Title: Water-soluble dinuclear iridium( iii ) and ruthenium( ii ) bis-terdentate complexes: photophysics and electrochemiluminescence
The synthesis, photophysics, and electrochemiluminescence (ECL) of four water-soluble dinuclear Ir( iii ) and Ru( ii ) complexes (1–4) terminally-capped by 4′-phenyl-2,2′:6′,2′′-terpyridine (tpy) or 1,3-di(pyrid-2-yl)-4,6-dimethylbenzene (N^C^N) ligands and linked by a 2,7-bis(2,2′:6′,2′′-terpyridyl)fluorene with oligoether chains on C9 are reported. The impact of the tpy or N^C^N ligands and metal centers on the photophysical properties of 1–4 was assessed by spectroscopic methods including UV-vis absorption, emission, and transient absorption, and by time-dependent density functional theory (TDDFT) calculations. These complexes exhibited distinct singlet and triplet excited-state properties upon variation of the terminal-capping terdentate ligands and the metal centers. The ECL properties of complexes 1–3 with better water solubility were investigated in neutral phosphate buffer solutions (PBS) by adding tripropylamine (TPA) as a co-reactant, and the observed ECL intensity followed the descending order of 3 > 1 > 2. Complex 3 bearing the [Ru(tpy) 2 ] 2+ units displayed more pronounced ECL signals, giving its analogues great potential for further ECL study.  more » « less

In this work, we investigated bonding features of 15 ruthenium(II) nitrile complexes of the type [Ru(tpy)(L)-(CH 3 CN)] n+ , containing the tridentate tpy ligand (tpy = 2,2′:6′,2″-terpyridine) and various bidentate ancillary ligands L; 12 compounds originally synthesized by Loftus et al. [J. Phys. Chem. C 123, 10291–10299 (2019)] and three new complexes. We utilized local vibrational force constants derived from the local mode theory as a quantitative measure of bond strength complemented with the topological analysis of the electron density and the natural bond orbital analysis. Loftus et al. suggested that nitrile dissociation occurs after light induced singlet–triplet transition of the original complexes and they used as a measure of nitrile release efficiency quantum yields for ligand exchange in water. They observed larger quantum yields for complexes with smaller singlet–triplet energy gaps. The major goal of this work was to assess how the Ru–NC and Ru–L bond strengths in these 15 compounds relate to and explain the experimental data of Loftus et al., particularly focusing on the question whether there is a direct correlation between Ru–NC bond strength and measured quantum yield. Our study provides the interesting result that the compounds with the highest quantum yields also have the strongest Ru–NC bonds suggesting that breaking the Ru–NC bond is not the driving force for the delivery process rather than the change of the metal framework as revealed by first results of a unified reaction valley approach investigation of the mechanism. Compounds with the highest quantum yield show larger electronic structure changes upon singlet–triplet excitation, i.e., larger changes in bond strength, covalency, and difference between the singlet and triplet HOMOs, with exception of the compound 12. In summary, this work provides new insights into the interplay of local properties and experimental quantum yields forming in synergy a useful tool for fine tuning of existing and future design of new nitrile releasing ruthenium compounds. We hope that this work will bring theoretical and experimental studies closer together and serves as an incubator for future collaboration between computational chemists and their experimental colleagues.

    To ascertain the influence of binary ligand systems [1,1-dicyanoethylene-2,2-dithiolate (i-mnt −2 ) and polyamine {tetraen = tris(2-aminoethyl)amine, tren = diethylene triamine and opda = o -phenylenediamine}] on the coordination modes of the Ni( ii ) metal center and resulting supramolecular architectures, a series of nickel( ii ) thiolate complexes [Ni(tetraen)(i-mnt)](DMSO) ( 1 ), [Ni 2 (tren) 2 (i-mnt) 2 ] ( 2 ), and [Ni 2 (i-mnt) 2 (opda) 2 ] n ( 3 ) have been synthesized in high yield in one step in water and structurally characterized by single crystal X-ray crystallography and spectroscopic techniques. X-ray diffraction studies disclose the diverse i-mnt −2 coordination to the Ni +2 center in the presence of active polyamine ligands, forming a slightly distorted octahedral geometry (NiN 4 S 2 ) in 1 , square planar (NiS 4 ) and distorted octahedral geometries (NiN 6 ) in the bimetallic co-crystallized aggregate of cationic [Ni(tren) 2 ] +2 and anionic [Ni(i-mnt) 2 ] −2 in 2 , and a one dimensional (1D) polymeric chain along the [100] axis in 3 , having consecutive square planar (NiS 4 ) and octahedral (NiN 6 ) coordination kernels. The N–H⋯O, N–H⋯S, N–H⋯N, N–H⋯S, N–H⋯N, and N–H⋯O type hydrogen bonds stabilize the supramolecular assemblies in 1 , 2 , and 3 respectively imparting interesting graph-set-motifs. The molecular Hirshfeld surface analyses (HS) and 2D fingerprint plots were utilized for decoding all types of non-covalent contacts in the crystal networks. Atomic HS analysis of the Ni +2 centers reveals significant Ni–N metal–ligand interactions compared to Ni–S interactions. We have also studied the unorthodox interactions observed in the solid state structures of 1–3 by QTAIM and NBO analyses. Moreover, all the complexes proved to be highly active water reduction co-catalysts (WRC) in a photo-catalytic hydrogen evolution process involving iridium photosensitizers, wherein 2 and 3 having a square planar arrangement around the nickel center(s) – were found to be the most active ones, achieving 1000 and 1119 turnover numbers (TON), respectively.

A series of five ruthenium complexes containing triphenyl phosphine groups known to enhance both cellular penetration and photoinduced ligand exchange, cis -[Ru(bpy) 2 (P( p -R-Ph) 3 )(CH 3 CN)] 2+ , where bpy = 2,2′-bipyridine and P( p -R-Ph) 3 represent para -substituted triphenylphosphine ligands with R = –OCH 3 (1), –CH 3 (2) –H (3), –F (4), and –CF 3 (5), were synthesized and characterized. The photolysis of 1–5 in water with visible light ( λ irr ≥ 395 nm) results in the substitution of the coordinated acetonitrile with a solvent molecule, generating the corresponding aqua complex as the single photoproduct. A 3-fold variation in quantum yield was measured with 400 nm irradiation, Φ 400 , where 1 is the most efficient with a Φ 400 = 0.076(2), and 5 the least photoactive complex, with Φ 400 = 0.026(2). This trend is unexpected based on the red-shifted metal-to-ligand charge transfer (MLCT) absorption of 1 as compared to that of 5, but can be correlated to the substituent Hammett para parameters and p K a values of the ancillary phosphine ligands. Complexes 1–5 are not toxic towards the triple negative breast cancer cell line MDA-MB-231 in the dark, but 3 and 5 are >4.2 and >19-fold more cytotoxic upon irradiation with blue light, respectively. A number of experiments point to apoptosis, and not to necrosis or necroptosis, as the mechanism of cell death by 5 upon irradiation. These findings provide a foundation for understanding the role of phosphine ligands on photoinduced ligand substitution and show the enhancement afforded by –CF 3 groups on photochemotherapy, which will aid the future design of photocages for photochemotherapeutic drug delivery.

  4. ABSTRACT

    We report the synthesis, photochemical and biological characterization of two new Ru(II) photoactivated complexes based on [Ru(tpy)(Me2bpy)(L)]2+(tpy = 2,2':6',2''‐terpyridine, Me2bpy = 6,6'‐dimethyl‐2,2'‐bipyridine), where L = pyridyl‐BODIPY (pyBOD). Two pyBOD ligands were prepared bearing flanking hydrogen or iodine atoms. Ru(II)‐bound BODIPY dyes show a red‐shift of absorption maxima relative to the free dyes and undergo photodissociation of BODIPY ligands with green light irradiation. Addition of iodine into the BODIPY ligand facilitates intersystem crossing, which leads to efficient singlet oxygen production in the free dye, but also enhances quantum yield of release of the BODIPY ligand from Ru(II). This represents the first report of a strategy to enhance photodissociation quantum yields through the heavy‐atom effect in Ru(II) complexes. Furthermore, Ru(II)‐bound BODIPY dyes display fluorescence turn‐on once released, with a lead analog showing nanomolar EC50values against triple negative breast cancer cells, >100‐fold phototherapeutic indexes under green light irradiation, and higher selectivity toward cancer cells as compared to normal cells than the corresponding free BODIPY photosensitizer. Conventional Ru(II) photoactivated complexes require nonbiorthogonal blue light for activation and rarely show submicromolar potency to achieve cell death. Our study represents an avenue for the improved photochemistry and potency of future Ru(II) complexes.

  5. Abstract

    Catalytic water oxidation is an important process for the development of clean energy solutions and energy storage. Despite the significant number of reports on active catalysts, systematic control of the catalytic activity remains elusive. In this study, descriptors are explored that can be correlated with catalytic activity. [Ru(tpy)(pic)2(H2O)](NO3)2and [Ru(EtO‐tpy)(pic)2(H2O)](NO3)2(where tpy=2,2′ : 6′,2“‐terpyridine, EtO‐tpy=4′‐(ethoxy)‐2,2′:6′,2”‐terpyridine, pic=4‐picoline) are synthesized and characterized by NMR, UV/Vis, EPR, resonance Raman, and X‐ray absorption spectroscopy, and electrochemical analysis. Addition of the ethoxy group increases the catalytic activity in chemically driven and photocatalytic water oxidation. Thus, the effect of the electron‐donating group known for the [Ru(tpy)(bpy)(H2O)]2+family is transferable to architectures with a tpy ligandtransto the Ru‐oxo unit. Under catalytic conditions, [Ru(EtO‐tpy)(pic)2(H2O)](NO3)2displays new spectroscopic signals tentatively assigned to a peroxo intermediate. Reaction pathways were analyzed by using DFT calculations. [Ru(EtO‐tpy)(pic)2(H2O)](NO3)2is found to be one of the most active catalysts functioning by a water nucleophilic attack mechanism.
