plasma membrane. These results indicate that multiple interventions compromising GlcCer biosynthesis disrupt cellulose deposition and CSC motility, suggesting that GlcCers regulate cellulose biosynthesis in plants.more » « less
