Fabrication of microfluidic devices by photolithography generally requires specialized training and access to a cleanroom. As an alternative, 3D printing enables cost-effective fabrication of microdevices with complex features that would be suitable for many biomedical applications. However, commonly used resins are cytotoxic and unsuitable for devices involving cells. Furthermore, 3D prints are generally refractory to elastomer polymerization such that they cannot be used as master molds for fabricating devices from polymers ( e.g. polydimethylsiloxane, or PDMS). Different post-print treatment strategies, such as heat curing, ultraviolet light exposure, and coating with silanes, have been explored to overcome these obstacles, but none have proven universally effective. Here, we show that deposition of a thin layer of parylene, a polymer commonly used for medical device applications, renders 3D prints biocompatible and allows them to be used as master molds for elastomeric device fabrication. When placed in culture dishes containing human neurons, regardless of resin type, uncoated 3D prints leached toxic material to yield complete cell death within 48 hours, whereas cells exhibited uniform viability and healthy morphology out to 21 days if the prints were coated with parylene. Diverse PDMS devices of different shapes and sizes were easily cast from parylene-coated 3D printed molds without any visible defects. As a proof-of-concept, we rapid prototyped and tested different types of PDMS devices, including triple chamber perfusion chips, droplet generators, and microwells. Overall, we suggest that the simplicity and reproducibility of this technique will make it attractive for fabricating traditional microdevices and rapid prototyping new designs. In particular, by minimizing user intervention on the fabrication and post-print treatment steps, our strategy could help make microfluidics more accessible to the biomedical research community.

                            A Facile Surface Modification Strategy for Antibody Immobilization on 3D-Printed Surfaces
                        
                    
    
            3D-printed microdevices have become increasingly important to the advancement of point-of-care (POC) immunoassays. Despite its great potential, using 3D-printed surfaces on the solid support for immunorecognition has been limited due to the non-ideal adsorption properties for many photocurable resins. In this work, we report a simple surface modification protocol that works for diverse commercial photocurable resins, improving ELISAs performed directly on 3D-printed devices. This surface modification strategy involves surface activation via air plasma followed by the one-step incubation of GLYMO-labeled streptavidin. We successfully immobilized biotinylated anti-activin A antibodies on the 3D-printed surfaces and performed the complete ELISA protocol on the 3D-printed surfaces. We demonstrated that this protocol achieved an improved performance over passive adsorption for ELISAs. The present method is also compatible with diverse commercial resins and works with both microwells and microchannels. Finally, this method demonstrated a comparable limit of detection to the ELISA performed using commercial microwells. We believe the simplicity and broad compatibility of the present surface modification strategy will facilitate the development of 3D-printed POC ELISA devices.

            Abstract Though 3D printing shows potential in fabricating complex optical components rapidly, its poor surface quality and dimensional accuracy render it unqualified for industrial optics applications. The layer steps in the building direction and the pixelated steps on each layer's contour result in inevitable microscale defects on the 3D‐printed surface, far away from the nanoscale roughness required for optics. This paper reports a customized vat photopolymerization‐based lens printing process, integrating unfocused image projection and precision spin coating to solve lateral and vertical stair‐stepping defects. A precision aspherical lens with less than 1 nm surface roughness and 1 µm profile accuracy is demonstrated. The 3D‐printed convex lens achieves a maximum MTF resolution of 347.7 lp mm−1. A mathematical model is established to predict and control the spin coating process on 3D‐printed surfaces precisely. Leveraging this low‐cost yet highly robust and repeatable 3D printing process, the precision fabrication of multi‐scale spherical, aspherical, and axicon lenses are showcased with sizes ranging from 3 to 70 mm using high clear photocuring resins.             The additive manufacturing (AM) industry increasingly looks to differentiate itself by utilizing materials and processes that are green, clean, and sustainable. Biopolymers, bio‐sourced raw materials and light weighting of parts 3D printed with photopolymer resins each represent critical directions for the future of AM. Here, we report a series of bio‐based composite resins with soybean oil derivatives, up to 20% by weight of surface‐methacrylated micro‐crystalline cellulose (MCC) and 60% total bio‐based content for vat photopolymerization based additive manufacturing. The ultimate tensile strengths of the materials were found to increase up to 3X, the Young's moduli increased up to 10X, and the glass transition temperature increased by 11.3°C when compared to the neat resin without surface‐methacrylated MCC as a filler. Working curves and shrinkage factors were used to demonstrate how the surface‐methacrylated MCC causes changes in the dimensions of printed parts, to facilitate development of optimized print parameters based on the UV intensity of the 3D printer being used. These results will allow further development of commercial 3D printable resins with a high concentration of bio‐based fillers that print well and perform on par with conventional resins.more » « less

            Abstract Fused deposition modeling 3D printing provides a cost-effective and streamlined method for producing electrochemical sensors, overcoming the challenges associated with material selection, complex fabrication processes, and reproducibility issues. This study introduces an innovative approach utilizing a dual-printer setup to simplify the manufacturing of sensor electrodes. A critical enhancement in this process is the surface modification with reduced graphene oxide (rGO), which not only improves the electrochemical characteristics but also induces a wrinkled structure on the 3D printed surface. These wrinkles significantly increase the surface area, directly boosting the electrode’s electrochemical performance. Comprehensive characterization of the electrode surfaces, both before and after rGO modification, demonstrates a substantial increase in sensitivity, with a fortyfold improvement observed in hydrogen peroxide (H2O2) amperometric measurements. This breakthrough paves the way for advanced applications in 3D printed electrochemical sensors.more » « less

            Abstract Charge‐programmed 3D printing enables the fabrication of 3D electronics with lightweight and high precision via selective patterning of metals. This selective metal deposition is catalyzed by Pd nanoparticles that are specifically immobilized onto the charged surface and promises to fabricate a myriad of complex electronic devices with self‐sensing, actuation, and structural elements assembled in a designed 3D layout. However, the achievable property space and the material‐performance correlation of the charge‐programmed printing remain unexplored. Herein, a series of photo‐curable resins are designed for unveiling how the charge and crosslink densities synergistically impact the nanocatalyst‐guided selective deposition in catalytic efficiency and properties of the 3D printed charge‐programmed architectures, leading to high‐quality 3D patterning of solid and liquid metals. The findings offer a wide tunability of the structural properties of the printed electronics, ranging from stiff to extreme flexibility. Capitalizing on these results, the printing and successful application of an ultralight‐weight and deployable 3D multi‐layer antenna system operating at an ultrahigh‐frequency of 19 GHz are demonstrated.more » « less
