Naturally occurring amino acids have been broadly used as additives to improve protein solubility and inhibit aggregation. In this study, improvements in protein signal intensity obtained with the addition of l -serine, and structural analogs, to the desorption electrospray ionization mass spectrometry (DESI-MS) spray solvent were measured. The results were interpreted at the hand of proposed mechanisms of solution additive effects on protein solubility and dissolution. DESI-MS allows for these processes to be studied efficiently using dilute concentrations of additives and small amounts of proteins, advantages that represent real benefits compared to classical methods of studying protein stability and aggregation. We show that serine significantly increases the protein signal in DESI-MS when native proteins are undergoing unfolding during the dissolution process with an acidic solvent system ( p -value = 0.0001), or with ammonium bicarbonate under denaturing conditions for proteins with high isoelectric points ( p -value = 0.001). We establish that a similar increase in the protein signal cannot be observed with direct ESI-MS, and the observed increase is therefore not related to ionization processes or changes in the physical properties of the bulk solution. The importance of the presence of serine during protein conformational changes while undergoing dissolution is demonstrated through comparisons between the analyses of proteins deposited in native or unfolded states and by using native state-preserving and denaturing desorption solvents. We hypothesize that direct, non-covalent interactions involving all three functional groups of serine are involved in the beneficial effect on protein solubility and dissolution. Supporting evidence for a direct interaction include a reduction in efficacy with d -serine or the racemic mixture, indicating a non-bulk-solution physical property effect; insensitivity to the sample surface type or relative placement of serine addition; and a reduction in efficacy with any modifications to the serine structure, most notably the carboxyl functional group. An alternative hypothesis, also supported by some of our observations, could involve the role of serine clusters in the mechanism of solubility enhancement. Our study demonstrates the capability of DESI-MS together with complementary ESI-MS experiments as a novel tool for understanding protein solubility and dissolution and investigating the mechanism of action for solubility-enhancing additives.

Transforming protein-polymer conjugate purification by tuning protein solubility
Abstract Almost all commercial proteins are purified using ammonium sulfate precipitation. Protein-polymer conjugates are synthesized from pure starting materials, and the struggle to separate conjugates from polymer, native protein, and from isomers has vexed scientists for decades. We have discovered that covalent polymer attachment has a transformational effect on protein solubility in salt solutions. Here, protein-polymer conjugates with a variety of polymers, grafting densities, and polymer lengths are generated using atom transfer radical polymerization. Charged polymers increase conjugate solubility in ammonium sulfate and completely prevent precipitation even at 100% saturation. Atomistic molecular dynamic simulations show the impact is driven by an anti-polyelectrolyte effect from zwitterionic polymers. Uncharged polymers exhibit polymer length-dependent decreased solubility. The differences in salting-out are then used to simply purify mixtures of conjugates and native proteins into single species. Increasing protein solubility in salt solutions through polymer conjugation could lead to many new applications of protein-polymer conjugates.

Abstract Protein‐polymer bioconjugates present a way to make enzymes more efficient and robust for industrial and medicinal applications. While much work has focused on mono‐functional conjugates, that is, conjugates with one type of polymer attached such as poly(ethylene glycol) or poly(N‐isopropylacrylamide), there is a practical interest in gaining additional functionality by synthesizing well‐defined bifunctional conjugates in a hetero‐arm star copolymer architecture with protein as the core. Using ubiquitin as a model protein, a synthetic scheme is developed to attach two different polymers (oligo(ethylene oxide) methacrylate and N,N‐dimethylacrylamide) directly to the protein surface, using orthogonal conjugation chemistries and grafting‐from by photochemical living radical polymerization techniques. The additional complexity arising from attempts to selectively modify multiple sites led to decreased polymerization performance and indicates that initiators for continuous activator regeneration atom transfer radical polymerization and reversible addition‐fragmentation chain transfer polymerization are not well‐suited to bifunctional bioconjugates applications under the studied conditions. Nonetheless, the polymerization conditions preserve the native fold of the ubiquitin and enable production of a hetero‐arm star protein‐polymer bioconjugate.more » « less

Antibodies against poly(ethylene glycol) (PEG) have been found to be the culprit of side reactions and efficacy loss of a number of PEGylated drugs. Fundamental mechanisms of PEG immunogenicity and design principles for PEG alternatives still have not been fully explored. By using hydrophobic interaction chromatography (HIC) under varied salt conditions, we reveal the “hidden” hydrophobicity of those polymers which are generally considered as hydrophilic. A correlation between the hidden hydrophobicity of a polymer and its polymer immunogenicity is observed when this polymer is conjugated with an immunogenic protein. Such a correlation of hidden hydrophobicity vs. immunogenicity for a polymer also applies to corresponding polymer–protein conjugates. Atomistic molecular dynamics (MD) simulation results show a similar trend. Based on polyzwitterion modification and with this HIC technique, we are able to produce extremely low-immunogenic protein conjugates as their hydrophilicity is pushed to the limit and their hydrophobicity is eliminated, breaking the current barriers of eliminating anti-drug and anti-polymer antibodies.more » « less

PEGylation is the gold standard in protein‐polymer conjugation, improving circulation half‐life of biologics while mitigating the immune response to a foreign substance. However, preexisting anti‐PEG antibodies in healthy humans are becoming increasingly prevalent and elicitation of anti‐PEG antibodies when patients are administered with PEGylated therapeutics challenges their safety profile. In the current study, two distinct amine‐reactive poly(oxanorbornene) (PONB) imide‐based water‐soluble block co‐polymers are synthesized using ring‐opening metathesis polymerization (ROMP). The synthesized block‐copolymers include PEG‐based PONB‐PEG and sulfobetaine‐based PONB‐Zwit. The polymers are then covalently conjugated to amine residues of lysozyme (Lyz) and urate oxidase (UO) using a grafting‐to bioconjugation technique. Both Lyz‐PONB and UO‐PONB conjugates retained significant bioactivities after bioconjugation. Immune recognition studies of UO‐PONB conjugates indicated a comparable lowering of protein immunogenicity when compared to PEGylated UO. PEG‐specific immune recognition is negligible for UO‐PONB‐Zwit conjugates, as expected. These polymers provide a new alternative for PEG‐based systems that retain high levels of activity for the biologic while showing improved immune recognition profiles.more » « less

Sea salt aerosols contribute significantly to the mass loading of ambient aerosol, which may serve as cloud condensation nuclei and can contribute to light scattering in the atmosphere. Two major chemical components commonly found in sea salts are ammonium sulfate (AS) and sodium chloride (NaCl). It has been shown that alkylamines, derivatives of ammonia, can react with ammonium salts in the particle-phase to displace ammonia and likely change the particle properties. This study investigated the effects of atmospheric alkylamines on the composition and properties of sea salt aerosols using a chemical system of methylamine (MA, as a proxy of alkylamines), AS and NaCl (as a proxy of sea salt aerosol). The concentrations of ammonia and MA in aqueous/gas phases at the thermodynamic equilibrium were determined using the Extended Aerosols and Inorganics Model (E-AIM) under varying initial inputs, along with the deliquescence relative humidity (DRH) and the corresponding particle water content. Our findings indicated a notable negative relationship between MA concentration and the DRH for both AS and NaCl while the effect of MA on NaCl is smaller than that on AS. The salt of MA in the particle phase may absorb water vapor and may lead to the displacement reaction between AS and NaCl due to the low solubility of sodium sulfate. The acidity in the particle phase also played a significant role in affecting the DRH of sea salt aerosols. Since both sea salt aerosol and alkylamines are emitted into the atmosphere from the ocean in large quantities, our study suggested the potential impact of alkylamines on the environment and the climate via the modification of sea salt aerosol properties.more » « less
