Title: Lower Energy Excitation of Water Soluble Near‐Infrared Emitting Mixed‐Ligand Metallacrowns
Abstract By combining advantages of two series of lanthanide(III)/zinc(II) metallacrowns (MCs) assembled using pyrazine‐ (pyzHA2−) and quinoxaline‐ (quinoHA2−) hydroximate building blocks ligands, we created here water‐soluble mixed‐ligand MCs with extended absorption to the visible range. The YbIIIanalogue demonstrated improved photophysical properties in the near‐infrared (NIR) range in cell culture media, facilitating its application for NIR optical imaging in living HeLa cells.  more » « less

  1. A family of Zn 16 Ln(HA) 16 metallacrowns (MCs; Ln = Yb III , Er III , and Nd III ; HA = picoline- (picHA 2− ), pyrazine- (pyzHA 2− ), and quinaldine- (quinHA 2− ) hydroximates) with an ‘encapsulated sandwich’ structure possesses outstanding luminescence properties in the near-infrared (NIR) and suitability for cell imaging. Here, to decipher which parameters affect their functional and photophysical properties and how the nature of the hydroximate ligands can allow their fine tuning, we have completed this Zn 16 Ln(HA) 16 family by synthesizing MCs with two new ligands, naphthyridine- (napHA 2− ) and quinoxaline- (quinoHA 2− ) hydroximates. Zn 16 Ln(napHA) 16 and Zn 16 Ln(quinoHA) 16 exhibit absorption bands extended into the visible range and efficiently sensitize the NIR emissions of Yb III , Er III , and Nd III upon excitation up to 630 nm. The energies of the lowest singlet (S 1 ), triplet (T 1 ) and intra-ligand charge transfer (ILCT) states have been determined. Ln III -centered total ( Q LLn) and intrinsic ( Q LnLn) quantum yields, sensitization efficiencies ( η sens ), observed ( τ obs ) and radiative ( τ rad ) luminescence lifetimes have been recorded and analyzed in the solid state and in CH 3 OH and CD 3 OD solutions for all Zn 16 Ln(HA) 16 . We found that, within the Zn 16 Ln(HA) 16 family, τ rad values are not constant for a particular Ln III . The close in energy positions of T 1 and ILCT states in Zn 16 Ln(picHA) 16 and Zn 16 Ln(quinHA) 16 are preferred for the sensitization of Ln III NIR emission and η sens values reach 100% for Nd III . Finally, the highest values of Q LLn are observed for Zn 16 Ln(quinHA) 16 in the solid state or in CD 3 OD solutions. With these data at hand, we are now capable of creating MCs with desired properties suitable for NIR optical imaging.

  2. Abstract Multiplex imaging in the second near‐infrared window (NIR‐II, 1000–1700 nm) provides exciting opportunities for more precise understanding of biological processes and more accurate diagnosis of diseases by enabling real‐time acquisition of images with improved contrast and spatial resolution in deeper tissues. Today, the number of imaging agents suitable for this modality remains very scarce. In this work, we have synthesized and fully characterized, including theoretical calculations, a series of dimeric LnIII/GaIIImetallacrowns bearing RuIIpolypyridyl complexes,LnRu‐3(Ln=YIII, YbIII, NdIII, ErIII). Relaxed structures ofYRu‐3in the ground and the excited electronic states have been calculated using dispersion‐corrected density functional theory methods. Detailed photophysical studies ofLnRu‐3have demonstrated that characteristic emission signals of YbIII, NdIIIand ErIIIin the NIR‐II range can be sensitized upon excitation in the visible range through RuII‐centered metal‐to‐ligand charge transfer (MLCT) states. We have also showed that these NIR‐II signals are unambiguously detected in an imaging experiment using capillaries and biological tissue‐mimicking phantoms. This work opens unprecedented perspectives for NIR‐II multiplex imaging using LnIII‐based molecular compounds.

  3. Abstract Series of lanthanide‐containing metallic coordination complexes are frequently presented as structurally analogous, due to the similar chemical and coordinative properties of the lanthanides. In the case of chiral (LnIII[15‐MCN(L‐pheHA)‐5])3+metallacrowns (MCs), which are well established supramolecular hosts, the formation of dimers templated by a dicarboxylate guest (muconate) in solution of neutral pH is herein shown to have a unique dependence on the identity of the MC's central lanthanide. Calorimetric data and nuclear magnetic resonance diffusion studies demonstrate that MCs containing larger or smaller lanthanides as the central metal only form monomeric host‐guest complexes whereas analogues with intermediate lanthanides (for example, Eu, Gd, Dy) participate in formation of dimeric host‐guest‐host compartments. The driving force for the dimerization event across the series is thought to be a competition between formation of highly stable MCs (larger lanthanides) and optimally linked bridging guests (smaller lanthanides).

  4. Abstract Nanothermometry is the study of temperature at the submicron scale with a broad range of potential applications, such as cellular studies or electronics. Molecular luminescent‐based nanothermometers offer a non‐contact means to record these temperatures with high spatial resolution and thermal sensitivity. A luminescent‐based molecular thermometer comprised of visible‐emitting Ga3+/Tb3+and Ga3+/Sm3+metallacrowns (MCs) achieved remarkable relative thermal sensitivity associated with very low temperature uncertainty ofSr=1.9 % K−1andδT<0.045 K, respectively, at 328 K, as an aqueous suspension of polystyrene nanobeads loaded with the corresponding MCs. To date, they are the ratiometric molecular nanothermometers offering the highest level of sensitivity in the physiologically relevant temperature range.

  5. Abstract Recent research suggests atmospheric cloud radiative effect (ACRE) acts as an important feedback mechanism for enhancing the development of convective self‐aggregation in idealized numerical simulations. Here, we seek observational relationships between longwave (LW) ACRE and the spatial organization of mesoscale convective systems (MCSs) in the tropics. Three convective organization metrics that are positively correlated with the area of MCS, that is, convective organization potential, the area fraction of precipitating MCS, and the precipitation fraction of MCS, are used to indicate the degree of convective organization. Our results show that the contrast in the LW ACRE inside and outside an MCS is consistent across different MCS precipitation intensities throughout the life cycle of an MCS, typically 90–100 W/m2, and provides important positive feedback to the circulation of the given MCS. However, the LW ACRE inside and outside an MCS as well as their difference are not strongly related to the degree of organization, suggesting that the LW cloud radiative feedback may be supportive of MCS formation and maintenance without necessarily being a dominant factor for spatial organization of MCSs. The domain average vertical velocity does tend to be related to the measures of convective organization, suggesting that factors that favor large‐scale low‐level convergence may exert a leading effect in creating an environment favorable for mesoscale organization of deep convection.
