Title: Predation differentially structures immature mosquito populations in stormwater ponds
1. Understanding the factors underlying the abundance and distribution of species requires the consideration of a complex suite of interacting biotic and abiotic factors operating on multiple spatial and temporal scales. Larval mosquitoes inhabiting small human‐constructed ponds represent a unique opportunity to investigate the relative importance of these structuring mechanisms while simultaneously generating applied knowledge on mosquito control. 2. A multi‐year field survey of 32 stormwater ponds was conducted in central Illinois (Champaign County). From each pond, data were collected on pond structure type and hydroperiod, the presence/absence of cattails (Typhaspp.), and measures of total nitrogen, phosphorus, and organic carbon, and chlorophylla. The communities of crustacean zooplankton and aquatic insects were characterised, and these taxa were assigned into two main groups: predators and competitors of larval mosquitoes. Structural equation modelling was used to explore the direct and indirect effects of these biotic and abiotic factors on larval density for three species of culicine mosquitoes (Culex pipiens,Culex restuans, andAedes vexans). 3. Hydroperiod had an indirect negative effect onC. pipiensdensity. However, this effect was mediated by predator density; more permanent ponds had more predators, which therefore reduced the density ofC. pipienslarvae.Aedes vexansdensity was positively correlated with predator density. No predictor variables were found that explained variation inC. restuansdensity. 4. This study show that the relative importance of these biotic and abiotic factors varies among species of culicine mosquitoes inhabiting stormwater ponds.  more » « less

Abstract Dormant propagules can provide a rapid colonization source for temporary aquatic habitats and set the trajectory for community dynamics, yet the egg banks of stormwater management systems have received little attention. We asked which species hatched from the sediment of drainage ditches in Champaign County, IL, and found bdelloid rotifers and ostracods (Heterocypris incongruens) to be the most common taxa. These sites also are colonized by mosquitoes, and we established laboratory experiments to examine interspecific interactions between common co‐occurring taxa.Culex restuanslarvae were reared in the presence or absence ofH. incongruensat two intra‐ and interspecific densities (20 or 40 total individuals) and their survivorship to adulthood, development time to adulthood, adult body size, and sex ratio were determined. Survival forCx. restuanswas significantly lower at high larval density than at low larval density in both treatments.Culex restuanslarvae reared in the presence ofH. incongruenshad a shorter development time to adulthood and emerged as larger adults compared to those reared in the absence ofH. incongruens. The sex ratios in theH. incongruenstreatments were female‐biased whereas those in theCulex‐only treatments were male‐biased. These differences may have epidemiological implications, as only female mosquitoes serve as disease vectors. Our results emphasize the importance of understanding interspecific interactions in influencing larval mosquito development traits.

  2. Abstract BackgroundCulexmosquitoes are the primary vectors of West Nile virus (WNV) across the USA. Understanding when these vectors are active indicates times when WNV transmission can occur. This study determined the proportion of femaleCulexmosquitoes that were in diapause during the fall and winter and when they terminated diapause and began blood feeding in the spring. MethodsMosquitoes were collected from parks using various traps and/or aspirated from culverts in Franklin County, Ohio, from October to mid-May from 2019 to 2022.Culexmosquitoes were morphologically identified to species, and the ovaries of females were dissected to determine their diapause and parity statuses. ResultsBy early October 2021, roughly 95% ofCulex pipienscollected in culverts were in diapause and 98% ofCx. erraticuswere in diapause. Furthermore, gravid and blood-fedCulex salinarius,Cx. pipiens, andCx. restuanswere collected in late November in 2019 and 2021 in standard mosquito traps. In the winter of 2021, the proportions of non-diapausingCulexdecreased within culverts. The last non-diapausingCx. erraticuswas collected in late December 2021 while the final non-diapausingCx. pipienswas collected in mid-January 2022, both in culverts. Roughly 50% ofCx. pipiensterminated diapause by mid-March 2022, further supported by our collections of gravid females in late March in all 3 years of mosquito collection. In fact, male mosquitoes ofCx. pipiens,Cx. restuans, andCx. territanswere collected by the 1st week of May in 2022, indicating that multiple species ofCulexproduced a second generation that reached adulthood by this time. ConclusionsWe collected blood-fed and gravidCulexfemales into late November in 2 of the 3 years of our collections, indicating that it might be possible for WNV transmission to occur in late fall in temperate climates like Ohio. The persistence of non-diapausingCx. pipiensandCx. erraticusthroughout December has important implications for the winter survival of WNV vectors and our overall understanding of diapause. Finally, determining whenCulexterminate diapause in the spring may allow us to optimize mosquito management programs and reduce the spread of WNV before it is transmitted to humans. The predator-permanence hypothesis predicts that as hydroperiod increases in lentic ecosystems, biotic interactions—mainly predation—replace physical factors like drying as the main determinant of community structure and population dynamics. We propose that the same transition occurs over time in seasonally flooded ecosystems that are connected to permanent water bodies. To test for evidence of successional changes that are similar to spatial changes in the relative importance of drying and predation, we used a 12-y time series of snail density, predator density, and water depth at 4 sites arranged along a nutrient gradient in a subtropical, seasonally flooded wetland, the Florida Everglades, USA. The rate of change in snail population size was negatively correlated with their density at all 4 sites, suggesting that density-dependent factors such as resource limitation regulate snail dynamics. The strength of the relationship varied among sites such that when water depth changes were less important, snail population size was more important in predicting changes in snail population size. At the site that consistently had the greatest snail density, crayfish density negatively affected the rate of snail population change, suggesting that crayfish predation may limit snail population growth in areas with more or higher-quality resources that support larger snail populations. Tethering studies were also conducted, which revealed higher snail mortality in the wet season, primarily because crushing predators (e.g., molluscivorous fishes) were more common at that time and added to the chronic mortality by entry-based predators (e.g., crayfish, which access snails through their aperture). In summary, 3 of the sites resembled temporary or permanent fishless ponds where snail populations were primarily structured by abiotic factors, intraspecific competition, and invertebrate predators (e.g., crayfish) during the wet season, whereas 1 site showed evidence that snail populations were also influenced by molluscivorous fish. This temporal change in importance of water permanence factors to fish that affected population dynamics supports the spatial pattern proposed by the predator-permanence hypothesis.

  4. Climate change is significantly impacting the geographic range of many animal species and their associated microorganisms, hence influencing emergence of vector-borne diseases. Mosquito-borne viruses represent a potential major reservoir of human pathogens, highlighting the need for improved understanding of ecological factors associated with variation in the mosquito viral community (virome). Here, a subtractive hybridization method coupled with RNAseq of individual mosquito specimens was used to profile RNA viromes of individual co-occurringAedes albopictusandAedes vexansmosquitoes across a 2,000 km spatial scale. Samples were collected and archived by the National Ecological Observatory Network (NEON) from four ecologically variable sites in the Southeastern United States between 2018 and 2019. Results of multivariate analysis suggest that mosquito species are an important factor in RNA viral community composition. Significantly higher viral diversity was detected inA. albopictuscompared toA.vexans. However, season, year, and site of sample collection did not show strong association with virome profiles, supporting the hypothesis that factors unique to the mosquito host species (e.g., larval habitat or vector competence) influence the structure of mosquito viromes.

    Abstract Background The bacterial communities associated with mosquito eggs are an essential component of the mosquito microbiota, yet there are few studies characterizing and comparing the microbiota of mosquito eggs to other host tissues. Methods We sampled gravid female Culex pipiens L. and Culex restuans Theobald from the field, allowed them to oviposit in the laboratory, and characterized the bacterial communities associated with their egg rafts and midguts for comparison through MiSeq sequencing of the 16S rRNA gene. Results Bacterial richness was higher in egg rafts than in midguts for both species, and higher in Cx pipiens than Cx. restuans . The midgut samples of Cx. pipiens and Cx. restuans were dominated by Providencia. Culex pipiens and Cx. restuans egg rafts samples were dominated by Ralstonia and Novosphingobium , respectively. NMDS ordination based on Bray-Curtis distance matrix revealed that egg-raft samples, or midgut tissues harbored similar bacterial communities regardless of the mosquito species. Within each mosquito species, there was a distinct clustering of bacterial communities between egg raft and midgut tissues. Conclusion These findings expand the list of described bacterial communities associated with Cx. pipiens and Cx. restuan s and the additional characterization of the egg raft bacterial communities facilitates comparative analysis of mosquito host tissues, providing a basis for future studies seeking to understand any functional role of the bacterial communities in mosquito biology.
